1.Expression of PcG related protein RYBP in pancreatic carcinoma and its clinical significance

Purpose To investigate the expression and clinical significance of RINGI and YYI binding protein (RYBP)in pancreatic carcinoma (PC).Methods RYBP expression was detected in 51 cases of PC tissues and paired adjacent nontumor tissues by immunohistochemistry.The correlation between RYBP expression and the survival time of PC patients after surgery was analyzed by Kaplan-Meier method.Results Compared to the expression in adjacent non-tumor tissues,RYBP was significantly lower expressed in PC tissues (P < 0.05).In addition,RYBP expression had a significant correlation with tumor diameter and tumor staging of patients (P < 0.05),but no significant correlation with other characteristics of patients,such as gender,age,smoking,alcohol intake,tumor number,metastasis or CA199 level (P > 0.05).The survival analysis showed that the survival time of PC patients with positive expression of RYBP was significantly longer than patients with negative expression of RYBP (P < 0.05).Conclusion RYBP is significantly lower expressed in PC tissues,which might be involved in the pathogenesis of PC.
2.Investigation of psychological health in middle-school students of Zhuang nationality

BACKGROUND: The middle-school stage is the key period of personality figuring and taking shape.The mentation in this period is filled with a lot of special psychological contradictions and conflicts. Long-term psychological worry will affect the formation of perfect personality.OBJECTIVE: To understand the psychological health status in the middle-school students of Zhuang nationality and the main factors of influence.DESIGN: Randomized cluster sampling and questionnaire investigation based on middle-school students.SETTING: Department of Preventive Medicine, Youjiang Medical College of Nationalities.PARTICIPANTS: 2 000 middle-school students of 34 classes from 3 middle schools in Baise City and Longlin County were selected to attend the investigation with randomized cluster sampling method in November 2003.METHODS: The psychological health statuses of middle-school students were evaluated with symptom checklist (SCL).The list contained 90 items,including 9 factors of psychological symptom such as somatization,compulsion,sensitivity to human relations,anxiety,depression,hostility,dread,paranoia and psychosis.The grading system from 1 to 5 grades was used and the standard of the adult norm of the whole country was refered to:Factor score<1.8 was normal;Factor score from 1.8 to 2.0 was mild abnormality;Factor score from 2.1 to 3.0 was moderate abnormality;Factor score>3.0 was severe abnormality. Instructive words were unified.The investigative method and behavior were normalized.The questionnaires were filled in without recording the names and taken back from the spot and then compared with the norm of the whole country.MAIN OUTCOME MEASURES: The evaluated result of psychological health in middle-school students.RESULTS: 2 000 questionnaires were provided and 1 855 were valid after return with the effective rate 92.75%. ①The average scores of each factor of SCL were all higher in the middle-school students of Zhuang nationality than in the norm of the whole country. The significance of difference existed(P=0.000).②The scores of somatization,depression,anxiety,dread and psychosis were all higher in the schoolgirls than in the schoolboys.The significance of difference existed (P=0.000).③Except hostility factor,the scores of other factors were higher in the senior middle-school students than in the junior middle-school students.The difference was statistically significant(P=0.000).④The arranged sequences(from bigness to smallness) of each factor that affected psychological health of middle-school students of Zhuang nationality in Baise were depression,somatization,compulsion,anxiety,sensitivity to human relations,psychosis,paranoia,hostility and dread.The sensitivity to human relations ranked 4th and the anxiety ranked 6th in the schoolboys,but the orders of the 2 factors were just the opposite in the schoolgirls and the orders of other factors were the same in both the schoolboys and the schoolgirls.The first 3 factors all were:depression,somatization, compulsion.CONCLUNSION: The psychological health status was lower in the middle-school students of Zhuang nationality than in the norm of the whole country and the first 3 factors that affected their psychological health were:depression,somatization and compulsion. The levels of psychological health on somatization,depression,anxiety,dread and psychosis were lower in the schoolgirls than in the schoolboys. The psychological problems were more serious in the senior middle-school students than in the junior middleschool students. The levels of psychological health in middle-school students of Zhuang nationality were relatively lower and the psychological education and intervention study should be provided aiming at psychological characteristics of different population.
3.Protective Effects of Folic Acid on Cardiac Development Related Genes GATA-4 and NKx2.5 Expression of Offspring Rats with Maternal Coxsackievirus B3 Infection

Objective To investigate the effects of oral folic acid on cardiac development related gene expression of offspring in an experimental model of coxsackievirus B3(CVB3) infection of pregnant rats. Methods SD female rats were randomized into control group, folic acid group, CVB3 group and CVB3+folic acid group. The female rats were given folic acid by gavage for 2 weeks before pregnancy in folic acid group and CVB3+folic acid group. After conception for 7 days, rat model was established by intraperitoneal injection of CVB3 for 5 days in CVB3 group and CVB3+folic acid group. After nat-ural childbirth neonatal heart was taken and stored in liquid nitrogen. The morphological changes of neonatal rat myocardial tissues were observed by HE staining. The expressions of GATA-4 and NKx2.5 mRNA were detected by RT-PCR and West-ern blot assay. Results There was significant myocardial injury, such as myocardial fiber disarray and myocardial fiber breakage, in neonatal rats in CVB3 group. These damages were improved in CVB3+folic acid group. The expression levels of GATA-4 and NKx2.5 genes in myocardial tissues were significantly lower in CVB3 group than those of control group (P<0.05). The expression levels of GATA-4 and NKx2.5 proteins in myocardial tissues were significantly higher in CVB3+folic acid group than those of CVB3 group (P<0.05). Conclusion CVB3 infection in the early pregnancy inhibited the expres-sion of neonatal rat cardiac development factor. Folic acid supplementation has obvious protective effects on the neonatal rat cardiac development.
4.Expression of FOXC1 and its relationship with E-cadherin in nasopharyngeal carcinoma tissues.

OBJECTIVE:
To investigate the significance and relationship between the expression of FOXC1 and clinicopathological features, and to explore its correlation with E-cadherin.
METHOD:
Immunohistochemical SP method was used to detected the expression of FOXC1 in nasopharyngeal carcinoma tissues and nasopharyngitis tissues.
RESULT:
(1) Immunoreaction to FOXC1 was mainly located in nucleus of nasopharyngeal carcinoma cells. The positive expression rate of FOXC1 in nasopharyngeal carcinoma tissues was 85.3% (81/95), which was significantly higher than that in nasopharyngitis tissues (59.4%) (P < 0.05). (2) The expression of FOXC1 was not related to patients' age and gender, clinical stage of cancer and lymph node metastasis (P > 0.05). (3) There was a correlation between the expression of FOXC1 and down-regulated expression of E-cadherin in nasopharyngeal carcinoma tissues (P < 0.05).
CONCLUSION
FOXC1 may play an important role in generation and progression of nasopharyngeal carcinoma, there may be a correlation between the expression of FOXC1 and down-regulated expression of E-cadherin, also FOXC1 may play an important role in the process of EMT in nasopharyngeal carcinoma by regulating E-cadherin.

5.Relationship between hepatocellular carcinoma and the interaction between hMSH2 polymorphisms and environmental factors.

OBJECTIVETo use a hospital-based case-control study design to investigate the relationship between hepatocellular carcinoma (HCC) and the interaction of polymorphisms in the human mismatch repair gene,hMSH2,with environmental factors.
METHODSCases of new-onset,histopathology-diagnosed,and untreated (no chemotherapy or radiation therapy) HCC were enrolled between September 2009 and September 2012.A non-HCC healthy control group was also enrolled and was composed of individuals living in the same region as the cases for more than 10 years and age-/sex-matched with similar socioeconomic characteristics.All enrollees underwent hMSH2 genotyping by real-time PCR.T-test,chi-square test and unconditional logistic regression analysis was used to analyze the difference in allele frequencies among the case and control groups and the relationship between hMSH2 polymorphisms and environmental factors.
RESULTSFrequencies of hMSH2 rs2303428 CC,CT and TT genotypes in the HCC group were significantly different than in the control group (14.13% vs.8.21%,47.02% vs.49.47%,and 38.85% vs.42.32%;x 2=8.289,P =0.016).Individuals carrying the hMSH2 rs2303428 T allelic gene had a significantly increased risk compared to those with the hMSH2 rs2303428 C allelic gene (adjusted OR=1.228).Interactions were found between the hMSH2 genotype and hepatitis B surface antigen (HBsAg)-positive hepatitis infection (adjusted OR=1.865) and history of cancer (adjusted OR=5.634).There was no relation between hMSH2 gene rs4952887 and rs2059520 and liver cancer development or interaction with environmental factors.
CONCLUSIONThe hMSH2 rs2303428 genotype is positively related to risk of HCC in Chinese,with HBsAg-positive hepatitis infection starus and history of cancer increasing the risk.
