Objective To explore the effect of the combination of benazepril and beraprost sodium on urinary microalbumin (UMA), serum high sensitivity C-reactive protein (hs-CRP) and TNF-α in patients with diabetic nephropathy(DN) in early stage. Methods From Mar. 2010 to Mar. 2013, 120 patients with early-stage DN were randomly divided into Benazepril Group, Beraprost Sodium Group and Combination Group. Each group consisted of 40 patients with medical therapy in 20-week duration. The levels of 24 h-UMA, serum hs-CRP and TNF-α were detected and compared before and after therapy. Results After treatment, the levels of 24 h-UMA, serum hs-CRP and TNF-αall decreased significantly in three groups (P<0.05) and there were lower 24 h-UMA, serum hs-CRP and TNF-αin Combination Group than those in Benazepril Group and Berapros sodium Group (P <0.05). Conclusion There is coordinating protection of combination of benazepril and beraprost sodium on diabetic nephropathy in early stage, possibly being related with the inhibiting of micro-inflammation.

Objective To investigate the clinical characteristic of vernier acuity in age-related macular degeneration (AMD) patients. Methods The vernier acuity test soft wear system was developed to detect the 23 cases (39 eyes) of AMD patients. Tweenty-one eyes were atrophic type and 17 eyes were exudative type. Two fixed targets and a movable target are shown on the computer screen. The examinee was asked to adjust the position of the central target and the relationship between it and align them by using a track ball. The computer automatically recorded the deviations of distances between the movable target and the specific one, and then computed and analysed the results of average threshold and variance. ObjectiveTo investigate the methods of reducing eye injury in phacoemulsification．MethodsPhacoemulsification with intraocular lens（IOL）implantation was performed on 98 cases（98 eyes）of cataract using a new type of phaco tip（MicroTip）with high vaccum and low energy．All the nuclei was harder than Grade Ⅳ．Postoperative visual acuity and eye injury were observed，including corneal edema，anterior chamber in flammation，intraocular pressure，IOL position and retinal condition．ResultsPostoperative visual acuity is 0.5 or better in all cases，1.0 ? or better in 28 cases（28.6％）．The complication during operation，such as anterior chamber hemorrhage，posterior capsule rupture and vitreous loss，did not occur．Postoperative complications were very slight．Two eyes（2％）suffered mild corneal edema that lasted no more than a week．No other complications were found．ConclusionMicroTip is a kind of safe and effective facility for phacoemusification，with which we can reduce the eye injury，especially dealing with the hard nuclei．

AIM: To explore the role of PI3K /PKB pathway in the growth inhibition effect of rabbit lens epithelial cells (LECs) induced by (-)-epigallocatechin-3-gallate (EGCG). METHODS: Rabbit LECs were obtained by tissue culture; MTT colormetric assay was used to study growth of LECs; Western blotting was used to study the kinsae phosphorylation- and nonphosphorylation- level of PKB. RESULTS: (1) When LECs were preincubated with 25, 50 ?mol/L LY294002 for 1 h, 50 ?mol/L EGCG had little influence on the proliferation of LECs. OBJECTIVE： To confirm the efficacy and safety of latanoprost in treating glaucoma.METHODS： In a double blind， randomized control clinical trial， we compared the efficacy and adverse drug reactions of once daily topically applied 0.005％ latanoprost with those of twice daily 0.5％ timolol for 12 weeks in patients with open angle glaucoma or ocular hypertension.RESULTS： The study included 46 patients（22 pts.randomized to latanoprost treatment， 24 pts.to timolol） ， 46 patients remained at the end of the study.Comparing with baseline diurnal intraocular pressure（IOP） ， the IOP reduction（mean±standard deviation） achieved with latanoprost（7.86±2.39） mmHg， （31.1％， P<0.001），and timolol（6.24±2.58）mmHg （24.9%，P<0.001），the difference between the two groups（1.62mmHg） being significant（P<0.01）. Two patients treated with latanoprost had foreign body sensation. No other ocular and systemic adverse reactions related to the two drugs were found. CONCLUSION: The results of this study demonstrated that 0.005% latanoprost applied once daily is well tolerated and more effective in reducing IOP than 0.5% timolol applied twice daily. Thus, latanoprost has the potential for becoming one of the ideal antiglaucoma drugs.

Objective To explore the expression difference of cytokines related to nuclear factor kappa B (NF-κB) signaling pathway among different traditional Chinese medicine (TCM) syndromes in the patients with diabetic nephropathy (DN).Methods Serum tumor necrosis factor-α (TNF-α),interleukin-1 (IL-1),interleukin-6 (IL-6) and interleukin-8 (IL-8) in 146 patients with DN and 62 individuals undergoing healthy physical examination in the control group were measured.Results The levels of serum TNF-α,IL-1,IL-6 and IL-8 in various DN groups were significantly higher than those in the control group (P＜0.05).The levels of serum TNF-α,IL-1,IL-6 and IL-8 were significantly different among groups of different TCM syndromes (P＜0.05).With the progression of DN,all cytokines levels showed a gradually increasing trend,moreover the increased extents and time were different in different TCM syndrome types of DN.Conclusion The levels of serum TNF-α,IL-1,IL-6 and IL-8 related to NF-κB signaling pathway are correlated with different TCM syndromes types and may play a role in the occurrence and progression of different TCM syndrome types of DN.

Objective To compare and evaluate the performance and applications of three assays in preliminary screening of dengue infections.Methods This study was designed as a retrospective study.Clinical data were obtained from 2 137 borderline cases from September to October in 2014 and 2015.Markers of dengue infections of serum samples were detected by NS1 antigen captured ELISA, dengue NS1 detect rapid test and dengue IgM detect rapid test, respectively.Chi square test was used to compare the preliminary screening value of these 3 methods.Forty-eight diagnosed patients were also examined in the 1st-3rd day, 4th-5th day, 6th-7th days and 14th day after infection to access the sensitivities and specificities of three assays.Results Nine hundred and fifty-seven ( 57.2%) cases in 2014 and 48 ( 10.4%) cases in 2015 were diagnosed to be Dengue fever of 2 137 borderline cases.The overall sensitivity of NS1-ELISA was superior to NS1 and IgM rapid detect test (χ2 =40.865,P<0.001;χ2 =151.383,P<0.001).No significant differences were found in specificity between three assays(χ2 =0.661,P=0.416;χ2 =0.548,P=0.459; χ2 =2.397,P=0.122).The NS1 detecting assays were sensitive in 7 days after infection, but the sensitivity of IgM detecting assay increased over time.Conclusions NS1 detecting assays had good sensitivities and specificities, which can be used as an important method in preliminary screening of dengue infection.ELISA or rapid test can be selected according to epidemic situation.

Background Mesenchymal stem cells (MSCs) have been applied in basic and clinical researches of organ transplantation.Our previous study showed that intravenous injection of MSCs prolonged corneal allograft survival in rat.However,the effect of local administration of MSCs on corneal allograft rejection remains unclear.Objective The aim of this study was to investigate the effect of subconjunctival injection of MSCs on corneal allograft rejection in rat model of keratoplasty.Methods MSCs were isolated and cultured from femur and tibia bone marrow of clean Wistar rats,and then the cells were identified by induced differentiation of osteoblast and adipocyte.The third generation of MSCs was used in subsequent experiment.Allogenic penetrating keratoplasty was performed with the bilateral corneas of 20 Wistar rats as donor grafts and the right eyes of 40 Lewis rats as recipients.PBS 0.1 ml containing 2 × 106 MSCs and 0.1 ml PBS only was subconjunctivally injected immediately and postoperative 3 days respectively in randomized two groups,and another 6 normal Lewis rats served as the normal control group.Corneal rejection response was evaluated under the slit lamp after surgery based corneal opacity,edema and neovascularization,and the grafts were scored according to the criteria of Larkin.The corneal samples were extracted from 12 rats of the PBS control group and the MSCs group separately 10 days after surgery.The relative expressions of Th1 cytokines (interferon-γ [IFN-γ] mRNA and interleukin-2 [IL-2] mRNA) and Th2 cytokines (IL-4 mRNA and IL-10 mRNA) were detected by real-time quantitative PCR.Protein levels of IL-4 and IL-10 proteins in the corneas were assayed by ELISA.All experimental protocols involving rats were approved by the laboratory animal care and use committee of the Tianjin Medical University and treated with the ARVO statement for the use of animals in ophthalmic and vision research.Results The cells grew well with the orange stain for alizarin red in differentiated the osteoblasts and red stain for Oil red O in differentiated adipocytes.The survival time of corneal graft in the MSCs group was (11.8±1.6) days,it was significantly longer than (9.6±1.4) days in the PBS control group (P=0.004).The levels of IL-4 mRNA and IL-10 mRNA in the MSCs group were significantly higher than those in the PBS control group (both at P =0.00);while the levels of IFN-γ mRNA and IL-2 mRNA were not significantly different between the groups (both at P＞0.05).The IL-10 protein contents were (22.74 ±7.06),(68.40±12.83) and (215.41 ±44.66)pg/ml in the normal control group,PBS control group and MSCs group,showing significant difference among the three groups (F =55.06,P =0.00) and a significant increase in the MSCs group compared with the PBS control group and the normal control group (both at P ＜ 0.05).Conclusions Subconjunctival injection of MSCs prolongs the survival time of cornea allograft in penetrating keratoplasty probably by modulating the balance between Th1 and Th2 cytokines,especially by up-regulating Th2 cytokines.
