1.Endoscopic diagnosis and treatment of biliary complications after liver transplantation: Clinical analysis of 250 times of procedures

Objective To evaluate the clinical value of endoscopic retrograde cholangiopancreatography(ERCP) in the diagnosis and treatment of biliary complications after orthotroptic liver transplangtation.Methods We retrospectively reviewed the clinical data of 139 patients with biliary complications after orthotropic liver transplangtation,and summarized 10 categories of biliary complications.The location of biliary complications included donor liver,anastomotic stoma,receptor biliary duct,duodenal papilla,and so on.Common complications were biliary duct or anastomotic strictures(102/139,73.4%),cholangitis(94/139,67.6%),common bile duct or intrahepatic cholelithiasis(75/139,54.0%),dysfunction of the duodenal papilla(20/139,14.4%),bile leakage(17/139,12.2%),and complications related to T-tube(4/139,2.9%).Infrequent complications involved duodenal papillary stenosis(1/139,0.7%),complications related to PTC(2/139,1.4%),biliary bleeding(3/139,2.2%),and too long and twisted bile duct(4/139,2.9%).The treatment methods included EST(112/139,80.6%),dilatable bougie or balloon dilatation(157/250,62.8%),plastic stent placement(139/250,55.6%),ENBD and irrigation(87/250,34.8%),and gallstone removal(27/250,10.8%).Results A total of 250 times of ERCP was performed in the 139 patients,the success rate of intubation being 99.2%(248/250).The total cure rate reached 70.5%(98/139) and the improvement rate was 18.7%(26/139),with a total effective rate of 89.2%(124/139).There were 9 patients with conversions to open surgery(9/139,6.5%),4 patients with no improvement(4/139,2.9%),and 2 patients with spontaneous quitting treatment or loss of follow-up.The postoperative complications included cholangitis in 37 patients,the dislodgement of ENBD tube in 26 patients,and biliary sludge or stone in 18 patients.Conclusions ERCP is an effective way to diagnose and treat the biliary complications after orthotroptic liver transplantation.
2.Survey of self-efficacy of condom use among mobile female population and its related factors

Objective To investigate self efficacy of condom use among mobile female population and to analyze related factors so as to provide evidence for relevant interventions.Methods One hundred and eighteen women working as waitresses in hotels in Beijing were interviewed with questionnaires.Results The average score of self-efficacy of condom use among the studied women was 45.42?8.26,and the self-efficay of condom use among them varied with their age,marital status and incomes;their attitude towards condom use and knowledge about correct condom use had obvious effect on the score of self-efficacy.Conclusion Mobile female population have a middle level of condom use self-efficacy,and there is a need to improve their condom use efficacy through training,especially for those unmarried young women with lower incomes.
3.THE ORIGIN OF NORADRENERGIC FIBERS IN THE SEPTUM OF THE RAT——A STUDY OF COMBINED RETROGRADE FLUORESCENT LABELING AND HISTOFLUORESCENCE

21 albino rats were used. The origin of the noradrenergic (NA) fibers in the septum was studied with a simultaneously combined retrograde fluorescent tracing and histofluorescence method. The results revealed that the NA neurons projecting to the septum were in A_1, A_2 and A_6 groups. The projections of A_1 were bilateral, But A_2 and A_6 projected dominantly to the ipsilateral septum, with a few to the contralateral. Retrograde labeled NA cells were localized in the rostrodorsal part of A_6 group, and scattered over the whole rostrocaudal length of A_(1,2) groups. No labeling was seen in A_4 and A_7 groups. In five of the animals, retrograde labeled NA cells were occasionally observed in A_5 group. In addition, some scattered labeled non-NA neurons were seen in the raphe and the ventrolateral reticular formation of the rostral medulla.
4.Application of low power laser irradiation and olfactory ensheathing cell transplantation in treatment of spinal cord injury

Spinal cord injury has been considered the disease seriously suffering people.Their oligodendrocyte cells cannot form the channel guide to axons' migrating and secrete inhibitors,as well as astrocytes format glial scar in the injured district after rapidly reactive proliferation and inhibit axon growth.Thus,the treatment of spinal cord injury is still a thorny issue in medical field at present.Olfactory ensheathing cells have a good specific process axis linking of axon regeneration and accurately target.The effects of low power laser irradiation on the nervous system and olfactory ensheathing cells not only included its reservations and even promotion of the activity of damaged neurons,decrease in scar formation,prevention of the degeneration of neurons,but also increase in olfactory ensheathing cell activity by intervening some factors.Olfactory ensheathing cell transplantation and low power laser irradiation play an important role in the repair of spinal cord injury.
5.Smooth muscle progenitor cells and cardiovascular disease

It is possible for bone marrow cells,peripheral blood cells,myocardial cells,muscle cells and embryonic stem cells differentiating into smooth muscle progenitor cells(SPCs),which can express special marks of myosin heavy chain.SPCs remained quiescent,until mobilized in response to injury or disease.With various growth factors,cytokines,SPCs are mobilized following physiological stress and subsequently home to site of vascular damage,where they contribute to the remodeling process.Recently,various studies have found that SPCs from different sources contributed to the remodeling process of vessels and development in atherosclerosis.It is generally accepted that SPCs contribute to pathological changes,leads to restenosis.But several recent papers reported findings that injection of SPCs increased collagen and SMC content and reduced macrophage content,consistent with a more stable plaque.SPCs may secrete cytokines and growth factors both locally and systemically that enhance plaque stability.It remains to be determined whether smooth progenitor cells,which are less well studied than their endothelial counterparts,can likewise be manipulated to achieve therapeutic genes benefit.This review has elucidated the origin and distribution of SPCs,mobilization and recruitment,and their roles in cardiac vascular diseases.
6.The role of glutamine and arginine in rat hepatic fibrosis

Objective To study the effect of glutamine and arginine in rats with hepatic fibrosis.Methods Thirty male Wistar rats with hepatic fibrosis induced by CCl4 for 6 weeks were randomly divided into control group(n=10)、glutamine treatment group(n=10) and arginine treatment group(n=10).At 8 weeks after treatment,the liver appearance、collagen protein level and the degree of hepatic fibrosis were observed.Results At the end of 8 weeks,there was no death of any group of the rats.The glutamine group showed the most severe hepatic fibrosis,the control group showed less,and the arginine group showed the least fibrosis.Conclusions The degree of hepatic fibrosis is aggravated by glutamine,while arginine obviously relieves rat hepatic fibrosis.
7.Establishment of a cisplatin-resistant mouse model of 4T1 triple negative breast cancer

Objective To establish a cisplatin-resistant 4T1 mouse model of triple negative breast cancer .Meth-ods A drug resistant mice model was established with cisplatin ( DDP ) induction and in-vivo/in-vitro tumorigenic ap-proach.Its resistance characteristics were identified by MTT assay .Changes of drug resistance gene ( MDR1, BCRP, MMP7, GST-π) and protein ( P-gp, BCRP, MMP7) expression, and phosphorate-Akt and total-Akt protein expression were evaluated by real-time PCR, immunohistochemistry and western blot method , respectively.Small animal live imaging technology was applied to detect tumor growth .Results Resistance fold (RF) of cisplatin-resistant 4T1 mouse model was 12.84.The expression of MDR1, BCRP, MMP 7, GST-πmRNA and P-gp, BCRP, MMP 7 proteins in the resistant mice were higher than that in the non-resistant mice .The result of western blot showed that a statistically higher expression of p-Akt in resistant mice than that in non-resistant mice at protein levels (P<0.01).No significant difference of tumor growth rate was observed between non-resistant and resistant mice ( P>0.05 ) .Given same dose of DDP , resistant mice showed lower sensitivity than non-resistant mice significantly (P<0.01).Conclusions We have successfully established a cis-platin-resistant triple negative breast cancer model in mice , which provides a new platform for further study on chemoresis-tant reversal strategy and individualized clinical treatment of this disease .

9.Surgical treatment of hallux valgus

Hallux valgus is a frequent forefoot disease, which has the high incidence, more happened among females, hallux valgus becomes more serious along with the age increasing, it usually needed to be done with surgery treatment owing to deformity and pain. The surgery with regard to hallux valgus had emerged widespread at the end of the 18~(th) century, most of the operation styles had been eliminated due to their disadvantages along with the carried out and the follow up judgment of the operation styles , however, some operation styles had been certified effective, which have been modified and consummated endlessly, have been added in the contemporary therapeutic methods. Operation styles mostly consist of soft issue repairment, osteotomy, arthroplasty and arthrodesis , selecting the operation styles reasonably is the key point of guarantee the treatment effectiveness.
10.The role of TRPM7 ion channel in the proliferation and migration of oral squamous cancer cells

Objective:To investigate the role of the transient receptor potential melastatin subfamily member 7 (TRPM7) ion channel in the proliferation and migration of oral squamous cancer cells OC2.Methods:The expression of TRPM7 in OC2 cells was examined by western blotting,RT-PCR and indirect immunofluorescence assay.The influence of TRPM7 on cell proliferation and migration under the inhibition of TRPM7 ion channel with 2-APB or siRNA TRPM7 in OC2 cells was examined by MTT and Transwell respectively.The influence of PI3K/AKT signal pathway on the expression of TRPM7 in OC2 cells was tested by Western blotting.The change of positive ion current when the OC2 cells were treated with TRPM7 inhibitor and activator was explored with patch clamp technique.Results:TRPM7 overexpression in OC2 cells mainly in the cytoplasm was observed,siRNA TRPM7 downregulated the expression of TRPM7.Inhibition of TRPM7 (2-APB or siRNA TRPM7) inhibited the growth and proliferation of OC2 cells in a time-and dose-dependent manner.Blockage of TRPM7 ion channel inhibited the migration capacity of OC2 cells.Inhibition of TRPM7 downregulated the expression of phosphorylated AKT and phosphorylated ERK.Patch clamp assay showed the activation of TRPM7-like current in OC2 cells.2-APB could weaken this current while Bradykinin could enhance the current.Conclusion:TRPM7 may regulate the proliferation and migration of oral cancer cells by the regulation of PI3K/AKT and MAPK/ERK signal pathways and cell positive current.
