Journal of Guangzhou University of Traditional Chinese Medicine 2017;34(5):729-733
Objective To observe the hypoglycemic, hypolipidemic and antioxidant effects of Zhuye Shigao Decoction (ZSD) on experimental type 2 diabetes mellitus rats. Methods Type 2 diabetes mellitus rat model was induced by intragastric administration of high fat diet(10 mL/kg) and intraperitoneal administration of streptozotocin (30 mg·kg-1). Sixty male Sprague-Dawley rats were randomized into normal group, model group, low-, moderate-and high-dose ZSD groups (in the dosage of 3.6, 7.2, 14.4 g·kg-1·d-1 respectively), and Metformin group (0.15 g·kg-1·d-1) , 10 rats in each group. With fasting blood glucose, urine glucose, glycosylated hemoglobin, total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol(LDL-C), serum insulin, serum superoxide dismutase(SOD) and malondialdehyde (MDA) as the main indexes, we evaluated the influence of ZSD on the blood glucose and lipid levels as well as oxidative stress in type 2 diabetes mellitus rats. Results Compared with the model group, the levels of blood glucose, urine glucose and glycosylated hemoglobin, and the contents of TC, TG, MDA were decreased (P <0.05), whereas SOD activity and serum insulin level were obviously increased (P < 0.05) in ZSD groups. ZSD showed dose-related effectiveness in the treatment of diabetic rats. Conclusion ZSD has certain hypoglycemic, hypolipidemic and antioxidant effects in treating type 2 diabetes mellitus.

Objects To assess the effect of BMP(bone morphogenetic protein) in orthotopit grafting with an autogenous cranial bone flap in cranioplasty. Methods 45 cases (48 locations ) have been reconstructed by orthotopic repair with autogenous bone flap, it has been divided into two group;26 locations of them using porous autogenous bone flap plus BMP (group Ⅰ), 22 locations of them only using porous autogenous bone flap(group Ⅱ), All cases have been closely followed up for 1~2 years with X-ray and CT , including postoperative complications, absorption or regeneration of the bone flap. Results No flexible with the porous autogenous bone flap was seen , and the appearance was perfectly. The bone graft survival and besetting apposition were all right. The gap disappears gradually. It is obvious that the osteotylus come into being. The density of bone flap was normal in group Ⅰ. 5 cases in group Ⅱ have shown that bone partial absorption occurred and the density reducing in the center of the flaps, and with the significant difference. Conclusions The local application of BMP can avoid bone flap absorption and stimulate osteotylus morphofogenesis.

Objective To observe the characteristic of the functional magnetic resonance imaging (fMRI) brain map in health adult undergoing clenching and relaxing the fist, for exploring the essence of the fMRI brain map in patients suffering from motor dysfunction by cerebrovascular accidents. Methods Twelve healthy volunteers had been chosen to partake the experience. Everyone had accomplished the following three actions separately: (1) Only clenching and relaxing the fist of left hand. (2) Only clenching and relaxing the fist of right hand. (3) Clenching and relaxing the fist of both hands at one time. The data had been analyzed statistically using analysis of functional neuroimages (AFNI) software. Results Under condition of F (6,1121), P = 0.005. Only clenching and relaxing the fist of left hand had gained the following brain functional area: right precentral gyms, left parietal, right superior temporal gyrus, right parietal, right parahippocampal gyrus, right superior frontal gyrus, right medial frontal gyrus, left precuneus, right superior parietal lobule, right middle frontal gyrus, left superior frontal gyrus. Only clenching and relaxing the fist of right hand had gained the following brain functional area: left precentral gyms, left postcentral gyrus right parietal, right medial frontal gyrus. Clenching and relaxing the fist of beth hands simultaneously had gained the following brain functional area: left precentral gyms, left postcentral gyrus, right precentral gyrus, right postcentral gyrus. Conclusions Hand movement (clenching and relaxing the fist) has its own specific brain activated areas. The brain areas activated by clenching and relaxing the fist of both hands simultaneously concentrate in the motor area of both cerebral hemisphere. The brain areas activated by clenching and relaxing the fist of single hand contain not only the motor area, but also the supplementary motor area. As compared with the right handedness, the brain areas activated by clenching and relaxing the fist of left hand is more widespread.

Objective To explore the relationship of UHRF1 with the clinicopathological characteristics of colorectal cancer (CRC) patients, as well as the effects of lentivirus transfection overexpression and knockdown of UHRF1 on the proliferation, invasion, and migration of CRC cells and the possible signaling pathways. Methods The expression of UHRF1 mRNA and protein in CRC tissues and adjacent tissues was detected by immunohistochemical staining and RT-PCR. The effects of the constructed UHRF1 overexpression- and knockdown-group cells on the expression of UHRF1, related molecules in the WNT signaling pathway, and MMPR9 were examined by Western blot and RT-PCR. EDU and Transwell assays were used to detect changes in the proliferation, migration, and invasion of CRC cells. Results (1) In the TCGA database and clinical data, the mRNA and protein expression levels of UHRF1 in CRC cancer tissues were significantly higher than those in adjacent normal tissues. UHRF1 expression was closely correlated with TNM stage, N stage, and M stage. Patients with low UHRF1 expression in TCGA had better 5-year OS and disease-specific survival. The area under the ROC curve of UHRF1 for predicting 1-, 3-, and 5-year OS were 0.634, 0.652, and 0.771, respectively. The 3-year OS in the clinical data also showed the same survival benefit. UHRF1 overexpression was a poor prognostic factor for CRC patients. (2) After UHRF1 overexpression, the expression of WNT3a, GSK3β, and MMP9 in SW480 cells significantly increased, whereas the expression of p-β-catenin decreased (
