1.DETERMINATION OF VITAMIN E IN EGG BY REVERSE PHASE HPLC

A method for separation and determination of vitamin E from egg by reverse phase HPLC was introduced.The accuracy and precision of this method were much better than the ordinary method. Standrd deviation of retention time and peak area were 0.012 and 146.92, and the corresponding coefficient of variation was 0.22% and 0.44% respectively. The recovery rate was 100.6 ?3%. This method was suitable for measuring VE in egg, milk powder and cod-liver oil.

The article summarizes the academic thought of Professor ZHANG Qi in treating rheumatism.In the cause and the pathogenesis of rheumatism,he thinks that disease invading and asthenia of health energy are the basic pathogenesis of rheumatism,besides the three evils:wind,cold and dampness,fever,the blood stasis,the phlegm stasis and blockade of collaterals have important meaning in rheumatism morbidity;In treating rheumatism,we must strengthen the body’s natural resistance and remove harmful factors,and use method of attacking(relieve wind,chill,dampness,fever and phlegm stasis),removing(smoothen blood and collateral circulation),reinfocing(regulating and nourishing yi-yang,qi,blood and zang-fu).In the prescription and medication,we must strengthen the body's natural resistance at the same time of removing harmful factors,systematic treating,make good use of the ancient Chinese medical classical recipe,treat the obstinate rheumatism,swelling and pain by the insect medicine.
3.Molecular biological characteristics of the transcription factor RORγt and the expression in immune-related diseases

RORγt, an immune cell-specific isoform of RORγ( retinoic acid receptor-related orphan nuclear receptor gam-ma) , is a key transcription factor for the development of Th 17 cells both in human and mouse .RORγt is required for the induction of IL-17 transcription and for the manifestation of Th 17-dependent autoimmune diseases in mice .RORγt natural ligands are retinoic acid . RORγt is closely implicated in the pathology of numerous autoimmune diseases , infectious diseases and cancer .With the further re-searches about the role of RORγt, we will clarify the mechanism of RORγt in autoimmune diseases , which will provide new ideas to di-agnose and treat autoimmune diseases .
4.The effect of prosthetic valve at preventing reflux esophagitis

Objective To approach the effect of prosthetic valve at preventing reflux esophagitis.Methods After the total gastrectomy and esophagojejunostomy,interrupted suture was apphed to form a prosthetic valve at output and input beyond Braun's stoma.Results Only one got light reflux esophagitis in 15 patients.Conclusion Prosthetic valve can prevent reflux esophagitis,and the operation is convenient and safe,which is deserved to be in generalization and approach the latest method.
5.A STUDY ON THE SYNTHESIS OF CELLULAR PROTEINS OF THE REPOPULATING RAT SPERMATOCYTE AFTER THE WITHDRAWAL OF GOSSYPOL TREATMENT

The synthesis of cellular and nuclear basic proteins in the first generation of the repopulated pachytene spermatocytes (RPS) after the withdrawal of gossypol treatment and the normal control pachytene spermatocyte (CPS) were studied by the incorporation of ~3H-arginine into proteins. After treatment with gossypol for various periods of time, the synthetic activity for cellular protein in the RPS was slightly inhibited. In comparison to CPS, the inhibitory rate ranged from 12% to 26% (gossypol treatment for 3 to 12 weeks, respectively). However, the synthetic activity for nuclear basic protein in the RPS was drastically reduced in comparison to that of the CPS. The reductive rate ranged from 46% to 61% in RPS (gossypol treatment for 3 to 12 weeks, respectively).Gel electrophoretic separation of the basic protein extracted from normal pachytene cell indicated that the major basic proteins were nucleosomal linker and core proteins, i. e., histone H_1, H_(2a), H_(2b), H_3 and H_4, and with a lesser amount of spermspecific BP and X_1 proteins. In the RPS, the synthesis of sperm-specific proteins(BP and X_1) and core histones (H_(2a) and H_4) reduced markedly.Finally, the effect of gossypol on the ratio change of nuclear basic protein to DNA in RPS and its relation to nucleosomal spacing and chromatin structure are discussed.
6.ISOLATION AND CULTURE OF NEURAL STEM CELLS FROM BRAIN OF RAT

Objective To investigate the effect of optimized conditions on proliferation and differentiation of the neural stem cells(NSCs) from different age brain of rat in vitro. Methods The brain tissues from embryonic(E12~E15d),newborn(1~2?d) hippocampus and olfactory bulb from adult of rat were isolated.After proliferation in conditional culture medium including growth factor and induced differentiation,the cell type specific antibodies were used for the immune fluorescence staining to identify the cells. Results A lager scale NSCs were obtained from brain tissue and could proliferate in conditional culture in vitro. They could self renew and generate both nerve and glia.Conclusion\ The NSCs got from rat brain can be used for basic and clinical research.\;[
7.Study on relationship between extravascular lung water,TNF-α,E-selectin,vasostatin-2 and the prognosis of patients with ARDS

Objective To analyze the correlations between acute respiratory distress syndrome ( ARDS) and extravascular lung water(EVLW),tumor necrosis factor alpha (TNF-α),E-selectin,vasostatin-2.To discuss the value of EVLW, TNF-α, E-selectin and vasostatin-2 in evaluating the severity of ARDS and prognosis of this disease . Methods There are thirty-six patients with ARDS were selected .According to the level of EVLWI shown by pulse indicator continuous cardiac output ( PiCCO) monitor,they were divided into the two groups:EVLWI≥14mL/kg group (n=18) and EVLWI<14mL/kg group(n=18).To monitor and record the levels of EVLWI ,PVPI,CI,ITBV,record oxygenation index ,calculate APACHEII score .The levels of TNF-α,E-selectin and vasostatin-2 in serum were meas-ured by ELISA.Results In EVLWI≥14mL/kg group,the APACHEⅡscore was (23.93 ±4.44),28-day mortality was 61.1%,the stay time in ICU was (19.05 ±4.0) d,which were higher than those in EVLWI <14mL/kg group [APACHEII score (18.58 ±3.66),28-day mortality(27.8%),the stay time in ICU(14.35 ±3.39) d](t=3.941,χ2 =4.050,t=3.797,all P<0.01).And the levels of TNF-α[(85.28 ±18.25)μg/L]and E-selectin[(74.07 ± 14.57)μg/L]in serum were higher than in EVLWI <14mL/kg group[TNF-α(63.27 ±20.28)μg/L,E-selectin (40.99 ±16.35)μg/L](t=3.422,5.847,all P<0.01).The oxygenation index of EVLWI≥14mL/kg group was (122.86 ±25.45),which was lower than that in EVLWI <14mL/kg group[(156.77 ±37.58)](t=-3.170,P<0.01).In the two groups of patients still alive within 28 days,after active treatment for 72 hours,the level of EVLWI was significantly lower than before in these 20 patients(Z=-0.392,P<0.01),oxygenation index was significantly higher than before(Z=-3.845,P<0.01),and the levels of TNF-αand E-selectin were significantly lower than before(t=7.194,4.025,all P<0.01).But 16 patients in the death group,after active treatment for 72 hours,the levels of EVLWI ,TNF-α,E-selectin and oxygenation index had no significant change ,there was no statistically signifi-cant difference(P>0.05).EVLWI had positive correlation with PVPI,TNF-αand E-selectin (r=0.605,0.649, 0.549,all P<0.01),and EVLWI had a negative correlation with oxygenation index (r=-0.715,P<0.01).Results showed no difference in vasostatin-2 between the two groups(P>0.05).Conclusion There were significant correla-tions between EVLWI and TNF-α,E-selectin in patients with ARDS.The levels of EVLWI,TNF-α,E-selectin were higher,and the mortality rate was higher,the the stay time in ICU was longer.The levels of EVLWI,TNF-αand E-selectin can provide a good reference value to the judgement of the severity of disease and prognosis in patients with ARDS.The level of vasostatin-2 had no correlation with EVLWI .

ObjectiveTo study the expression of Twist in invasive ductal carcinoma of breast and its relationship with cell proliferation and angiogenesis.MethodsThe expression of Twist and Ki-67 was detected in 60 cases with invasive ductal carcinoma of breast by immunohistochemistry.Ki-67 index and microvascular density(MVD) were calculated.ResultsThe positive expression rate of Twist was 56.7% (34/60) in invasive ductal carcinoma of breast.Ki-67 index and MVD in the patients with positive expression of Twist was higher than those in the patients with negative expression of Twist[(57.05 ± 16.37)% vs.(25.32 ± 16.16)%,(34.30 ± 12.25)% vs.(23.04 ± 10.45)%,P< 0.05 ].ConclusionsThe overexpression of Twist is found in invasive ductal carcinoma of breast.To stimulate cell proliferation and angiogenesis may be one of the pathways of Twist to contribute to the invasion and metastasis of breast cancer.
9.Analysis of the level and significance of IL-16 and IL-17 in nasal secretion and in serum of patients with allergic rhinitis and non-allergic rhinitis.

OBJECTIVE:
To test the immunoglobulin free light chain (FLC) from nasal secretion and serum of patients with allergic rhinitis(AR)and non-allergic rhinitis(NAR) for the purpose of exploring the possible immunological mechanism.
METHOD:
Ninety consecutive patients were selected between January 2009 and January 2012, involving 45 patients with AR and 45 patients with NAR diagnosed by symptoms,signs,skin prick tests(SPT) and specific IgE (slgE). Forty-five volunteers were chosen as healthy control (HC). According to the visual analogue scale (VAS) scores,the nasal symptoms of AR and NAR,including sneeze. Nasal discharge. Nasal obstruction and nasal itching were compared. ELISA was used to detect the total IgE, IL-16, IL-17 in nasalsecretion and serum. The data was analyzed by SPSS 17.0 software.
RESULT:
There was no statistical difference between AR and NAR group in nasal symptoms (P > 0.05); In serum, IL-16 and IL-17 increased in AR group comparared to NAR group (P < 0.05); IL-16 and IL-17 increased in NAR group comparared to HC group (all P < 0.05); In nasal secretion, IL-16 and IL-17 increased in NAR and AR group comparared to HC group (all P < 0.05).
CONCLUSION
IL-16, IL-17 takes part in the path of physiological process of AR and NAR with the immunological mechanism.

10.Correlation between soluble E-selectin and interleukin-8 levels and the development of severe pneumonia

Objective This study was designed to evaluate the correlations between soluble E-selectin (sE-selectin) and intedeukin-8 (IL-8) levels and the condition and prognosis of severe pneumonia. Methods A total of 67 patients with pneumonia were identified at the intensive care unit and the Respiratory Department of Zibo Central Hospital between April 2007 and March 2008. The patients were divided into two groups according to the severity of pneumonia: severe (Group A, n = 35) and non-severe (Group B, n = 32). Group A was also subdivided into two groups of patiems: patients with Multiple Organ Dysfunction Syndrorae(MODS) (Group A_1, n = 13) and pa-tients with severe pneumonia alone (Croup A_2, n= 22). Thirty healthy people whose age and sex matched with the patients were enrolled as a control group (Group C). Patients with cancer, who had undergone surgery within the past 1 month, connective tissue disease or acute conplications of diabetes, for example, were excluded from the study. The serum levels of s-Eselectin and IL-8 were measured by EI.ISA, and correlations with Acute physiol-ogy and chronic Health Evaluation Ⅱ (APACHE-Ⅱ)score, Oxygenation index(PaO_2/FiO_2), percentage of poly-morphonuclear leukocyte (PMN)and high sensitive C-reactive protein (hs-CRP)levels were determined. The data were analyzed using t tests, one-way ANOVA, X~2 tests and linear correlation analysis using SAS 8.2 software. Results The sermn levels of sE-selectin and IL-8 in Croup A and Group B were significant higher than Group C (P < 0.05), and the levels in Group A were higher than those in Group B (P < 0.05). Furthermore, the levels of sE-selectin and IL-8 were higher in Group A_1 than in Group A_2(P < 0.001). The correlation analysis showed a positive correlation between the levels of sE-selectin and IL-8 in patients with pneumonia (r = 0.781,P < 0.01) ; and both were positively correhted with APACHE-Ⅱ score, PMN% and hs-CRP (P < 0.01), and nega-tively correlated with PaO_2/FiO_2 (P < 0.01).Conclusions sE-selectin and IL-8 levels are important indices for the assessment of the severity of pneumonia in tetras of the condition and prognosis.
