2.Anti-tumor and Anti-metastasis Effects of Tumor Cells Transduced with the Gene, Cording for Human Interleukin-2

Reccent investigations demonstrated that genetically modified tumor cells could be used as vaccines against cancer. In this light, we introduced human interleukin -2 (IL-2) gene into a poorly immunogenic rat fibrosarcoma cell line WBT, a human lung abenocarcinoma cell line AGZY and a highly metastatie sub - cell line Anip973 derived from AGZY. The transduced cells showed no obvious changes in morphology, growth and colony - forming ability in soft agar.Transduction with IL-2 gene rendered the cells more sensitive than the parental cells and cells transduced with NeoR gene alone to the lysis by cytotoxic lymphocytes. High levels of IFN and TNF were detected in the supernatants of cocultured lymphocytes and tumor cells containing IL-2 gene. Tumorigenicity of WBT/IL-2 cells was greatly reduced in syngeneic rats. Metastatic capacity of Anip973 in nude mice was blocked by the transduction of IL-2 gene, but not the transduction of NeoR gene alone. Inoculation of WBT/IL-2 cells conferred rats resistance to the challenge of parental WBT cells.Splenocytes from WBT/IL-2-inoculated rats restimulated with WBT cells in vitro showed high CTL activity against WBT cells. Our results demonstrated the potential application of tumor cells transduced with IL— gene as active vaccine against cancer.

Objective:Combined laparoscopy and hysteroscopy in the diagnosis and therapy of the female infertity is and advanced means in the gynecologcial field.We analysed the causes and the treatments of the 71 infertile patients.Pelvic adhesions,endometrosis and salpingtis were the main causes of infertity in our 71 cases.Of 71 cases,29 cases were primary infertity,in whichc endometriosis,PCOS and anatomic uterine defect were the main causes,and 42 cases were secondary infertity,in which 35 cases (83%) had previous intrauterine operations.All the patients were treated for the causes.We concluded that combined laparoscopy and hysteroscopy can diagnose the causes of infertity and can observe the tubal patency by hysteroscopic tubal cathertetization and hydrotubation,which will direct the treatments and estimate the results of the therapy.Methods:Infertity;Combined Laparoscopy and Hysteroscopy.

Objective To investigate the effects of IFN ? and Somatostatin analog octreotide (SMS) on hormone secretion by cultured human GH secreting pituitary adenomas.Methods Each cultured GH secreting pituitary adenoma cells were equally distributed to culture tube and divided into 4 groups: control, IFN ?, SMS, and IFN ?+SMS. Different drugs were added into different groups. The hormone secretion in each group were detected after 2 and 4 days' incubation.Results After 2 and 4 days' incubation with IFN ?(100 IU/ml) GH secretion was significantly inhibited in 6 of 10 and 7 of 10 pituitary adenoma cultures ,reduced by 22%~54% and 30%~61% vs control ( P
9.Interventional therapy of traumatic pseudoaneurysm

Objective To introduce the different therapeutic methods for the different types of traumatic pseudoaneurysms. Methods Traumatic pseudoaneurysm was treated in 8 patients. Intraarterial embolization was performed with gelfoam in 3 patients for terminal type. Endovascular covered stent graft was inserted in 2 patients with trunk type. One patient with branch type, we used metallic coils to embolize both broken ends. One patient with perforation in femoral artery was treated with compression. For 1 patient with huge haematoma and pseudoaneurysm, operation for elimination the haematoma was performed after embolization. Results Pseudo- aneurysm kept closed after treatment achieved in all 8 patients. Distal arterial pulse was normal in 2 patients treated with stent graft. A new pseudoaneurysm presented as a complication at puncture site in 1 patient due to other factors. There were no other symptomatic complications. Conclusions Pseudo- aneurysms can be treated effectively with interventional techniques instead of surgical operation.
10.The significance of the changes of HBV DNA and it's antigen and antibody in 35 cases with liver transplanting

Objective To study the changes and significance of HBV DNA and it's antigen and antibody in patients with liver transplanting.Methods 35 cases with liver transplanting in 302th hospital from Oct.2006 to Mac.2007 has been studied.The HBV antigen and antibody has been studied by ELISA in 188 samples including one from before transplanting and 4 to 7 from after transplanting.The quantitative of HBV DNA has been detected by real time PCR technology.45 cases of hepatitis B,whose sera HBV DNA is positive no more than 10 000U/ml,treated with lamifudine in 2 weeks about has been used in controls.Results 95(34/35)percent of liver-transplanting patients acquired serum-transfer in HbsAg to it's antibody,and 6 patients with HBeAg positive acquired from HbeAg to it's antibody in 2 days after transplanting.The HBV DNA in sera is distinguished of 34 cases from 35s in 6 months after liver-translpanting.Only one patient is insisting with HBsAg and HBV DNA positive.In lamifudine treatment controls,none of patients acquired sera-transferred from HBsAg to antibody in 45 cases.The HBV DNA is decreased from positive to negative under cove in 11 controls.29 cases of HBV DNA are decreased in 2 to 3 Log E after treatment with lamifudine.The HBV DNA is insisting in high levels in 5 patients.The HBeAg is negative in 29 cases after lamifudine treatment,but it's antibody is not acquired serum transferring.Conclusion The HBV DNA,HbsAg and HbeAg in patients with HBV infection may be transferred into negative forever transplanting with healthy liver underlined in HBIG and lamfudine treatment.
