Objective To explore the source of endothelial cells in tumor vessels by A 549 tumor model with GFP nude mouse. Methods To establish the A 549 lung cancer models with GFP nude mice, expression of CD 31 was determined by immunofluorescence to label tumor vessels; to observe and take a picture of the tumor frozen section by confocal microscopy and invert microscope;expression of GFP in tumor vessels was determined by immunohistochemistry. Result The results of immunofluorescence showed:Tumor interstitial vascular endothelial cells or endothelial cells clusters and micro-vascular lumen size and shape are clearly visible by immunofluorescence, and part of vessels with no obvious lumen or irregular lumen. We can see green fluorescent in tumor cells of tumor tissue and endothelial cells which form of tumor vessels. The results of immunohistochemistry showed: expression of GFP was determined in cytoplasm of tumor stromal cells and endothelial cells in tumor vessels. Conclusion The endothelial cells which formed tumor neovessels that derived from GFP nude mice partly and the other part derived from tumor cells.
3.Gene expression of osteoprotegerin and osteoclast differentiation factor in giant cell tumor.
Chinese Journal of Pathology 2002;31(2):128-131
OBJECTIVETo investigate the gene expression of osteoprotegerin (OPG) and osteoclast differentiation factor (ODF/TRANCE/RANKL), two new members of the TNF-receptor superfamily, in giant cell tumor (GCT); to discuss the molecular mechanism of extensive bone resorption caused by GCT.
METHODSUsing TRIzol reagent to prepare total RNA from GCT sample and normal bone tissue. By a first-strand complementary DNA (cDNA) synthesis kit, cDNA was synthesized from 2.0 micro g RNA according to the manufacturer's instructions. cDNA was then amplified by PCR. Amplification products were resolved by electrophoresis on a 1.5% agarose gel and stained with EB. The relative quantity of the PCR products were determined and the mRNA levels of OPG, ODF, M-CSF (cofactor of ODF), and RANK (receptor of ODF) were compared with that of the normal bone.
RESULTSGCT contained highly expressed mRNA of ODF, OPG, M-CSF and RANK. There was mRNA expression of OPG, M-CSF and RANK and less expression of ODF in normal bone. The ODF mRNA and RANK mRNA in GCT were more abundant than that in normal bone. In GCT, the ratio of ODF mRNA exceeded OPG expression. But in normal bone, the OPG mRNA exceeded ODF expression.
CONCLUSIONSThe results suggest that GCT contains all signals including OPG, ODF, M-CSF and RANK that are essential for inducing osteoclastogenesis and promoting bone resorption.

Objective:To design the mode of supervision, forwarn and communication system in barrier enviroment of laboratory animal. Methods and Results:Distribution inspect system of industrial computer center was used for live monitoring, displaying, reserving, printing temperature humidity, pressure, difference and ammonia density, according to necessity. The upper and low limit of parameter was set up according to national standard of laboratory animal,if parameter surpass the limit,it will warn the people on duty in multiple spots.Working and animal illumination were established respectively on the basis of illuminace light period of standard, the switch of working illumination was controlled by the people, but the later was controlled, automatically and slowly in order to disinterfere with physiology activity of animal. The mode of moniter add video together with multiple pickup camera in cable television were used, which binded wirh the intellectual industral control group. The communication system was designed using miniature program control changer and terminal telephone, which joined the unit inner telephone to communicate with outside. The double and stereophonic was used to install background music in this enviroment, which was timely played with suitable volume. Conclusion:This system provided the method for the administration of laboratory animal standardization?mathematics and informatics.

To observe the effect of various doses of oil of Piper longum unsaponifiable matter (OPUM) to cholesterol gallstones in experimental mice. C57BL/6 mice (n = 60) were randomly divided into 6 groups: control group, model group, OPUM (15, 30 and 60 mg x kg(-1)) group and ursodeoxycholic acid (UDCA, 60 mg x kg(-1)) group, administered for 10 weeks. The level of serum lipid and liver function enzymes were tested. The gallbladder was removed and bile was obtained by centrifugation. Next, the levels of the bile total cholesterol (TC), phospholipid (PL) and bile acid (TBA) were measured. The indicators of lipid peroxidation were determined and cholesterol saturation index (CSI) was calculated. The liver histological changes were observed by HE staining. The results showed that serum TC, TG (triglycerides) and AST (aspartate transaminase) contents, gallbladder cholesterol crystallization and CSI increased significantly (P < 0.05). In addition, the activity of SOD decreased significantly and MDA content increased significantly in liver (P < 0.05). HE staining results showed that the hepatic cord disorder and intracellular lipid droplets increased significantly. All results indicate that lithogenic diet lead to the formation of cholesterol gallstones. In OPUM (30 and 60 mg x kg(-1)) group, serum TC, TG and AST content, gallbladder cholesterol crystallization and CSI decreased significantly, the activity of SOD increased significantly and MDA content decreased significantly. HE staining results showed that OPUM can improve the morphology of liver cell, reduce the degree of hepatic cord disorders and restore the cell morphology close to normal. The cause of OPUM prevents cholesterol gallstone formation maybe due to protect the integrity of the liver cells, lower CSI, and reduce cholesterol crystal formation and hence prevent cholesterol gallstone formation.

Chinese Journal of Traumatology 1999;2(1):3-7
OBJECTIVE: To observe the expression and cellular localization of interleukin-8 (IL-8) mRNA and protein in the area of xenogenic bone implant. METHODS: The bovine cancellous bone granules were implanted into the thigh muscles of mice. The samples were taken 4, 7, 14 and 21 days after implantation. IL-8mRNA and protein in the site of implant were assayed by in situ hybridization and immunocytochemical techniques. RESULTS: The expression of IL-8mRNA and protein were observed in all specimens 4, 7, 14 and 21 days after implantation. IL-8mRNA was expressed mainly by the neutrophils, monocytes, macrophages and fibroblasts at 7th day post-implantation. Some mesenchymal cells, multinucleated giant cells, vascular endothelial cells and smooth muscle cells also expressed IL-8mRNA in the area of xenogenic bone implant at 14th and 21st days. Immunocytochemical studies demonstrated the same results as that of in situ hybridization. CONCLUSIONS: Many different kinds of cells express IL-8mRNA and secret IL-8 in the area of xenogenic bone implant, suggesting that IL-8 may play an important role in local immunity of xenogenic bone graft.
