3. Effects of PEG-6000 stress on tanshinones accumulation in hairy roots of Salvia miltiorrhiza
Chinese Traditional and Herbal Drugs 2013;44(9):1181-1185
Objective: To study the effects of polyethylene glycol-6000 (PEG-6000) stress on the accumulation of tanshinones in hairy roots of Salvia miltiorrhiza. Methods: Agrobacterium rhizogenes ATCC15834 was used to induce the hairy roots of S. miltiorrhiza. After 20 d suspension culture, the PEG-6000 (1.2%, 2.0%, 5.5%, and 10%, respectively) was added into the suspension cultures and at the same time, the contents of tanshinones (including tanshinone I, cryptotanshinone, dihydrotanshinon I, and tanshinone IIA) were quantified by HPLC on day 7. Results: The growth of the hairy roots of S. miltiorrhiza was inhibited by PEG-6000. After PEG-6000 (1.2%, 2.0%, 5.5%, and 10%) treatment, the dry weights of the hairy roots of S. miltiorrhiza were reduced to 75.1%, 83.0%, 76.2%, and 76.1% of the control group, respectively. Addition of PEG-6000 at different levels could significantly increase the yields of four tanshinones in the hairy roots of S. miltiorrhiza. The yields of tanshinone I, cryptotanshinone, dihydrotanshinon I, and tanshinone IIA were significantly increased by 2.0%-5.5%, 1.2%, 2.0%, and 5.5% PEG-6000, respectively. And the tanshinone IIA increased most. Conclusion: PEG-6000 could stimulate the accumulation of tanshinones in the hairy roots of S. miltiorrhiza.

Microbiology 1992;0(05):-
The fresh conidia powder of Beauveria bassiana SGBB8702 produced with diphasic technology was dried using 36-h procedures of vacuum-freeze drying (VFD) or vacuum drying (VD). The VFD and VD procedures reduced water content of the fresh conidia powder from 58.56% to 3.97% and 4.26%, resulting in preparations containing 1.29, and 1.25?10 11 conidia/g. The VFD or VD conidia had the same viability (≥98%) as the fresh ones but germinated slightly more slowly than the fresh ones. The estimates of LC 50 s for the fresh, VFD, and VD conidia against Myzus persicae on day 7 after inoculation were 1.15, 5.89, and 2.95?10 4 conidia/ml, respectively. At the concentration of 10 6 conidia/ml, the LT 50 of the fresh conidia against M. persicae was estimated as 3.6 d, corresponding to 3.9 d and 4.4 d for the VFD and VD conidia, respectively. Due to much lower cost, the VD procedure was of greater potential for drying B. bassiana conidia in mass production though the VFD procedures resulted in slightly better quality of conidia powder. The viability and virulence of the VFD conidia were assessed periodically during 12-mon storage at 4℃ and 20℃, respectively. No viable conidia stored at 20℃ were detected 255 d after storage whereas those stored at 4℃ had a viability of 90.15% and an LT 50 of 4.7 d at the end of 12-mon storage. The results showed that storage of B. bassiana conidia powder at ambient temperature was unable to maintain shelf life at commercially acceptable level even though its water content was reduced to

Chinese Journal of Emergency Medicine 2006;0(06):-
Objective To investigate the effect of TanshinoneⅡA (TSN) on the cell hypertrophy induced by angiotensinⅡ(AngⅡ) in the primary culture of neonatal rat cardiomyocytes. Method The effect of TSN on cardiomyocyte was evaluated by the 3-[4, 5-dimethylthiazol-2-yl] -3, 5-diphenylformazan (MTF) assay. As the index of eardiomyocyte hypertrophy, protein synthesis rate was measured by H-Leucine incorporation and the cell size was determined by phase contrast microscope. The proto-oncogene c-los mRNA and c-jun mRNA expression was assessed using reverse transcription polymerase chain reaction (RT-PCR). Results Exposure of the myocytes to TSN (5~80 mmol/L) for 24hours produced no cytotoxicity. Protein synthesis rate and proto-oneogene c-fos and c-Jun mRNA expression of eardinmyoeytes increased significantly after AngⅡtreatment, and TSN inhibited these effect of AngⅡ.Conclusions TSN can prevent the hypertrophy of eardiomyocytes induced by AngⅡ, which be attributable relate to the decreased expression of proto-oncogene c-los and c-jun mRNA by TSN.
