1.Operative treatment of humeral super-middle fracture combined with brachial artery injury in 9 cases

Objective To investigate the clinical characteristics and operative treatment of humeral super-middle fracture combined with brachial artery injury. Methods 9 cases of humeral super-middle fracture combined with brachial artery injury have been treated in our department since 2000 with reduction, axial external fixation, and autotransplantation of great saphenous vein to repair brachial artery. Results All patients were followed up for 1 to 2 years. No stenosis occurred at the anastomosed veins and the limbs survived well. All fractures achieved clinical healing except 1 case who had nonunion following refracture and had to undergo secondary operation. Conclusion The operation is simple and easy, but effective in treatment of the humeral super-middle fracture combined with brachial artery injury.
2.The primary study on the newborn screening of congenital hypothyroidism with the dry blood spot in the filter paper by surface enhanced Raman spectroscopy

Objective To develop a method of detecting dry blood spot (DBS) samples collected in the filter paper by using Surface Enhanced Raman Spectroscopy (SERSp,also known as Raman molecular fingerprint spectrum) technology.The possibility and reliability of applying the technology to the newborn screening of congenital hypothyroidism (CH) was investigated.Methods The case-control study was used.The results of CH-hTSH screening test based on the DBS samples collected in the filter paper,and the clinical diagnosis were all provided by neonatal screening center of Shanghai Children's Hospital.The results of SERSp analysis were provided by Dalian University of Technology.Six positive and six negative samples,which were confirmed respectively by CH-hTSH screening using time resolved fluoroimmunoassay (TRFIA) were chosen.With the spectrum lines of 725 cm-1 as internal standard,the SERSp from the aqueous solution of these samples were analyzed.The specific spectrum line analysis,the principal component analysis (PCA) and the scatter diagram of PCA were used to compare the results of TSH test.Results With the spectrum lines of 725 cm-1 as internal standard,the CH specific spectrum lines of 1373 cm-1 and 1400 cm-1 were discovered.They showed the most significant difference of the peak height between the positive and negative samples of CH,while TSH specific spectrum lines of 785 cm-1,827 cm-1 and 853 cm-1 failed to distinguish positive from negative samples with distinct peak height.The consistency between the results of CH-Raman Screening and CH-hTSH Screening was 91.7% (11/12),which was proven by PCA,the scatter diagram of PCA and the specific spectrum line analysis.The sensitivity of CH-Raman Screening was 100% and the specificity was 83.3%.Conclusion There is a new CH-Raman Screening method using the SERSp analysis,which was proven to be a promising technology in the newborn screening of congenital hypothyroidism with the DBS in the filter paper.
3.The relationship between the degradation of five RNAs and early PMI in brain tissue of rats with different causes of death

Objective To establish a mathematical model of early PMI and RNA △Cq under different causes of death,which provides a reference for early PMI inference under the condition of changing causes of death.Methods Thirty adult male SD rats with SPF level were randomly divided into 5 groups with 6 animals in each group(Death by carotid artery major hemorrhage;Death by strangle;Death by drown;Death by organophosphorus pesticides;Death by CO poisoning).The rats were placed in a constant temperature and humidity incubator at 20℃and 55%humidity after death.RNA was extracted from brain tissue at 7 time points within 0~24 h postmortem.The Cq values of five RNA(β-actin,GAPDH,miR-9,miR-124,miR-125b)were measured at each time point.QBase+V(3.4)software geNorm program was used to select the internal parameters.The correlation between early PMI and RNA △Cq was analyzed by SAS 9.4 software,and a mathematical model was established.Another 15 adult male SD rats with SPF level were randomly divided into 5 groups.The RNA from brain tissue were collected at 10 h and 20 h after death for back-inference verification.Results(1)The fluorescence of miR-124 could not be captured in this experimental system,and this gene could not be used as a candidate gene in this experimental system.(2)The stability of the four genes(β-actin,GAPDH,miR-9 and miR-125b)in rat brain tissue varied under different conditions of death.Using SAS 9.4 software,the correlation analysis were conducted between early postmortem interval(PMI)and RNA △Cq values,and the mathematical models were set up for early PMI estimation.Conclusion(1)The causes of death can affect the degradation of RNA in postmortem brain tissue of rats,and different reference genes should be selected under different causes of death.(2)The mathematical model established under this experimental system can provide reference for early postmortem PMI inference of different causes of death.
