1.Effects of different intensities of treadmill running on bone composition in rats

Objective To evaluate the effects of different intensities of treadmill running on changes in tibial bone composition in a rat model.Methods A total of 24 female Wistar rats were randomly assigned into 4 even groups (n =6) of free movement (FM),low-intensity running (LR),medium-intensity running (MR),and high-intensity running (HR).Rats in LR,MR,and HR groups underwent treadmill running with respective intensities (15 m/min and a 0° gradient,20 m/min and a 5° gradient,25 m/min and a 10° gradient) for 8 weeks (1 h/d and 5 d/week).FM group were taken as controls.The cortical and trabecular bone at the left tibial metaphysis was harvested after 8 weeks in each group for Raman spectroscopy to analyze mineral-matrix ratio,carbonate-phosphate ratio and crystallinity.Results In MR group,the mean mineral-matrix ratio was 4.883 ± 0.128 for cortical bone and 4.216 ±0.213 for trabecular bone,both significantly higher than those in FM group (4.113 ±0.132 and 3.773 ±0.122) (P < 0.05).The mean mineral-matrix ratio was 3.222 ± 0.329 for trabecular bone in HR group,significantly lower than that in FM group (P < 0.05).In MR group,the mean carbonate-phosphate ratio was 0.166 ± 0.013 for trabecular bone,significantly lower than that in FM group (0.177 ± 0.011) (P < 0.05).In HR group,the mean carbonate-phosphate ratio was 0.195 ± 0.012 for cortical bone and 0.187 ± 0.010 for trabecular bone,both significantly higher than those in FM group (0.183 ± 0.014 and 0.177 ± 0.011) (P < 0.05).In HR group,the mean crystallinity was 0.0521 ± 0.0012 for cortical bone and 0.0522 ± 0.0017 for trabecular bone,both significantly higher than those in the other 3 groups (P < 0.05).Conclusions Running with medium intensity may increase bone mineralization and turnover,thus enhancing bone quality.However,running with high intensity may decrease bone mineralization and thus bone quality.
2.Therapeutic effect of Hcy-lowering therapy on patients with coronary heart disease after PCI

Objective:To analyze therapeutic effect of homocysteine (Hcy)‐lowering therapy on serum levels of Hcy and inflammatory factors in patients with coronary heart disease (CHD ) after percutaneous coronary intervention (PCI) .Methods :A total of 82 CHD patients who received PCI in our hospital were selected .According to random number table ,they were randomly and equally divided into routine treatment group (received routine postoperative therapy) and Hcy‐lowering group .Serum levels of Hcy ,inflammatory factors ,N‐terminal pro‐brain natriuretic pep‐tide (NT‐proBNP) and soluble intercellular adhesion molecule‐1 (sICAM‐1) were compared between two groups . Results :1) On six months and one year after treatment ,compared with routine treatment group ,serum Hcy level significantly reduced [after six months: (15.39 ± 1.83) μmol/L vs . (13.21 ± 1.35) μmol/L ,after one year :(15.61 ± 1.62)μmol/L vs . (8.73 ± 0.72)μmol/L] in Hcy‐lowering group ;2) after six‐month treatment ,compared with routine treatment group ,there were significant reductions in serum levels of CRP [ (67.27 ± 7.51) mg/L vs . (37.11 ± 6.32) mg/L] ,IL‐6 [ (87.58 ± 7.21)μg/L vs . (60.17 ± 5.45)μg/L] ,procalcitonin [PCT , (21.34 ± 3.04) ng/L vs .(15.61 ± 2.32) ng/L] ,NT‐proBNP [ (298.37 ± 53.28) pg/ml vs .(104.28 ± 13.17) pg/ml] and sI‐CAM‐1 [ (391.83 ± 75.04) ng/ml vs .(162.18 ± 30.26) ng/ml] in Hcy‐lowering group , P<0.05 all .Conclusion:Hcy‐lowering therapy is help to reduce serum Hcy level ,relieve systemic inflammatory response and protect myocar‐dial function in CHD patients after PCI .
3.Intensity-dependent effect of treadmill running on type Ⅱ Collagen of knee articular cartilage in rats

Objective To investigate the effect of treadmill running with different intensities on type Ⅱ collagen (Col2) of knee joint articular cartilages in rats.Methods A total of 48 adult Sprague-Dawley rats were randomly divided into a control (C) group,a low-intensity exercise (L) group,a moderate-intensity exercise (M)group and a high-intensity exercise (H) group,each of 12.Rats in three exercises groups were regularly trained on treadmill at low,moderate,and high intensities respectively.Eight weeks later,all the animals were sacrificed.The right tibial plateau samples were collected to observe collagen fibers under polarizing light microscopy,and the collagen Ⅱ content were examined using immunohistochemistry.The mRNA expression of biglycan (BGN),fibromodulin (FMOD) and Col2 was tested using the quantitative real-time reverse transcription-polymerase chain reaction.Results Compared with group C,collagen fibers in group L and M exhibited almost the same organization,whereas,alteration in organization and shape of collagen fibers was found in group H.Significantly lower content of type Ⅱ collagen was found in group H than that in group C.In comparison with group C,group L had significantly higher gene expression of Col2,whereas group H had significantly higher BGN mRNA expression.Conclusion Low-or moderate-intensity treadmill running appears to have beneficial effect on articular cartilages to maintain its integrity.Highintensity exercises induce lower content and disorder of type Ⅱ collagen in articular cartilages,but the self-healing of cartilage may still exist.
4.Therapeutic effect of rosuvastatin on inflammatory factor levels,insulin resistance and vascular endo-thelial function in patients with hypertension complicated dyslipidemia

Objective:To analyze therapeutic effect of rosuvastatin on inflammatory factor levels ,insulin resistance (IR) and vascular endothelial function in patients with hypertension complicated dyslipidemia .Methods:A total of 136 outpatients with hypertension complicated dyslipidemia were selected .According to random number table ,they were divided and equally into routine treatment group (received routine therapy ) and rosuvastatin group (received rosuvastatin based on routine treatment ) .Serum levels of C reactive protein (CRP) ,interleukin‐6 (IL‐6) and IL‐8 , IR and vascular endothelial function were compared between two groups before and after treatment .Results:Com‐pared with before treatment ,there were significant reductions in serum levels of CRP ,IL‐6 and IL‐8 ,homeostasis model‐insulin resistance index (HOMA‐IR) and number of endothelial microparticles (EMPs) ,significant rise in in‐sulin sensitivity index (ISI) and flow‐mediated dilation of brachial artery (FMD) after treatment , P< 0.05 all . Compared with routine treatment group ,there were significant reductions in serum levels of CRP [ (67.27 ± 7.51) mg/L vs .(37.11 ± 6.32) mg/L] ,IL‐6 [ (87.58 ± 7.21)μg/L vs .(60.17 ± 5.45)μg/L] and IL‐8 [ (121.31 ± 8.57)μg/L vs .(84.44 ± 5.21)μg/L] ,HOMA‐IR [ (3.08 ± 0.51) vs .(2.31 ± 0.47)] and number of EMPs [ (852.18 ± 115.37) /μl vs .(573.29 ± 72.18)/μl] ,and significant rise in ISI [(-4.39 ± 0.61) vs .(-3.42 ± 0.53)] and FMD [ (4.35 ± 0.52)% vs .(5.82 ± 0.69)% ] in rosuvastatin group after treatment ,P<0.05 all .Conclusion:Rosuvasta‐tin could reduce inflammatory factor levels ,relieve insulin resistance and improve vascular endothelial function in patients with hypertension complicated dyslipidemia .

Objective The clinical effect of the lateral femoral cutaneous artery flap for soft tissue defects of leg wounds. Methods From October 2007 to January 2016, VSD was firstly used to promote the growth of granulation tissue. When the growth of granulation tissue became satisfactory, flaps were designed based on the anatomical characteristics of the lateral femoral cutaneous artery. We repaired 20 cases of wound defects by cutting flaps that coincide with the recipient vessels. Result 20 cases were followed up for 6 to 24 months, 12 months on average. All flaps were survived and only one case had small area of necrosis flap which was healed by replacing medicines. In all cases, wounds were healed and flaps showed good color and good texture. The strength of quadriceps muscle was good and the extension of knee flexion was 0° to 150°. Conclusion To The lateral femoral cutaneous artery flap is used for soft tissue defects of refractory wounds on leg , flap donor sites are sutured directly, the treatment period is shorten and the method is safe and effective. The lateral femoral cutaneous artery flap is one of ideal choices for wound tissue defects.
6.Level and clinical value of serum cystain C in patients with HBeAg-negative chronic hepatitis B

Objective To investigate the change and clinical value of serum cystain C(CysC)level in the patients with HBeAg-negative chronic hepatitis B(CHB).Methods 212 cases of CHB in our hospital were selected and divided into the HBeAg-negative group and the HBeAg-positive group according to the HBeAg status.105 healthy individuals at the same time were enrolled as the control group.The levels of serum CysC,ALT,AST,Cr,BUN and HBV DNA in 3 groups were detected and compared.The corre-lations between serum CysC with AST,AST,Cr,Bun,HBV DNA were analyzed by the Pearson correlation analysis.Results The mean CysC level in the HBeAg-negative group was (1 .40±0.36)mg/L,which was higher than (0.93±0.12)mg/L in the control group and (0.96±0.18)mg/L in the HBeAg-positive group,the differences between them was statistically significant(P <0.05). The CysC was significantly positively correlated with serum Cr level in the HBeAg-negative group(r=0.840,P <0.01).Conclusion The CysC level might be used as an effective index for monitoring the early kidney injury in the patients with HBeAg-negative CHB,which has important significance in the prevention and treatment of HBeAg-negative CHB.
7.Effects of cyclic tensile stress on metabolism and apoptosis of chondrocytes inrat

Objective To investigate the effect of cyclic tensile stress (CTS) on the metabolism inand apoptosis of rat chondrocytes.Methods Primary rat chondrocytes were cultured on a Bioflex plate for one day and then stretched cyclically for 24 hours at a frequency of 0.5 Hz using a Flexcell-5000T apparatus.The cells were divided into 5 groups according to their stretching ratio:0% (the control group),2%,6%,10% and 14%.After the stretching,Col Ⅱ,Aggrecan,MMP-13 and ADAMTS-5 mRNA were measured using qPCRs,and the NO and PGE2 levels were measured using ELISA kits.Moreover,TUNEL staining and Annexin V-FITC/PI were used to analyze the apoptosis of chondrocytes.Results Compared with the control group,the average levels of Col Ⅱ and Aggrecan mRNAdecreasedin 10% and 14% groups [(0.738±0.11) and (0.58±0.13),(0.75±0.11) and (0.55±0.09)].In those groups,the MMP-13 [(2.47±0.47) and (2.88±0.36)] and ADAMTS-5 mRNA level [(2.39±0.33) and (2.75±0.49)],the NO [(6.96±0.96) and (8.28±0.82)] and PGE2 level [(6.83±0.66) and (7.15±0.71)] had increased significantly.In the 6% group the average levels of Col Ⅱ(1.76±0.30) and Aggrecan mRNA (1.93±0.14)of 6% group were significantly higher than the control group,but the NO level of the former (3.07±0.20) was significantly lower than the control group (3.89 ± 0.33).The apoptosis rate of chondrocytesin 2% and 6% groups were (0.065±0.013) and (0.063 ± 0.147),without significant differences to that of the control group (0.045 ± 0.008).However,compared with the control group,apoptosis in the 10% and 14% groups [(0.135 ±0.026) and (0.184±0.020)] increased significantly.Conclusion The effect of cyclic tensile stress on chondrocyte metabolism and apoptosis was magnitude-dependent.Ten percent and 14% CTS can increase the catabolism and apoptosis of chondrocytes.Ten percent and 14% strain can increase the catabolism and apoptosis of chondrocytes.Cyclic 6% strain can increase the anabolism of chondrocytes,but 2% strain has no impact on metabolism or apoptosis.
