International Journal of Laboratory Medicine 2015;(13):1844-1845,1848
Objective To explore the application value of combined detection of serum carcinoembryonic antigen(CEA),carbo-hydrate antigen 1 53(CA1 53),carbohydrate antigen 125(CA125)and carbohydrate antigen 1 99(CA1 99)in the diagnosis of lung canc-er.Methods The direct chemiluminescence assay was used to detect the expression levels of serum CEA,CA1 53,CA125 and CA1 99 in 128 cases of patients with lung cancer,84 cases of patients with lung benign disease and 30 cases of healthy individuals. Results The expression levels of serum CEA,CA1 53,CA125,CA1 99 in the lung cancer group was significantly higher than those in the lung benign disease group and the healthy control group(P <0.05).The sensitivity and specificity of serum CEA was high in the lung cancer group, which was 5 1.56% and 92.1 1%,respectively.The optimized group of combined detection was CEA +CA125+CA1 99 and CEA+CA125,and the sensitivity was 75.00% and 64.84%,respectively.Conclusion Single tumor markers have certain auxiliary value in diagnosing lung cancer.Combined detection could increase diagnostic sensitivity and accuracy for lung cancer, but its specificity may decrease accordingly,so clinical diagnosis should pay attention to whether there is the occurrence of false-positive.The combined detection of serum CEA,CA125 and CA1 99 could improve diagnostic sensitivity of lung cancer.
2.Tumor necrosing factor receptor-associated factor 2 gene deletion leading to fulminant liver failure and its mechanism
Chinese Journal of Infectious Diseases 2001;0(03):-
Objective To clarify the relationship between tumor necrosing factor(TNF) receptor-associated factor 2(TRAF2)gene deletion and liver failure and to explore the probable mechanism. Methods TRAF2 gene deficient mice(TRAF2~(lox/lox)/Cre~+,TRAF2~(lox/lox)/Cre~+/TNF~(-/-)) and control mice(TRAF2~(lox/lox)/Cre~-,TRAF2~(lox/lox)/Cre~-/TNF~(-/-))were simultaneously injected with PolyI:PolyC(300?L per injection one for 3 days,3 injections in succession)to induce TRAF2 gene knockout. The gene deletion was verified by polymerase chain reaction(PCR).The dynamic change of sera alanine aminotransferase(ALT)in animal model was investigated. These mice were sacrificed for liver when TRAF2~(lox/lox)/Cre~+ mice were severe sick and dying.Several pieces of liver tissues were speedily frozen, embedded by OCT and sectioned. Then, hematoxylin and eosin staining was performed. The pathological manifestation was investigated by light microscope.Expression of Fas and CD40 on hepatoeytes isolated from these mice' livers was determined by flow analyzers and cell sorters(FACS).The purified hepatocytes incubated with TNF(100?g/L)were stained by thiazolyl blue(MTT)at 30 rain,1 h,4 h和20 h respectively, then optics density values were determined. The proportion of viable hepatocytes was counted. Results TRAF2~(lox/lox)/Cre~+ mice were sick 5 to 9 days after injection with Poly(?):PolyC; and TRAF2 genes were successfully deleted.ALT started to increase at 2 days after injection, peaking at day 4(234.80?12.34)U/L and beginning to decrease before death. In TRAF2~(lox/lox)/Cre~+/TNF~(-/-) mice whose TRAF2 genes were also successfully deleted, although ALT levers also increased(90.30?15.63)U/L,the mice did not show any illness symptom up to day 15.Pathological results showed that there were obvious liver cells necrosis and inflamma- tion cells infiltration in TRAF2~(lox/lox)/Cre~+ mice livers. Moreover,Fas expression increased in TRAF2~(lox/lox)/Cre~+/TNF~(-/-)mice compared with that in control, hut CD40 decreased. Conclusions These results suggest that TRAF2 gene deletion can lead to hepatocytes death, and induce fulminant liver failure in mice.It is probably the reason why depletion of TRAF2 in hepatocytes sensitizes hepatocytes to TNF-mediated death
3.Relationship between Superoxide Dismutase and Gastric Cancer
Chinese Journal of Gastroenterology 2015;(5):308-310
Superoxide dismutase( SOD)is an important antioxidant enzyme, and plays an important role in active oxygen metabolism to regulate the balance between oxidation and anti-oxidation. Helicobacter pylori( Hp)infection, one of the risk factors of gastric cancer, can induce large number of reactive oxygen species by chronic inflammation, which causes DNA damage and high risk of gastric cancer. Expression of SOD is correlated with risk, metastasis and prognosis of gastric cancer. Studies on SOD can provide great help for the prevention and treatment of gastric cancer.
4.Advances in Study on CMTM3 and CMTM7 in Tumors of Gastrointestinal Tract
Chinese Journal of Gastroenterology 2015;(7):428-430
CMTM3 and CMTM7 are two newly discovered anti-oncogenes,which exist in various kinds of organs and tissues and having multiple biological functions including cell proliferation, differentiation, migration, and apoptosis. Recently,it has been shown that these two anti-oncogenes have closely relationship with gastrointestinal tract tumor, which can inhibit the growth of esophageal, gastric, and colorectal cancer. In this review article, the advances in study of the relationship between these two anti-oncogenes and tumors of gastrointestinal tract were summarized.

[Summary] Gastric cancer is seriously dangerous to human health with constantly high incidence .Human gastric mucosa can express gastrokines that can inhibit cancer cell growth .Gastrokines is expressed abundantly in normal tissues while downregulated or absent in gastric cancer and precancerous lesion .It plays an important role in the suppression of gastric cancer proliferation combining with the other factors such as helicobacter pylori infection or trefoil factors .In this paper we made a review about the suppression of gastrokines on gastric cancer cell proliferation .
8.Advances in Study on Bile Reflux Gastritis
Chinese Journal of Gastroenterology 2014;(5):305-308
Bile reflux gastritis (BRG)has been recognized as a chemical gastropathy due to excessive duodenogastric reflux (DGR).Abnormalities in pyloric anatomic structure,as well as antropyloric and duodenal dysmotility are considered to be implicated in the occurrence of pathologic DGR.Bile acid may induce apoptosis of gastric mucosal cells, and high concentration of bile acid plays a crucial role in the induction of intestinal metaplasia in stomach. In this review article, advances in study on BRG,including the mechanisms of DGR,the pathogenic effect of bile acid on gastric mucosa, and the diagnosis and treatment of BRG were summarized.
9.Endothelin-1 and stress ulcer in obstructive jaundice in rats
Chinese Journal of General Surgery 2001;0(07):-
Objective To determine the relationship between endothelin-l(ET-1)with blood flow of gastric mucosa and gastric mucosal injury (GMI) in obstructive jaundice rats induced by coldly restrainting stress. Methods 161 rats were randomly assigned into the experimental (obstructive jaundice) group (n=70), the control (sham operative) group (n=70) and the anti-ET-1 preconditioning group (n=21). The obstructive jaundice model was set up in the experimental group and the anti-ET-1 preconditioning group. Both the experimental group and the control group were re-assigned into 5 subgroups which underwent non-coldly restraint and coldly restraint for 10, 30, 60 and 120 min respectively. The anti-ET-1 preconditioning group underwent anti-ET-1 serum precondition 30,60 or 120min before the coldly restraint. The ET-1 was detected by radioimmunologic method and the gastric mucosal blood flow (GMBF) by laser Doppler flowmeter. Results The concentrations of ET-1 in gastric mucosa and ulcer index significantly increased (P
