1.Antitumor Activity of Dichloromethane Extract from Salvia plebeia and Induction of Apoptosis on K562 Cells

Objective To study the antitumor activity of extract from Salvia plebeia and investigate whether the extract induce apoptosis of K562 cells. Methods The aqueous, petroleum ether, dichloromethane (CH2Cl2), ethyl acetate, and butanol extracts were prepared from the aerial parts of 5. plebeia. Taking fluorouracil as reference, the cytotoxic activities of these extracts on HeLa, A549, SGC-7901, HCT-116, K562, LoVo, DU-145, and HepG2 cells were evaluated. To clarify the apoptosis of K562 cells induced by CH2Cl2 extract, the methods of Hoechst 33258 staining, flow cytometry assay, and DNA ladder assay were investigated. Results The CH2Cl2 extract showed the most potent cytotoxic effect against K562 cells, with an IC50 < 15 μg/mL for 3 d treatment. The characteristic apoptotic symptoms such as DNA fragmentation and chromatin condensation were also observed in the K562 cells. Conclusion The CH2Cl2 extract from S. plebeia may inhibit the cancer cell proliferation by inducing cell apoptosis.
2.Establishment of health literacy evaluation index system for Alzheimer's disease

Objective To establish an objective health literacy evaluation index system of dementia care for scientific assessment.Methods Literature reviews,in-depth interviews,participatory rapid appraisal,and focus group discussion were adopted to developed a scale and checked its items.Then the scale was utilized to investigate 280 residents,and its reliability and validity were examined.Results The health literacy evaluation index system of dementia care included 56 items which belong to four dimensions of knowledge,attitude,behavior and skills.The total Cronbach α was 0.818,split-half reliability was 0.881,the correlation between the total score and each dimension were significant (r =0.417-0.747).KMO statistic in factor analysis was 0.739 and 8 factors predicted 46.305% of total variance.What's more,the correlation between each items score and dimension were significant (r =0.221-0.717).Conclusions The health literacy scale presents acceptable reliability and validity in the surveyed sample,which can be used as a valid measure of health literacy in community health service centers and as the basis of health education.
3.Efficacy of nystatin vaginal effervescent tablets and nifuratel tablet combined with recombinant human interferon alpha 2b suppository in the treatment of candida vaginitis

Objective To explore the efficacy of nystatin vaginal effervescent tablets , nifuratel tablet combined with recombinant human interferon alpha 2b suppository in the treatment of candida vaginitis .Methods 124 patients with candida vaginitis were selected as the research subjects .The patients were randomly divided into two groups according to different treatment .The control group was treated with nystatin vaginal effervescent tablets +nifuratel tablet,the study group was given recombinant human interferon alpha 2b suppository on the basis of the control group.The symptoms,clinical curative effect,quality of life score,adverse reaction rate were observed and compared in the two groups .Results There were no statistically significant differences between the two groups in symptom and physical signs before treatment (all P>0.05).After treatment,the vulvar itching,burning pain,abnormal leucorrhea,redness symptoms and signs of the study group were (0.95 ±0.44)points,(1.02 ±0.51)points,(1.13 ± 0.62) points,(1.09 ±0.56) points,which were significantly lower than those of the control group [(1.42 ± 0.98)points,(1.28 ±1.02)points,(1.38 ±1.17)points,(1.37 ±1.12)points,t=2.529,2.512,2.505,2.618,all P<0.05].The total effective rate of the study group was 95.2%,which was significantly higher than 74.2% of the control group (Z=-2.839,P<0.05).After treatment,the quality of life scores of physiological function ,bodily pain,mental health,emotional function and social function in the study group were (80.16 ±9.23)points,(84.22 ± 9.95)points,(87.49 ±10.46) points,(89.27 ±12.35) points,(88.21 ±11.74) points,which were significantly higher than those in the control group[(67.58 ±5.34)points,(71.49 ±7.28)points,(73.52 ±7.61)points,(76.83 ±8.32)points,(75.51 ±8.16) points,t=9.728,8.577,9.036,7.039,7.537,all P<0.05].The incidence rate of adverse reactions of the study group was significantly lower than that of the control group (χ2 =10.978,P<0.05). Conclusion Nystatin vaginal effervescent tablets ,nifuratel tablet combined with recombinant human interferon alpha 2b suppository in the treatment of candida vaginitis can significantly improve clinical symptoms ,improve the therapeutic effect,and improve the quality of life of patients with significant clinical value ,it is worthy of reference .
4.Clinical effect of ethinyl estradiol cyproterone tablets combined with ovulation induction in the treatment of polycystic ovary syndrome infertility

Objective To explore the clinical effect of ethinyl estradiol cyproterone tablets ( Diane -35 ) combined with ovulation induction in the treatment of polycystic ovary syndrome infertility .Methods 70 patients with polycystic ovary syndrome were enrolled , and they were randomly divided into two groups according to the random number table of the computer ,35 cases in each group .The control group was treated with Diane -35,the observation group was given ovulation induction .The clinical parameters ,vaginal ultrasound results ,cycle ovulation rate ,pregnancy rate,adverse reaction rate and quality of life were compared between the two groups .Results There were no statistically significant differences in uterine volume , intima thickness , bilateral ovarian volume and bilateral follicle number between the two groups after treatment (t=0.055,0.308,0.991,0.435,0.473,0.248,all P>0.05).The cycle ovulation rate of the observation group was significantly higher than that of the control group (80.88% vs. 54.05%,χ2 =8.451,P<0.01).The shape,color,thought and energy score of the observation group were significantly higher than those of the control group (t=4.819,3.519,4.735,6.804,all P<0.01).The pregnancy rate of the observation group was significantly higher than that of the control group (25.71%vs.5.71%,χ2 =5.285,P<0.05). Conclusion Diane-35 combined with ovulation induction in the treatment of polycystic ovary syndrome infertility has significant clinical efficacy .
5.Epidemiological study of the genes conferring aminoglycoside resistance in 36 strains of multidrug-resistant Acinetobacter baumannii

Objective To investigate the prevalence of genes conferring aminoglycoside resistance in multidrug-resistant strains of Acinetobacter baumannii (MDR-ABA).Methods Multidrug-resistant A.baumannii strains were isolated during the period from August to November 2012 from patients in the affiliated hospital of Jiangsu University and the First Hospital of Zhen-jiang.Kirby-Bauer diffusion method was used to determine the susceptibility of these strains to antimicrobial agents.PCR was performed to detect the aminoglycoside resistance genes.Results The 36 MDR-ABA strains showed high resistance rates to most antimicrobial agents except cefoperazone-sulbactam.The prevalence of the genes conferring aminoglycoside resistance, aac (3)-I,aac (6’)-Ib,aph (3’)-I and armA,was 72.2% (26/36),72.2% (26/36),80.6% (29/36)and 80.6% (29/36), respectively.Conclusions The MDR-ABA strains in this study are highly resistant to antimicrobial agents,which is closely as-sociated with the genes conferring aminoglycoside resistance.
6.Stakeholder analysis of prevention and control of obesity among middle school students

Objective:
To identify the stakeholders of prevention and control of obesity among middle school students and analyze their motivations and interest demands for participation, in order to provide constructive recommendations for facilitating their effective engagement of stakeholders in prevention and control of obesity among middle school students.
Methods:
Stakeholders and their types were identified through a combination of literature analysis and consultation with 17 experts in children and adolescents for prevention and control of obesity. From December 2023 to January 2024, by applying convenience sampling, face to face semi structured interviews were conducted with 11 individuals engaged in prevention and control of obesity for middle school students and focus group interviews were conducted with 48 students and parents. Thematic analysis was employed to obtain motivations and interest demands for stakeholder involvement in prevention and control of obesity.
Results:
A total of 29 subcategories within 7 major categories of stakeholders were identified, including government and relevant functional departments, non profit organizations and communities, forprofit organizations and individuals, schools, professional and technical personnel, families and individuals, and scientific research institutions, with 28 subcategories being confirmed stakeholders. Among these stakeholders, there were 3 types of cooperative relationships:management, cooperation, and service. There were some factors hindering cooperation among stakeholders including imperfections in specialized policies and lack of long term cooperative mechanisms, flaws and interest conflicts in the evaluation system, insufficiency of material resources, and poor health values. In response to these challenges, interviewees proposed to convene a working group for the prevention and control of adolescent obesity. They also suggested making a contract of responsibility, strengthening supervision over for profit organizations, enhancing advocating efforts for prevention and control of obesity, and increasing financial support.
Conclusions
The prevention and control of obesity for middle school students can be facilitated by forming a collaborative alliance based on the roles and relationships of stakeholders identified in the present study. Perceived conflicts of interest among stakeholders can be reconciled by employing strategies of interest integration, goal alignment, and spiritual integration, to enhance the feasibility, participation and sustainability of obesity intervention.
7.p16 (CDKN2) Gene Transfer Mediated by Retrovirus Vector Inhibits Proliferation of Leukemia Cell Line K562

The aim of this work was to test the effect of p16 on the proliferation of leukemic cells and its potential in gene therapy for leukemia. The full-length p16 cDNA was transfered by recombinant retrovirus vector into leukemia cell line K562, which is homozygous p16 deletion and retains functional retinoblastoma (RB) protein. The cell proliferation was tested in liquid and in soft agar culture after transduction of p16 retrovirus. The results showed a strong inhibition of cell proliferation. Phosphorylation of RB protein was also inhibited. The findings demonstrated that p16 (MTS/CDKN2) inactivation is a significant factor in the genesis and progression of leukemia and p16 could be a candidate gene for gene therapy in leukemia.
8.The effect of health education on medication compliance and rehabilitation effect of patients with epilepsy after discharge

Objective To evaluate the effect of health education on the compliance and health status of epileptic patients after discharge. Methods A total of 88 patients with epilepsy were selected and divided into control group (January to June in 2014) and experimental group (July to December in 2014)with 44 patients each.During hospitalization,the patients in control group were given routine health education,discharge guidance,on the basis of the control group,the patients in experimental group were added health education after discharge. The health status were evaluated at the time of admission and discharge(1 days before discharge)and 1,3,6 and 12 months after discharge.The medication compliance and clinical efficacy of the two groups were evaluated at 12 months after discharge. Results The medication compliance at 12 months after discharge was 97.73%(43/44)in experimental group and 63.64% (28/44)in control group,and there was significant difference(x2=15.92,P<0.01).The total effective rate was 84.09%(37/44)in experimental group and 40.91%(18/44)in control group,and there was significant difference (x2=12.31, P<0.01). Repeated measurement of variance analysis showed that there was no significant difference between two groups of self testing health score during hospitalization (P>0.05), there were significant differences in the health status at 1, 3, 6, 12 months after discharge between two groups, treatment effect, time effect, interaction effect (F =57.50, 5.69, 8.35, P<0.01). Conclusions Health education after discharge can improve the medication compliance, health status and clinical efficacy of patients with epilepsy and promote their rehabilitation.
9.Glutamine supplementation provides neuroprotection by inhibiting microglia-mediated neuroinflammatory response after traumatic brain injury in rats

Objective To investigate the effects and mechanisms of Glutamine (Gln) supplementation on neurobehavioral outcome,neuronal apoptosis,microglia polarization,and neuroinflammatory response after traumatic brain injury (TBI) in rats.Methods TBI animal models were established using Feeney's method.Sixty Wistar rats were randomly divided into three groups:control group (Con),traumatic brain injury group (TBI),and glutamine supplementation group (TBI+Gln).We measured rat behavioral outcomes by modified neurologic severity score (mNSS) tests at day 1,3,7 and 14 after TBI.Apoptotic neurons were determined by Nissl staining.The microglia polarization relatived protein (Iba-1,CD16,CD86) expressions in TBI cerebral cortices were determined by immunohistochemistry staining and western blotting,respectively.While,the serum levels of tumor necrosis factor-α (TNF-α),interleukin (IL)-1 and interferon (IFN)-γ were tested by enzyme linked immunosorbent Assay (ELISA).Results Compared with the Con group,the levels of neurobehavioral outcome,neurons apoptosis,microglia polarization and neuroinflammatory factors were significantly increased in the other two groups (P=0.00).Compared with the TBl group,glutamin supplementation improvedneurobehavioral outcome [7 d:(10.74±0.25) points vs.(8.94±0.24) points,P=0.01;14 d:(8.77± 0.16) points vs.(7.43±0.13) points,P=0.03].Meanwhile,glutamin supplementation suppressed the apoptotic rates of neurons [3d:(80.18±8.38)% vs.(65.47±7.02)%,P=0.01;7 d:(58.90±6.12)% vs.(42.73±4.88)%,P=0.01;14d:(39.56±2.95)%vs.(31.12±3.16)%,P=0.01],inhibited protein expressions of Iba-1 and CD16,and increased the protein expression of CD86,which promoted the phenotypic shift of microglia from M1 to M2 phenotype,inhibited microglial activation,and thus reduced TBI-induced neuroinflammatory factors [TNF-α:(125.42 ± 12.81) pg/ml vs.(74.36 ± 9.25) pg/ml,P =0.01;IL-1:(69.04±8.48) pg/ml vs.(34.73±3.92) pg/ml,P=0.01;TNF-α:(89.75±9.40) pg/ml vs.(45.62±6.64) pg/ml,P=0.02].Conclusion Glutamine supplementation can markedly reduce neuron apoptosis and improve neurological outcomes after TBI,possibly mediated by promoting the phenotypic shift of microglia from M1 to M2 phenotype and thus reducing TBI-induced neuroinflammatory factors.
10. Effects of transcatheter aortic valve replacement in patients with severe aortic valve stenosis

Objective:
To investigate the effects of transcatheter aortic valve replacement (TAVR) in patients with severe aortic valve stenosis.
Methods:
The clinical data of 130 patients with severe aortic valve stenosis, who underwent TAVR in our hospital with self-expanding valve between January 1,2010 and October 30, 2016, were analyzed retrospectively. The patients were divided into calcific aortic valve stenosis (CAS) group (112 cases) and non-calcific aortic valve stenosis (NCAS) group (18 cases) according to 3D volume-rendering reconstruction under multiple detector computed tomography before TAVR. The baseline clinical features, imageology results, procedural details, and clinical prognosis were compared between the 2 groups.
