1. Advance in Regulation of Expression of Brain Aquaporin 4 (review)
Chinese Journal of Rehabilitation Theory and Practice 2007;13(8):724-726
The expression of brain aquaporin 4 (AQP4) is different at the different stages of cellular differentiation, and distribution of brain AQP4 is affected by the existence of endothelial cells. Other factors including osmolality, ammonia, hypoxia, temperature, hormone, C-type natriuretic peptide, lead, complement inhibitor, lipopolysaccharide can also influence AQP4 expression in astrocytes. However, the regulating mechanisms for brain AQP4 expression are not clear. Protein interaction, protein kinase C (PKC) through protein phosphorylation, mitogen-activated protein kinase signal pathway, Ca2+ signaling cascade signal pathway and transcription factor pathway have been proposed, among which phosphorylation of PKC for the inhibition of AQP4 is frequently studied.

AlM: To investigate the relationship among carotid atherosclerosis, homocysteine ( Hcys ) and anterior ischmic optic neuropathy( AlON) .METHODS: Sixty-five AlON patients enrolled in AlON group and 70 non - AlON controls enrolled in control group. All the participants in 2 groups were examined by carotid artery color doppler flow imaging to evaluate the incidence of carotid atherosclerosis. Plasma levels of Hcys, vitamin B12 and folate were measured by automatic biochemical analyzer. The incidence of carotid atherosclerosis, plasma levels of Hcys, vitamin B12 and folate were compared statistically between AlON and control group.RESULTS: Fifty - six AlON patients ( 86%) exhibited carotid atherosclerosis, which was found in 43 control subjects ( 61%) . Fifty - nine AlON patients ( 91%) exhibited increased plasma levels of Hcys compared with that in 38 control subjects ( 54%) . Lowered vitamin B12 was found in 37 patients with AlON ( 57%) and in 43 control subjects ( 61%) . Lowered folate was found in 54 AlON patients ( 83%) and in 32 control subjects ( 46%) . The incidence of atherosclerosis in carotid artery ( P =0. 001), mean Hcys level (P=0. 0005) and lowered folate ( P = 0. 0006 ) were different statistically between AlON group and control group. Mean vitamin B12 level was indifferent between AlON group and control group ( P=0. 0722). Logistic regression analysis showed that carotid atherosclerosis was an a risk factor for AlON (OR=1. 312, 95%CI:0. 927~1. 772), whereas elevated Hcys level (OR=2. 378, 95% CI: 1. 479 ~ 3. 821 ) was also shown to be significant risk factor for AlON. CONCLUSlON: Carotid atherosclerosis and elevated Hcys were related with AlON. Reducing above-mentioned risk factors related to thrombus may be crucial to the prevention and cure of AlON.

Objective:To investigate the expression and correlation of cyclooxygenase-2(COX-2) and c-erbB-2 in human breast cancer. Methods: The expressions of COX-2 and c-erbB-2 in 48 breast cancer patients with breast cancer were detected with immunohistochemistry examination and was analyzed. The relationship between their expression and the clinicopathologic characters of breast cancer was also analyzed. Results: The positive rate of COX-2 was 60.4%(29/48) in these patients;the expression was higher in patients with tumor size larger than 2 cm,lymph node metastasis, and negative expression of ER or PR,but was not related with the ages, the pathologic types and the TNM stages.Expression of c-erbB-2 was identified in(39.6%)(19/48) of these patients;the expression was significantly higher in patients with lymph node metastasis or negative expression of ER and PR,but was not related with the ages, the sizes and the pathologic types.COX-2 was positive in 84.2%(16/19) of patients positive of c-erbB-2 and in 44.8%(13/29) of patients negative of c-erbB-2,with significant difference found between the 2 figures(P=0.006).Conclusion: The high expression of COX-2 in breast cancer is related to the expression of c-erbB-2,suggesting that they may regulate each other and jointly contribute to the tumorigenesis and progression of breast cancer.

Objective To investigate the molecular mechanisms for the development of cardiac hypertrophy in hypertension, the present study provided the differential protein expression analysis of hypertrophied heart at differ- ent stages in spontaneously hypertensive rats (SHR).Methods The profiles of protein expression of left ventricu lar myocardium in SHR and its normotensive control Wistar-Kyoto (WKY) rats at the age of 4 and 20 weeks were analyzed with two-dimensional gel electrophoresis (2-DE) in combination with matrix assisted laser desorption ioniza- tion-time of flight (MALDI-TOF-TOF) mass spectrometry. Results Although the blood pressure of SHR was normal at 4 weeks age, hypertrophy of the left ventricle had already developed. The expression pattern in the hy- pertrophic myocardium was found 27 modulated proteins,20 of which were identified. These proteins are involved in reactions of energy metabolism, mitoehondrial oxidative phosphorylation and oxidative stress, etc. The expres- sion of 13 proteins was significantly changed in SHR rats at early stage prior to the development of sustained hyper- tension, while the expression changes of other 7 proteins occurred only at late stage in SHR rats when the blood pressure was significantly elevated. Conclusions lncrease in glycolysis and decrease in oxidation of fatty acid and glucose was shown in the hypertrophied myocardium from early stage in SHR prior to the development of hyperten sion. The significant changes in protein expression of the mitochondrial electron transport chain and antioxidative molecules support the hypothesis that oxidative stress promotes and accelerates the development of hypertensive car- diac hypertrophy.

Chinese Journal of Rehabilitation Theory and Practice 2010;16(1):29-31
ObjectiveTo determine whether glutamate induces the alteration of aquaporin 4 (AQP4) expression in cultured rat astrocytes. MethodsThe secondary cultured astrocytes were treated with 1 mmol/L L-glutamate for 1 h, 3 h, 6 h, 12 h, 24 h and 48 h. The morphologic changes of astrocytes were observed through microscope after GFAP immunostaining and AQP4 mRNA expression were detected with real-time PCR. ResultsThe astrocytes swelled when exposed to glutamate for 1 h and remained with prolonged treatment. Meanwhile, the AQP4 mRNA expression were early down-regulated and subsequently up-regulated, featured with the lowest AQP4 mRNA level at 12 h after treatment (P<0.01) and higher at 48 h (P<0.05). ConclusionAquaporin 4 may be involved in the occurrence and development of astrocyte swelling induced by glutamate.

Objective To reproduce a scratch-wound model in cultured rat astrocytes (AST).Methods The secondary cultured AST prepared from newborn Wistar rat cerebral cortex were scratched with plastic pipette tips. The morphologic change of AST was observed through microscope at 10 min before and 1, 3, 6, 12, and 24 h after injury, meanwhile the lactate dehydrogenase (LDH) leakages in the cultured medium were determined. Results Immediately after injury the edge of the scratch was lined with irregularly shaped cell. 6 h after injury the AST processes began extending to cell-free area, and elongated further at 12 and 24 h after injury, with presented of new generated cells in the denuded area. At different times after injury, the LDH leakages of the experiment group were higher than that before injury ( P<0.05), and were higher than that of the control group ( P<0.05).Conclusion According to observed AST morphologic changes and determined LDH leakages in culture medium, the scratch-wound model in cultured rat AST is successfully reproduced.
