2.MRI diagnosis of the meniscus and ligament injury of the knee joint

Objective To evaluate the clinical significance and value of MRI for the diagnosis of the meniscus and ligament injury of the knee joint. Methods 27 patients were put under MRI examinations with 1.5 T unit(Magnetom Vision plus, SIEMENS)and confirmed by surgery or arthroscopy. In all patients, the sagittal and coronal multiplane scan was performed with 4mm section thickness. The posterior cruciate ligament (PCL) angle and PCL curvature index were measured in 8 patients with anterior cruciate ligament (ACL) tears and in 22 patients with ACL normal. The results of measuring were statistically analyzed. Results The accurate diagnosis of MRI was 18 of 24 patients (62.07%) and 24 of 31 cases with meniscus injury (77.42%). On MRI, the signs of meniscus injury were as follows: firstly, the linear high signal intensity inside meniscus was abnormal. Secondly, the size of meniscus diminished and the inner edge was pointless. The third, the triangle configuration of meniscus disappeared.The fourth, the lower or superior margin of meniscus appeared as wavelike configuration. The fifth,the distance from outer side of meniscus to articular capsule increased. Only 5 in 9 patients with ACL tears were exactly diagnosed and the accuracy ratio was 55.56%. Objective To investigate the efficacy of renal transplantation from living related renal donor.Methods A retrospective analysis of 26 cases of renal transplantation from living related renal donor and 84 cases cadaveric renal transplantation was conducted.Results All the operations in these two groups underwent smoothly.The rate of patient/renal survival was 96.15%/96.15%,96.15%/96.15%,96.15%/92.31% respectively in the group of living related renal transplantation,and 96.43%/95.23%,85.71%/80.95%,65.48%/60.71% respectively in the group of cadaveric renal transplantation 1,3,5 years after the operation.The time of cold blood-deficiency and the time of warm blood-deficiency in the living related renal transplantation group were both shorter than those of the cadaveric renal transplantation group(P
5.Research on PHA-CIK cells' immunophenotype and cytotoxicity

Objective:To research PHA-CIK cells' phenotype and cytotoxicity. Methods: Using phytohemagglutinin(PHA)to stimulate peripheral blood mononuclear cells(PBMNCs) for 24 h,then cultured like incubating cytokine induced killers by traditional method. On 15th day examined its immunophenotype by flow cytometry and using MTT method to evaluate cytotoxicity. Results: The ratio of CD3+,CD8+, CD3+ CD8+ and CD3+ CD56+ cells were high.PHA-CK cells cytotoxicity was strong in some degree.Conclusion:PHA-CIK cells had strong cytotoxicity.The result provides an experimental basis for biotherapy.
6.Contrastive Research the Diagnosis by MRI and MRCP with Operative Findings in Hepatic Hilar Cholangiocarcinoma

Objective To evaluate the role of MR imaging and MR cholangiopancreatography(MRCP) for the diagnosis of hepatic hilar cholangiocarcinoma and assessment of tumor resectability.Methods MR imagings were analyzed retrospectively and compared with surgical result in 20 patients with hepatic hilar cholangiocarcinoma.Results The tumor masses were found in the hilar regions on the MR images for 18 patients. It was found that only dilatation of the intrahepatic biliary tree and thickness of the bile duct wall for the other 2 patients. According to the Bismuth classifying, classification of MR was: typeⅠ (n=3), typeⅡ(n=4), type Ⅲ(n=6),type Ⅳ(n=7). Compared with the operative findings, the accurate ratio of classification was 80%, sensitivity specificity and coincidence was 75%, 75%,80% for the assessment of tumor resectablility by MR. Conclusion MRCP combined with MRI is very useful for diagnosis and classification of the hepatic hilar cholangiocarcinoma and assessment of tumor respectability.
7.Establishment of breast cancer cell sublines with different potential of metastasis and their biological characteristics

Objective To investigate the validity of selecting subclones of human breast cancer cells with high-and low-metastatic potentiality,based on their different ability of penetrating the matrigel in vitro,and analyze their biological characteristics.Methods Continuous in vitro invasion assays in matrigel were applied to screen two subclones of human breast cancer cell line MDA-MB-435s,characterized by high-and low-metastatic potentiality.Cell growth curve assay,invasion assay,adhesion assay and angiopoiesis-promoting assay were conducted to explore the different biological properties of these two subclone cells.Results Two subclones of MDA-MB-435s,of high-and low-metastatic potentiality respectively,were obtained through continuous in vitro invasion assay,manifesting significant difference in growth rate,invasion ability,adhesion ability and angiopoiesis-promoting ability.Conclusion It is feasible to obtain subclones of human breast cancer cells of high-and low-metastatic potentiality by utilizing their difference in penetrating the matrigel in vitro.
8.Corneal biomechanical properties in different anterior chamber angle of glaucoma

Background Recent studies have demonstrated that glaucoma progression had great relationship with corneal biomechanical properties.However,there are not enough studies about corneal biomechanical properties in different types of glaucoma.Objective This study was to evaluate the corneal biomechanical properties in chronic primary angle-closure glaucoma (CPACG) and primary open angle glaucoma (POAG).Methods Prospective observational study was adopted.Sixty-eight cases (68 eyes) with CPACG and 69 POAG patients (69 eyes) from Zhongshan Ophthalmic Center were investigated by a novel technique named Corvis-ST which can measure corneal deformation.The Cronbach's αt coefficient and intraclass correlation coefficient (ICC) were used to evaluate the repeatability of the parameters from Corvis-ST.The differences of corneal biomechanical properties between POAG and CPACG were evaluated by independent-samples t test.Multiple regression analysis was used to assess the association between parameters of deformation response and clinical factors.This study protocol was approved by Ethic Committee of Zhongshan Ophthalmic Center and complied with Helsinki Declaration.Written informed consent was obtained from each patient prior to any medical examination.Results There were no statistic differences about age,sex and intraocular pressure (IOP) between CPACG and POAG patients (all at P>0.05).However,differences existed in the number of anti-glaucoma medications (t =-2.388,P =0.020).Measurements of central corneal thickness (CCT),deformation amplitude (DA),and peak distance (PD) demonstrated excellent repeatability (ICC≥ 0.8).There were no statistic differences about CCT,first applanation time/length/velocity (A1 T,A1 L,A1V),second applanation time/length/velocity (A2T,A2L,A2V),DA between CPACG and POAG patients (all at P> 0.05).The highest concavity time was shorter (t =2.920,P =0.005),PD was bigger (t =-2.453,P =0.017),and central curvature radius (CCR) was smaller (t =1.997,P =0.050) in POAG than those in the CPACG patients.DA,A1V,A2T and PD were negatively associated with IOP (r =-0.709,-0.531,-0.645,-0.554;all at P<0.001).However,A1T and A2V were positively associated with IOP (r=0.744,0.546;both at P<0.001).And CCR was positively associated with CCT (r =0.181,P =0.039).Conclusions It is useable to evaluate corneal biomechanical properties of glaucoma by Corvis ST.Under the same level of IOP,the corneal concavity of POAG is easier than that of CPACG,which indicates that cornea in POAG patients is more deformable.
9.Surgical treatment for 310 elderly patients with non-small cell lung cancer

Background and purpose:Life expectancy has continuously improved in recently years with the development of medical level. In 2012, the amount of people who were above 60 years old has reached 810 million and account for 11% of worldwide population. The worldwide population shift towards older ages will inevitably lead to more elderly patients being diagnosed with non-small cell lung cancer (NSCLC). It still remains controversial whether sublobar resection is effective in such cases. In order to solve this question, we need to understand the clinical characteristics of these tumors. Methods:From 2006 to 2012, a total of 310 patients with NSCLC who were above 65 years old underwent surgical resection in Department of Thoracic Surgery, the First Afifliated Hospital of China Medical University and the Second Department of Thoracic Surgical Oncology, Jilin Province Tumor Hospital. The clinical data were retrospectively analyzed in sex, stage, histology, smoking status, smoking amount, drinking status, surgical approaches, multimodality therapy and overall survival. Survival was analyzed by Kaplan–Meier method and log-rank test. Results:There were 256 (82.6%) elderly patients who underwent standard lobectomies and 54 (17.4%) patients who underwent sublobar resections. The overall 5-year survival rate was 52.9%. Patients with different surgical approaches (lobectomy and sublobar resection) had nearly the same 5-year survival rates (60.5%vs 60.8%, P=0.381). The prognosis were signiifcantly inlfuenced by gender (P=0.024), stage of disease (P=0.028), smoking status (P=0.034) and smoking amount (P=0.028). The multivariate Cox proportional hazards analysis revealed that the smoking status was associated with the hazard ratio of 1.660 (1.058-2.606;P=0.028). Conclusion:In elderly NSCLC patients, sublobar resection is considered to be an appropriate treatment in comparison with lobectomy, as this procedure provides an equivalent long-term survival. The survival of elderly patients with lung cancer is closely related to the smoking status.
10.A flow cytometric method for the detection of alkaline phosphatase expression on the membrane of neutrophils

Objective To establish a flow cytometric method for the detection of alkaline phosphatase (ALP) expression on the membrane of neutrophils (mNAP).Methods EDTA-K2 anticoagulant venous bloods were collected.Expression of mNAP in peripheral blood was measured by flow cytometry using a phycoerythrin (PE)-labeled anti-ALP monoclonal antibody.BD QuantiBRITE PE was used to generate a calibration curve for PE fluorescence and ratios of PE to anti-ALP antibody and detect the bound ALP antibodies per cell (antibodies bound per cell,AB/c).Preanalytical handling including anticoagulants (EDTA-K2,citrate,and heparin),storage temperature,storage time,and plasma ALP were optimized and measured the precision.The expression levels of mNAP from 481 healthy controls were measured to establish a clinical reference range.The mNAP levels of 84 patients with severe infection and local infection,39 patients with virus infection were determined by this method.Results For preanalytical handling,application of PBS washing can effectively eliminate the interference of plasma ALP.The mNAP levels were not influenced by different anticoagulants and storage conditions (stored for 12 h either at room temperature or 4 ℃).This method had preferable reproducibility (CV in batch were 2.01%-3.33%,average 2.67% ; CV between batch were 5.80%-6.00%,average 5.90%).The median (quartiles) of mNAP in health controls were 1 758 (1 378-2 310) AB / c for men and 1 897 (1 369-3 249) AB / c for women.There was no significant difference between genders (U =27 140,P =0.243 8).The clinical reference ranges (2.5 percentile to 97.5 percentile) of mALP was 920.5-3 493.0AB / c.The expression levels of mNAP of patients with bacterial infections (13 532,9 756-16 869 AB / c) were significantly higher than those of patients with virus infection(1 143,536-2 012 AB / c) and healthy controls (1879,1399-2497 AB / c) (H=221.5,P<0.01).Conclusion BD QuantiBRITE PE kit can be used to standardize flow cytometer settings and quantitatively detect molecules per cell.The flow cytometric method for detection of mNAP has important clinical application for differentiating bacterial and virus infection.
