1.Cross-talk between nuclear factor-?B and other signaling pathways

Transcription factor NF-?B has attracted attention due to its important role in the regulation of the expression of a number of cellular genes involved in host defense,inflammation/immune responses,cytokine and growth factor regulation,cell survival,proliferation,and also in embryonic development and programmed cell death. In this review,we provide an overview of established TNF-?,TLR/IL-1R,TCR and BCR signaling pathways to NF-?B,the alternative NF-?B pathway,and the regulation of NF-?B transcriptional activity. Furthermore,we discuss on emphasis the cross-talks between NF-?B and other signaling pathways (TGF-?,p53,nuclear receptor family,Ras/MEK/MAPK...),which are very complex and remain to be understood. The activation or inhibition of NF-?B has effect on other signaling pathways. They compose a complex signaling network,in which many regulatory signals integrate systematically and coordinate transcriptional responses to the stimulations. So when NF-?B is used as an important target of drugs in human diseases,the pathological and physiological functions of related signal pathways also should be understood sufficiently.

3.Expression of bFGF mRNA in pulmonary artery of rat with chronic hypoxia and its effect on tension of rat pulmonary artery in vitro

AIM: To explore the role of basic-fibroblast growth factor (bFGF) in the development of pulmonary hypertension induced by hypoxia. METHODS: 1) The pulmonary arteries of SD rats with hypoxia for one and two weeks were isolated, from which the total RNA were extracted by acid guanidinium thiocyanate-phenol-chlorform .Then the levels of mRNA were measured by RT-PCR. 2) About 3mm-long arterial rings cut from SD rat pulmonary arterial stem were suspended between stainless steelhooks in chamber with warmed (37℃) Kreb's solution. Different concentrations of bFGF were added in a cumulative fashion into the chamber where the rings were suspended. The cumulative concentration response curve was obtained. Objective To analyze the difference of salivary microbiome between the patients with functional dyspepsia and healthy controls.Methods Saliva samples were collected from the patients with functional dyspepsia and healthy control.Genomic DNA of the samples was extracted,and the high-throughput sequencing technology was used to conduct DNA sequence of 16S rRNA-V4 region.Subsequently,all the data were performed by the bioinformatic analysis.Results The salivary microbiome in the functional dyspepsia group was dominated by Proteobacteria,while Bacteroidetes was the top microbiota in the heathy control group.In the functional dyspepsia group,the Chao1 index,ACE index,Shannon index and Simpson index were 1 295,1 351,4.93 and 0.90 respectively.In the healthy control group,the above indexes were 1 001,1 351,5.28 and 0.92 respectively.The PCoA ba sically separated the microbiome composition of the two groups.Sixteen kinds of microbiota were significantly different between two groups using linear discriminant analysis effect size tool,including Bacteroidetes,Prevotella,Prevotellaceae,Neisseria,Betaproteobacteria and Proteobacteria,etc.Conclusion Saliva in the patients with functional dyspesia has characteristic microbiome composition,which is dominated by Proteobacteria,but the richness and diversity between the two groups have no difference.Neisseria is one of the significantly different bacteria between the two groups.
5.A nested case-control study on genetic polymorphisms for patients with non-alcoholic fatty liver disease

Objective To investigate the relationship between the genetic polymorphisms, which played roles in the pathogenesis of metabolic syndrome (MS), and susceptibility of non-alcoholic fatty liver disease (NAFLD) in Han people in Guangdong province. Methods The subjects were selected from an epidemiologie survey in Guangdong province. Fifty to 117 adult NAFLD patients, who met the criteria of Chinese guideline for diagnosis of NAFLD and had typically clinical, biochemical signs and abdominal ultrasonography, were recruited in the study. By using 1 : 1 matched method of nested case-control study, same numbers of people without NAFLD were included as controls. The genetic analyses was performed by using genomic DNA extracted from peripheral blood. Polymerase chain reaction (PCR)-restriction fragment length polymorphism (RFLP) was applied to detect the single nucleotide polymorphisms (SNPs) at 9 sites in 7 candidate genes. Results Most SNPs of the genes were related to the susceptibility of NAFLD. Some of them had positive relation (increasing the risk) such as tumor necrosis factor (TNF)-α-238, adiponectin-45, leptin-2548, peroxisome proliferator-activated receptors (PPAR) γ-161 and phosphatidylethanolamine N-methyltransferase (PEMT)-175. Some had negative relation (decreasing the risk) including adiponectin-276 and hepatic lipase-514. And some had no relation (TNF-α-380 and PPAR g coactivator-1α-482). Conclusion Most cytokines' SNPs of candidate genes discovered in MS patients are related to the susceptibility of NAFLD.
6.Application of parameter assignment method in the review of parenteral nutrition prescription in our hospital

OBJECTIVE To provide reference for improving the working efficiency of prescription review for pharmacists and ensuring the safe use of parenteral nutrition solution in patients. METHODS The review module of parenteral nutrition prescription was developed with the pharmacy intravenous admixture services(PIVAS) MATE system. According to the instructions and relevant information of parenteral nutrition solution, the parameters to the review module for parenteral nutrition solution prescriptions were assigned, thresholds and calculation formulas were set, and the prescription was automatically determined by the system. The type and quantity of the inappropriate prescriptions of parenteral nutrition solution in our hospital from January to August 2020 and from January to August 2022 were counted, and the review efficiency of inappropriate prescriptions was compared before and after the application of the review module of parenteral nutrition solution prescription. RESULTS The module realized the informatized review of basic attribute parameters and custom parameters of parenteral nutrition solution prescription, such as ion concentration, ratio of heat to nitrogen, ratio of sugar to lipid, glucose concentration, ratio of sugar to insulin, maximum dosage of compound preparation, necessary drug collocation, incompatibility, repeated drug use, age limit, etc. The module also shortened the review time greatly. It took about 33 minutes for two pharmacists to review 100 prescriptions, and only took 6 seconds after the module went online. Compared with January to August in 2020, the average approval rate of inappropriate prescriptions for parenteral nutrition solution increased from 0.582% to 1.416% during the corresponding period in 2022 (P<0.05). CONCLUSIONS The review module of parenteral nutrition solution prescription established by parameter assignment method in our hospital can greatly reduce the phenomenon of missed and wrong review by pharmacists, reduce the risk of parenteral nutrition solution dispensing, reduces the work pressure of pharmacists reviewing prescriptions, and ensures the drug safety of patients.
