Objective To make clear the relationship between adenoma,malignant transformation of ade- noma and carcinoma in gallbladder.Methodes The nuclear morphometry was performed with a TJTY-300 Auto- matic Image Analyser and PCNA was measured with immunohistochemisty in 25 cases of carcinoma,20 cases of adenoma and 3 cases of normal gallbladder.Results (1) Cell DNA content,atypia and PCNA index were in- creased gradually from the normal cell to dysplasia to malignant transformation of adenoma to carcinoma in gallblad- der;(2) Aneuploidy in gallbladder adenoma and PCNA index had significant relationship to the size of adenoma and gallstones (P
2.Mutation of ras gene and overexpresion of PCNA as prognostic parameters in gallbladder cancer

Objective To estimative the relationship between ras gene mutation,PCNA overexpression and the recurrence and prognosis of gallbladder cancer.Methods PCNA was studied by ABC immunohistology, ras gene mutation by PCR-RELP analysis.Results Histology,Nevin staging and operative methods were relevant to the prognosis of gallbladder cancer.The patient with stag IV,V gallbladder cancer had a poor prognosis;radical operations could prolong patients survival time.The mortal risk of patients with ras gene mutation was 1.62 times higher than that without ras gene mutation,with high overexpression of PCNA was 2.2 times higher than that with low expression of PCNA. Conclusions Differentiation degree, Nevin′s staging, mutation of ras gene and overexpresion of PCNA could be the prognostic factors in gallbladder cancer,and could help to select the operative procedure and to eluvate the surrival.
3.The influnece and related factors analysis of postpartum community visit for 6 months old infants exclusive breastfeeding

Objective To explore the influnce and related factors analysis postpartum community visit for 6 months old infants exclusive breastfeeding (EBF).Methods 800 women in Daqing city communities delivered in January,2009 to December,2010 were selected into study based on simple random sampling.Using the questionnaire surveyed the influence of the postpartum community visit for 6 months old infants EBF.Results Reclaimed effective questionnaires were 790 ;6 months old infants EBF rate was 65.06% ;the receiving postpartum community visit group's 6 months of EBF rate 7 4.8 7 %) was significantly higher than the not receiving postpartum community visit group (38.50%),difference was statistically significant(x2 =90.54,P < 0.01).Logistic regression analysis showed that main related factors of 6 months old infants EBF were the first breast feeding time,whether or not to accept the postpartum visit,mode of delivery and mother's education degree,respectively.Conclusion Postpartum community visit can promote 6 months old infants for EBF,but still need to pay more attention to coordination and improvement.

Objective To study the antioxidant activity of the extract from north Ph.praecox leaves.Methods The extract from north Ph.Praecox leaves was made by ultrasonic-assisted extraction process,and the concentration of the total flavonoids was detected by NaNO3-Al(NO3)3-NaOH methods.The antioxidant capacity of the extraction was detected by DPPH free radical scavenging method and CuSO4-Phen-Vc-H2O2-DNA reaction with ultra-weak chemiluminescence method.Results The bamboo leaf extract could scavenges the DPPH free radical with the IC50 value as 7.02 mg /L and could delay the development of the emission peak value(P
5.Analysis of Fundamental Frequency and Voice Parameter From 4 to 8 Years Old Children

Objective To study and analyse the parameter values of voice in normal healthy preschool children and school-age children. Methods In present paper, voice quality of 120 cases were collected and analyzed with Dr. Speech software. Results There were no differences between preschool and school-age children on parameters jitter, shimmer, SNR, HNR, NNE, F 0, F 1,F 2,F 3 of voice acoustic and so in difference gender. Conclusion All parameters in preschool children and school-age children showed no difference. The tested parameters were significantly different between normal children and children with vocal nodule.
6.Changes in abilities of invasion and metastasis in surviving human lung adenocarcinoma (A549) cells after X-ray irradiation and related mechanism

Objective To investigate the changes in the abilities of invasion and metastasis in surviving human lung adenocarcinoma (A549) cells after X-ray irradiation and the related mechanism.Methods The change in radiosensitivity after X-ray irradiation was determined by colony formation assay.The abilities of invasion and metastasis were evaluated by MTF adhesion assay and Transwell invasion and migration assay in vitro.The mRNA and protein expression of E-cadherin,vimentin,tumor growth factor (TGF)-β1,matrix metalloproteinase (MMP)-2,and MMP-9 was measured by real-time RT-PCR using SYBR Green fluorescence and Western blot.Results The colony formation assay showed that the surviving A549 cells after X-ray irradiation were more resistant to irradiation (ratio of D0 values =0.94).Their abilities of adhesion,invasion,and migration were significantly increased by 1.46 times,1.40 times,and 1.45 times,respectively.In addition,the surviving cells showed enlarged intercellular spaces and had many long pseudopodi.Compared with those in the control group,the mRNA expression levels of vimentin,TGF-β1,MMP-2,and MMP-9 of surviving cells were increased by 1.37 times,2.37 times,1.80 times,and 1.50 times,respectively,the protein expression levels of the above substances were increased by 1.60 times,1.80 times,1.10 times,and 1.20 times,respectively,and the mRNA and protein expression levels of E-cadherin were decreased by 59.4% and 74.6%.Conclusions The surviving A549 cells after X-ray irradiation have significantly increased abilities of invasion and metastasis.This may be due to radiationinduced TGF-β1 expression increase that in turn promotes epithelial-mesenchymal transition and also due to radiation-induced MMP-2 and MMP-9 expression increase.

Objective:To construct Interleukin 21 (IL 21) expression plasimd pCDNA3.1 IL21 and express it in Hela cell and analysis its acitivity of costimilating T cell proliferation with anti CD3 monoclonal antibody in vitro.Methods:The CDs of IL 21 was amplified by PCR using the pMD IL21 as templet.The expression plasimd pCDNA3.1 IL21 was constructed by inserting the sequence of coding region of the IL 21 into pCDNA3.1/Hismyc B containing CMV promoter and transfected into Hela cells.The stability expression stain was screened by the condition media containing 400 mg/L G418 and cloned through the limited dilution method.The target protein was purified through Ni 2+ chelating Sepharose Fast Flow.The bioactivity was confirmed by MTT method on costimulating the T cell proliferation with anti CD3.SDS PAGE and Western blot analysis the rhIL 21 expressed.Results:hIL 21 was expressed in Hela cell successfully.SDS PAGE analysis showed the IL 21 fusion protein with Mr 18 000 or so was expressed in supernatant of the cells.The rhIL 21 has significant proliferation effect on mature human T cells in the presence of anti CD3 monoantibody.Conclusion:The rhIL 21 with bioactivity has been obtained,which may help researcher study its new function and effects. 8.Effect of Aromatic Herbs for Resuscitation on NO Content and Expression of NOS in Brain Tissue of Rats with Cerebral Ischemia and Reperfusion Injury

[Objective] To investigate the effect of aromatic herbs for resuscitation such as Moschus and Borneol on nitric oxide (NO) content and expression of endothelial NO synthetase (eNOS) and inducible NO synthetase (iNOS) in brain tissue of rats with cerebral ischemia and reperfusion injury. [Methods] Forty rats were allocated to 4 groups: mimic operation group (Group A), model group (Group B), herbs treatment group (Group C) and nimodipine treatment group (Group D). Rat model of cerebral ischemia and reperfusion injury was established by Pulsinelli 4VO method. NO content in brain tissue was determined by nitrate reductase method. Expression of eNOS and iNOS in micrangium and iNOS in neurons was measured by semi-quantitative analysis. [Results] NO content was decreased and approached normal level in Group C and Group D (P 0.05 as compared with Group A). Aromatic herbs with action of inducing resuscitation increased the expression of eNOS in micrangium and decreased the expression of iNOS in micrangium and neurons, the differences being significant as compared with those in Group B ( P

To investigate the effect of treatment based on syndrome differentiation combined with resuscitation inducing aromatic herbs for acute ischemic stroke (AIS).Forty four cases of AIS were randomly allocated to two groups. Twenty cases of Group A were treated with treatment based on syndrome differentiation combined with Tongqiao capsule(each capsule containing Moschus 0 04?g and Borneolum 0 1?g)and twenty four of Group B with treatment based on syndrome differentiation. Therapeutic effect were observed in the second week after medication. The total effective rate was 95% in Group A and 91 6% in Group B, the difference being not significant. Objective To study the characteristics of immune function,Epstein-Barr virus(EBV) antibodies and EBV-DNA in children with different clinical types of EBV infection,which provide basis for prevention and treatment of EBV infection.Methods Clinical data of 103 patients suffering from EBV infection were retrospectively analyzed in Xijing Hospital of the Fourth Military Medical University.A total of 103 children were divided into infectious mononualeosis(IM) group(n=68),chronic active Epstein-Barr virus infection(CAEBV) group(n=13) and Epstein-Barr virus-related hemophagocytic lymphohistiocytosis(EBV-HLH) group(n=22).The changes of EBV antibodies,EBV-DNA,immunoglobulin levels,lymphocyte subpopulation and complement series were detected and compared among the three groups.A total of 26 healthy children at the same stage were enrolled as a control group,immunoglobulin levels,lymphocyte subpopulation and complement series were detected in control group,then compared with the rest of the three groups.Results The levels of C3 and C4 in CAEBV group and EBV-HLH group were significantly decreased than those in the control group(P<0.05).The levels of IgA,IgG and IgE in EBV-HLH group,CAEBV group,IM group and control group gradually increased(P<0.05,respectively).The levels of IgA,IgG and IgE in EBV-HLH group and CAEBV group significantly decreased than those in control group(P<0.05,respectively).The levels of IgA,IgG and IgE in IM group decreased than those in control group,while there were no statistically significant differences(P>0.05).CD8+T cells in IM group significantly increased than those in the rest of the three groups(P<0.05,respectively).T cells,CD8+T cells,CD4+ T cells,CD4+/CD8+ ratios,NK cells,B cells of EBV-HLH group significantly decreased than those in the rest of the three groups(P<0.05).The positive rates of EBV antibodies in CAEBV group and IM group were significantly higher than those in EBV-HLH group(P<0.05).EBV-DNA in EBV-HLH group were significantly higher than those of CAEBV group and IM group(P<0.05).Conclusion EBV-DNA levels in the serum are positively correlated with disease types and severity,the pathogenesis of IM,CAEBV and EBV-HLH induced by EBV infection are associated with immune dysfunction.Dynamic monitoring of EBV load and cell immune function can reflect disease status and progress risk.
