Objective: To measure the expression of E cadherin (E cad) and ? catenin (CTN) in transitional cell carcinomas (TCC) of bladder and to study the relationship between the expression and biological behavior of the carcinoma. To measure the expression levels of matrix metalloproteinase 2 (MMP 2) and its inhibitor, tissue inhibitor of metalloproteinase 2 (TIMP 2), and to evaluate their relationship with the bladder transitional cell carcinoma.RT PCR and immunohistochemistry method were used to measure the levels of MMP 2 and TIMP 2 in 61 cases of bladder transitional cell carcinoma and 23 samples of tissues adjacent to the cancer. 3.Effects of angiostatin gene transfection on biological behaviors of bladder carcinoma BIU-87 cell line

Objective To explore the expression and effects of extragenous angiostatin gene in human bladder carcinoma BIU-87 cell line. Methods A full human angiostatin gene was cloned into expression vector pcDNA3.1(+) and transfected into human bladder carcinoma BIU-87 cells by lipofectamine, and the positive clone was screened by G418. The growth characteristics of BIU-87 cells before and after transfection were compared.The expressions of angiostatin protein in BIU-87 cells were examined by immunofluorescence assay, immunohistochemistry and Western blotting, and the activities of angiostatin protein were examined by endothelial cell inhibition assay and the chicken-embryo chorioallantoic membrane assay. Results The expression of extragenous angiostatin gene had no inhibitory effect on the growth of BIU-87. The transfected cells could inhibit the umbilical vein endothelial cell growth in vitro and chicken-embryo choriallantoic membrane angiogenesis in vivo. Conclusion The results indicated that the angiostatin gene clones could be transfected into BIU-87 bladder cancer cells, and angiostatin could inhibit endothelial cell proliferation and neovascularization.
4.Effect of L-arginine Therapy on Serum Nitric Oxide and Endothelin in Acute Hypoxic Rat

PurposeTo study the changes of serum nitric oxide(NO)and endothelin(ET)levels by L-Arg therapy in acute hypoxic model. MethodsNADPH diaphorase histochemical staining method, Griess biochemical assay and radioimmune assay were applied to investigate the changes of nitric oxide synthase (NOS) ,NO, cGMP and ET in normal, hypoxic and L-Arg treated hypoxic rats. Resultsduring acute hypoxia, while serum ET rose up significantly, both the levels of serum NO and cGMP and the activity of NOS were significantly lower. After the application of L-Arg therapy, the activity of NOS did not change significantly, but the levels of serum NO and cGMP rose up significantly, and the level of serum ET was to the opposite direction. ConclusionsNO and ET may modulate hypoxic pulmonary hypertension, and acute hypoxia can result in acute hypoxic pulmonary hypertension. L-Arg can reverse the acute hypoxic hypertension.
5.The expression of serum procalcitonin in sepsis patients and its influence on prognosis

Objective To investigate the expression of serum procalcitonin in patients with sepsis and its influence on prognosis.Methods From December 2014 to December 2016,119 cases of sepsis patients were enrolled in the hospital.According to the severity of the disease,49 cases in the early sepsis group,37 cases in the severe sepsis group and 33 cases in the septic shock group.According to the prognosis of 28 d after admis-sion,the patients were divided into the survival group(95 cases)and the death group(24 cases).Serum procal-citonin was measured by double antibody sandwich enzyme-linked immunosorbent assay.The changes of WBC count,C reactive protein and APACHE Ⅱ score,serum procalcitonin level,serum procalcitonin level and sur-vival group and death group were compared.Results Septic shock of white blood cell count,C reaction protein and APACHEⅡ were higher than those of early sepsis group and severe sepsis group(P<0.05);severe sep-sis group cell counts,C reactive protein and APACHE Ⅱ were higher than those of early sepsis group(P<0. 05).The septic shock group of serum procalcitonin levels higher than the early sepsis group,severe sepsis group(t=33.136,21.863,P<0.05);severe sepsis group serum procalcitonin levels higher than the early sep-sis group(t=16.876,P<0.05).The level of serum procalcitonin in the death group was higher than that in the survival group(P<0.05).Conclusion The level of serum procalcitonin in patients with sepsis is obvious-ly higher.With the severity of the disease,the serum procalcitonin level is higher,and can be used as an indica-tor of prognosis evaluation.
