1.Value of contrast -enhanced ultrasound in evaluating peripancreatic vascular invasion and resectability of pancreatic cancer

Objective To investigate the value of preoperative contrast -enhanced ultrasound (CEUS)in evaluating peripancreatic vascular invasion and resectability of pancreatic cancer.Methods A retrospective analysis was performed for the clinical data of 25 patients with pancreatic cancer confirmed by surgical exploration or postoperative pathological examination,who visited Shengjing Hospital of China Medi-cal University from December 2012 to October 2014,and all the patients underwent CEUS before surgery.The relationship between the le-sion and the peripancreatic vessels was observed,the degree of invasion was evaluated,and related scores were measured.These results were compared with the results of intraoperative exploration and postoperative pathological results.The rate of peripancreatic vascular invasion, sensitivity and specificity of preoperative CEUS in evaluating the degree of peripancreatic vascular invasion,and sensitivity,specificity,pos-itive predictive value,negative predictive value,overall coincidence rate,and Youden index of CEUS in evaluating the resectability of pan-creatic cancer were calculated.Results Preoperative CEUS showed that 42 vessels were invaded,while intraoperative exploration showed 41 vessels were invaded.A total of 17 patients were considered resectable by CEUS,while 15 patients were considered resectable by intraop-erative exploration.In evaluating the resectability of pancreatic cancer,CEUS had a sensitivity of 93.3%,a specificity of 70.0%,an over-all coincidence rate of 84.0%,a false positive rate of 30.0%,a false negative rate of 6.7%,a positive predictive value of 82.4%,a nega-tive predictive value of 87.5%,and a Youden index of 0.633.Conclusion Compared with intraoperative exploration,CEUS has higher sensitivity and specificity in evaluating the degree of peripancreatic vascular invasion and higher sensitivity,specificity,and coincidence rate in evaluating the resectability of pancreatic cancer.CEUS can accurately evaluate the degree of peripancreatic vascular invasion and provides a new method for preoperative evaluation of the resectability of pancreatic cancer.
2.Three-dimensional ultrasonography of normal vascular structures and vascular diseases

Objective To assess the clinical value of three-dimensional ultrasonography (3DUS) in normal vascular structures and vascular diseases. Methods Thirty-nine 3DUS images were reconstructed,including 17 normal vascular structures and 22 vascular diseases. 3DUS reconstruction was introduced by the maximum intensity projection (MIP) method. A series of 3DUS image was performed using GE Logiq 500 and Logiq 700. Results In all cases, 3DUS images were constructed within 10-30 seconds after free hand scanning. Internal, external and common carotid arteries were also showed in the same rotating 3DUS display simultaneously. The continuity of the transplanted hepatic portal vein were demonstrated clearly in rotating mode 3DUS display and consistent with angiography or computer tomography findings. In cases of various vascular diseases, the degree and location of stenosis or aneurysm of the vessels could be stereoscopically visualized, easy to understand especially for a surgeon who evaluates a preoperative state. Conclusions The 3DUS can display normal or abnormal vascular anatomy clearly. 3DUS image could supply more information than original two-dimensional image, and is useful in the diagnosis of various vascular diseases.
3.Overexpression of Bax and induction of apoptosis in human retinal pigment epithelial cell lines

Purpose To investigate bax expression and induction of apoptosis in normal cultured retinal pigment epithelium (RPE) cells . [WT5”HZ]Methods [WT5”BZ]Cultured human RPE cells were transfected by P MDNA3 hbax ,which incoded the whole bax gene and may be induced by Zn 2+ under the MTII promoter, through lepofectin mediated protocol.The tested RPE cells were divided into three groups of A, P MDNA3 hbax transfected ;B,P MDNA3 (nude vector) transfected and C,normal RPE cells.After transfection, DNA gel electrophoreses were performed ,the tested RPE cell cycles were analyzed with flow cytometry (FCM). Results The gel electrophoretogram showed DNA ladder phenomenon,FCM confirmed the apoptosis of RPE cells P MDNA3 bax transfected , consisting of significant apoptotic peak sited before the G 1 phase and the apoptotic rate was 36%. Conclusion The foreign bax gene can be effectively conducted into the RPE cell through lepofectin mediated protocol and induced expression . The foreign bax overexpression may induce the cultured human RPE cell susceptibility to apoptosis.
4.Dissociation and purifying of the retinal pigment epithelial cells by gradient centrifugation

Objective To investigate the effect of dissociation of the human retinal pigment epithelium (RPE) cells by ficoll hypaque gradient centrifugation. Methods The primary human RPE cells and subcultured human RPE cells were dissociated with ficoll gradient centrifugation solution (d= 1.077 g/ml) and the same divided cells as the control were dissociated with routine normal culture medium centrifugation. The Trypan blue (0.4%) rejection staining was used, and the mouse anti-human monoclonal antibody and fluorescein isothiocyanate (FITC) labeled rabbit anti-mouse IgG were utilized for indirect immunoreactivity for the test of human cytokeratin (CK) in active RPE cells cytoplasm. Flow cytometry assay was used to analyzed the percentages of CK positive staining RPE cells. simultaneously, the cells configuration, growth condition, the rate of clone formation, and the purifying result were observed under the fluorescent and confocal microscope. 5.Activation and role of signal transduction pathway of epidermal growth factor-epidermal growth factor receptor-mitogen activated protein kinase in proliferation of human retinal pigment epithelial cells

Objective To investigate the activation and role of signal transduction pathway of epidermal growth factor (EGF)-epidermal growth factor receptor (EGFR)-mitogen activated protein kinase (MAPK) in proliferation of human retinal pigment epithelial (RPE) cells. Methods Human RPE cells were stimulated with 0.1%,10% foetal calfserum (FCS) and EGF(0.1, 1, 10, 50 and 100 ng/ml)in 0.1% FCS Dulbeco′s modified Eagle′s medium (DMEM) and in 10% FCS DMEM for 3 days, respectively. Immunohistochemical staining and in situ hybridization were used to observe the expressions of EGFR protein and EGFR mRNA,respectively. Activation of MAPK was detected by immunohistochemical method with specific anti-phosphorylated ERK 1/2 antibody. Results The optimal concentrations of EGF were 10 ng/ml in 0.1% FCS DMEM and 1 ng/ml in 10% FCS DMEM. After 3 days of stimulation with EGF, phosphorylated ERK 1/2 staining was detectable in nucleus of RPE cells, whereas cells presented immunostaining for phosphorylated ERK 1/2 in the cytoplasm before stimulation. Conclusions EGF may improve the expression of EGFR protein and EGFR mRNA of RPE cells, and induced MAPK nuclear translocation in a concentration-dependent manner. EGF-EGFR-MAPK signal transduction pathway may play a key role in RPE cells proliferation, and serum exerts an important acceclerating function in the process.
6.Effect of α-phenyl-N-tert-butylinitrone on Expression of Nerve Growth Factor in Spinal Cord Injured Rats

Objective To explore the effect of α-phenyl-N-tert-butylinitrone (PBN) on expression of nerve growth factor (NGF) in serum and spinal cord tissue in rats after spinal cord injury (SCI). Methods 174 female Sprague-Dawley rats were randomly assigned to following groups: normal control group (n=54), normal saline control group (NS group, n=60, intrathecally injected normal saline 15 μl), and PBN group (n=60, intrathecally injected PBN, 3 mg, 15 μl). The model was established with New York University blow device (150 kDyne, 1 s dwell time). PBN was intrathecally injected into the damaged areas 30 min after operation, then once a day for 7 days. The Basso-Beattie-Bresnahan (BBB) Locomotor Rating Scale was used to assess the rats 3 days and 1 day before, and 1 day, 5 days, 10 days, 15 days, 20 days,25 days, 30 days and 35 days after SCI. NGF in the injured spinal cord tissue and serum was measured 1 h, 12 h, 24 h, 48 h, 3 days, 7 days,14 days and 21 days after SCI. Results NGF increased in serum but not in spinal cord. The ratio of NGF/total protein in serum rose and peaked 48 h after SCI, and the ratio was higher in NS group (0.92%±0.02%) than in PBN group (0.77%±0.05%) (P=0.021). BBB scores increased from the 9th day, and PBN group improved better than NS group (P<0.01). Conclusion PBN could reduce the expression of NGF in the SCI rats, and promote the recovery of neurol function.
7.Research advances in SULF2 and its relationship with hepatocellular carcinoma

Cell secreted sulfatase 2 is an endogenous sulphate esterase, which can hydrolyze the sulphate groups in extracellular matrix or on the chain of heparan sulphate on the outer surface of cell membrane, which can dissociate the growth factor bound to heparan sulphate proteoglycan, improve the local concentration of growth factor and activate the downstream signal. Sulfatase 2 is highly expressed in a variety of tumors, and the increased expression of sulfatase 2 in hepatocellular carcinoma is associated with poor prognosis. Sulfatase 2 can promote tumor progression by activating multiple signaling pathways in hepatocellular carcinoma cells. Inhibit the activity of hepatocellular carcinoma cell sulfatase 2, and then inhibit the proliferation, migration and tumorigenesis of hepatocellular carcinoma cells. In this paper, the research progress of sulfatase 2 and its role in the occurrence and development of hepatocellular carcinoma is summarized.
8.Activated hepatic stellate cells and its relationship with hepatocellular carcinoma

Interactions between hepatocellular carcinoma (HCC) cells and the tumor stromal microenvironment have profound effects on tumor growth,epithelial-mesenchymal transition (EMT),invasion and metastasis.Activated hepatic stellate cells (HSCs) are the major subtype of stromal cells in the liver tumor microenvironment.HCC cells can induce the activation of HSCs during carcinogenesis,while activated HSCs promote HCC cells growth and migration through secreting growth factors,inducing angiogenesis and immune suppression.Bidirectional interactions between HCC cells and HSCs may function as an "amplification loop" to further enhance metastatic growth in the liver.In this review,we summarized the most recent data from the research on HSCs and its relationship with HCC.
9.Logistic Regression Analysis of Gallbladder Lesions of≥1 cm in Diameter Diagnosed by Ultrasound

Purpose To establish Logistic regression model of gallbladder lesions of≥1 cm in diameter diagnosed by ultrasound, and to filter benign and malignant sonographic features. Materials and Methods The sonographic features were retrospectively analyzed in 165 patients with gallbladde apophysis lesions of≥1 cm in diameter which confirmed by pathology, including the number of lesions, size, shape and basal width, gallstones, continuous gallbladder wall continuous, blood flow signals detected by color Doppler flow imaging. Logistic regression model with bipartition was established by multivariate Logistic regression analysis, and the efficiency of Logistic regression model was evaluated to predict benign or malignant of these lesions. Results Three characteristic variables, including lesion morphology, basal width and flow signals, were took into the Logistic regression model by binary Logistic regression analysis, which was the sensitive indicators can differentiate the benign or malignant gallbladder lesions. The accuracy, sensitivity and specificity of this model were 97.0%, 93.8%and 97.3%for predicting the benign or malignant gallbladder apophysis lesions≥1 cm in diameter, respectively. Area under ROC was 0.979. Conclusion Binary Logistic regression analysis can filter the sonographic features which can differentiate the benign or malignant gallbladder apophysis lesions≥1 cm in diameter, and lesion morphology, basal width and flow signals are of important differential diagnosis value of benign lesions or malignant lesions.
10.Diagnosis of skull base chondrosarcoma and analysis on the causes of misdiagnosis

Objective To investigate the diagnosis and the causes of misdiagnosis of skull base chondrosarcoma.Methods The clinical data of 7 cases of skull base chondrosarcoma confirmed by pathology from April 2009 to February 2014 were analyzed retrospectively.Results Only 1 case of preoperative diagnosis was correct in 7 cases of skull base chondrosarcoma, other 6 cases were misdiagnosed, of which 2 cases were misdiagnosed as trigeminal schwannoma, 3 cases were misdiagnosed as chordoma, 1 case was misdiagnosed as meningioma,the misdiagnosis rate was 85.7%.Conclusion Chondrosarcoma of the skull base is rare, so clinicians are lack of knowledge, and non-characteristic clinical features and non-characteristic radiographic manifestations contribute to misdiagnosis.Strip calcification on CT and honeycomb reinforced on enhanced MRI existing at the same time,which prompts to diagnosis chondrosarcoma of the skull base.
