1.The probe of circulating immune complexes in thyroid diseases

Serum thyroglobulin(TG),anti-TG antibodies,circulating immune complexes(CIC)and circulating thyroglobulin-antithyroglobulin immune complexes(TG-IC)were studied using ??enzyme-linked immunosorbent assay(ELISA)in 147 sera of patients with thyroid diseases.The serum TG concentration,anti TG and IgG type CIC titer,and TG-IC were significantly increased in patients with Graves'diseases with clinical symptom phase and Hashimoto's thyroiditis,in patients with thyroid goiter,howerever,were sightly increased.The results found that CIC was detectable in 90% sera with TG-IC which contained high TG concentration and low or nagetive anti-TG antibodies.We belivedthat a part of CIC in thyroid autoimmune diseases were composed of TG-IC.TG-ICmight fix complement three(and thus bind to anti/C3 Serum)and play a pathogenic rolein thyroid autoimmune diseases.

3.ESTABLISHMENT OF COXSACKIE B_5 VIRUS INFECTED RAT HEART CELLS IN CULTURES

A micro-model of Coxsackie B5 virus infected rat beating heart cells was introduced.In the infected group, the cardiac enzymes-lactic acid dehydrogenase (LDH) and glutamic oxaloacetic transaminase(SGOT)werc significantly increased and the synthesis rate of DNA was decreased 48 h after inoculation with 400 TCID50 Coxsakie B6 virus.Virus titer(Lg 6.95 TCID50)in the supernatant of the infected cultures was detected 96 h after virus challenge.In addition, the specific fluorescence in cytoplasma of infected cells could be observed.These results suggest that Coxsackie B5 virus might be replicated, assembled and released in cultured rat myocardial cells.
4.Diagnosis and treatment standard of urinary tract infections in children

Urinary tract infection (UTI) are one of the most common infectious diseases in childhood, possibly with atypical symptoms.It is often diagnosed by urinary sediment analysis and urine bacteriological examination.Sometimes it is necessary to find out whether there is urinary tract malformation by imaging evaluation.Once a UTI is confirmed, sensitive antimicrobial drugs should be used based on the general status of the patient, incidence of drug resis-tance in the region where the patient is located, past medication history, and drug sensitivity of urine bacteriology.In addition, treatment of UTI children with vesicoureteral reflux using antimicrobial prophylaxis is still controversial.In this review, the diagnosis, laboratory examination and treatment for UTI children were summarized by referring to and analyzing guidelines, consensuses and clinical evidence on UTI in children, so as to provide reference for clinicians.
5.Establishment of relative quantification in real time reverse transcription polymerase chain reaction to measure cytokine expression

Objective To establish relative quantification in real time reverse transcription polymerase chain reaction to measure cytokine expression.Methods TNF? and GAPDH were used as target gene and internal reference respectively. Two gene fragment were cloned, vectors were purified and quantificated. Standard curves were established using a series dilution of quantificated plasmids to measure the amplification efficiency of TNF? and GAPDH. By changing reaction conditions, the amplification efficiency of two gene were nearly 100%. Ct value of TNF? and GAPDH were measured in 14 samples stimuteneously.Results The method can detected as low as 10 3 copies with the linear range was 10 3~10 9 copies, the intra assay and interassay variation was 1% and 8% respectively. TNF? increased 8 6~9 8 fold with the average 9 2 relative to untreated control group analyzed by the 2 -??Ct methods. Conclusions The method we established has fine sensitivity and reproducibility and the data analysis was simple and reliable and can be apply to any genes.
6.Progress in genetics of vesicoureteral reflux

Vesicoureteral reflux,a most common congenital anomaly of the kidney and urinary tract,is associated with the malformation of ureterovesical junction. It does not cause any specific symptoms or signs un-less it is part of a syndrome or complicated by urinary tract infection. The exact cause is not clear,and genes or environmental factors may result in vesicoureteral reflux. The prevalence of siblings and offspring of reflux pa-tients are higher than normal control groups,so the genetic screening is necessary. This article will review the ge-netics of vesicoureteral reflux and possible interactions.
7.Effect of crocetin on ATPase and hydroxyproline in myocardial hypertrophy rat induced by overload pressure

Object To study the influence of crocetin on the activity of ATPase and the content of hydroxyproline in the myocardial hypertrophy rats induced by overload pressure. Methods Abdominal aortic ligation was used to establish the myocardial hypertrophy model in rats. The rats were divided into two groups: 1) The early term group which included five groups: sham-operation group, model group, Captopril groups, crocetin 100, 50 mg/kg groups. 2) The long term group which also included five groups as the same as the early term groups. Heart indexes were examined. The ATPase activity of heart and the content of hydroxyproline in heart were assayed by colorimetric analysis. Results Compared with the model group, crocetin can significantly reduce the cardiac indexes, increase the activities of Na +, K +-ATPase; Ca 2+ , Mg 2+ -ATPase and markedly decrease the content of hydroxyproline in heart. Conclusion Crocetin could prevent the remodeling of cardiac hypertrophy induced by overload pressure. The ATPase may play an important role in myocardial hypertrophy induced by overload pressure.
8.Protection of crocetin on primary culture cardiac myocyte injured by hydroxyl free radicals

Object To study the cardio-protective effect of crocetin on primary culture cardiac myocyte injured by hydroxyl free radicals. Methods After adding crocetin, primary culture cardiac myocyte was injured by 0.5 mmol/L hydroxyl free radicals which were generated by Fenton systems. The activity of lactic dehydrogenase (LDH) and mitochondrion succinic dehydrogenase (MSD) was observed. Mitochondrion membrane potential (MMP) was detected by Rh123. The ratio of cardiac myocyte apoptosis was investigated by flow cytometry and DNA ladder electrophoresis. Results Compared with the model group, crocetin significantly decreased the activity of LDH in culture supernatant and the ratio of cardiac myocyte apoptosis, markedly increased the activity of MSD and MMP. Conclusion Crocetin has the protective effect on the apoptosis of cardiac myocyte injured by hydroxyl free radical.

Objective:To study adenovirus mediated transfer of TGF ?1 in treatment of mice autoimmune myocarditis. Methods: Experimental autoimmune myocarditis was induced in BALB/c mice using purified myosin from porcine heart. TGF ?1 was transferred into mice by intravenous injection of adenovirus encoding TGF ?1. Plasma level of myosin autoantibody was detected by indirect ELISA method and plasma level of troponin I was assayed using fluorescence immune assay method. The protective effects of TGF ?1 were evaluated from 3 aspects: (1) the degree of infiltrative inflammatory cells in the heart; (2) the plasma level of myosin autoantibody; (3) the plasma level of troponin I.
