2.Content Determination of Emodin and Chrysophanol in Cigu Xiaozhi Pill

Objective To establish a HPLC method for the determination of the content of emodin and chrysophanol in Cigu Xiaozhi pill.Methods The chromatographic procedure was carried out in Symmetry C18(4.6 mm?150 mm,5.0 ?m) column with methanol-0.1% phosphoric acid(85∶15) as the mobile phase,flow rate was 0.8 mL/min,column temperature was 20 ?C,and the detection wavelength was 423 nm.Results Emodin and chrysophanol were well separated with other components.The standard curves of emodin and chrysophanol showed linearity in the range of 0.25~0.75 ?g.The average recoveries were 101.35%(RSD=2.26%) and 98.66%(RSD=1.74%) respectively.Conclusion The method is sensitive,simple and accurate,and can be used for the determination of emodin and chrysophanol in Cigu Xiaozhi pill.
3.Protective and therapeutic effects of Sagittara Sagittifolia L. Xiao-Zhi Pills on nonalcoholic steatohepatitis in rats

Objective To observe the preventive and therapeutic effects of Sagittara Sagittifolia L. Xiao-Zhi Pills on nonalcoholic steatohepatitis (NASH) in rats. Methods Rat NASH models were established and the rats were divided randomly into five groups: the normal group, model group, high-dosage group and low-dosage groups of the treatment, and Dongbao Gantai positive drug group. Histopathological changes of the liver were observed. The levels of MDA, superoxide dismutase (SOD), and NO (nitric oxide) in the liver homogenate were determined. Expressions of cyclooxygenase-2 (cox-2) and inducible nitric oxide synthase (iNOS) in the liver were determined with immunohistochemical technique. Results In model group, extensive adipose degeneration and inflammatory cell infiltration were found in the liver. Compared with those in normal group, SOD activities in model group were obviously decreased (P<0.01), the contents of MDA and NO were obviously increased (P<0.01), and the expressions of cox-2 and iNOS proteins were obviously increased (P<0.01). Compared with those in model group, the activities of SOD in treatment groups were obviously increased (P<0.01), the contents of MDA and NO were obviously decreased (P<0.01), and the expressions of COX-2 and iNOS proteins were obviously decreased (P<0.01). The effect of high-dosage group was more obvious than that of Dongbao Gantai group (P<0.05). Conclusion Sagittara Sagittifolia L. Xiao-Zhi Pills can effectively prevent and treat nonalcoholic steatohepatitis in rats, and its mechanism may be associated with ameliorating hepatocellular steatosis, removing the free radicals and enhancing the anti-oxidation and anti-inflammation capability.
4.Clinical analysis of parapharyngeal space tumors—Report of 24 cases

Twenty-four patients with parapharyngeal space tumors were treated from 1964 to 1999.There were 11 males and 13 females,ranging in age from 10 to 71 years old.Among 24 patients, benign tumors were 17(70.8%),malignant tumors 7(29.2%).The diagnosis of these tumors were biopsy and CT scaning .Pathologic results showed that the original tumors were salivary gland tissue in 11(45.8%),cranial nerve in 7(29.2%),vascular tissue in 2(8.3%)and the unidentified in 4(16.7%).The surgical approaches for these tumors were neck side incision in 19,through mouth in 5.Results showed that 4 malignant tumors were died in one year,3 cases did not clear;the prognosis of 17 benign tumors were well.
5.Advances in the Modulation of Quercetin on Drug Metabolizing Enzymes

Objective: To summarize the recent research progress in the influence of quercetin on drug metabolizing enzymes. Methods:By referring to the relevant literatures at home and abroad in recent years, the paper summarized, analyzed and concluded the the influence of quercetin on drug metabolizing enzymes. Results: Quercetin could modulate the phase Ⅰmetabolic enzyme cyto-chrome P450 ( CYP) and the phase Ⅱ metabolic enzymes uridine diphosphate - glucuronosyltransferase enzyme ( UGTs) , sulfotrans-ferase ( SULTs) and glutathione S-transferase ( GSTs) to influence the in vitro and in vivo metabolism of a lot of drugs. Meanwhile, the modulation of quercetin on the metabolic enzymes demanded the participation of various nuclear receptors. Conclusion:Quercetin shows the inhibitory or inducing effect on a variety of drug-metabolizing enzymes, therefore, it can interact with other drugs.

8.The effect of PAT on PCNA of human colon carcinoma implants in nude mice

Objective To study the potential therapeutic effect of potassium antimonyl tartrate (PAT) on human colon cancer in transplanted tumor nude mice models. MethodsSixty transplanted animal models were constructed with colon cancer cell line SW480 injected in nude mice. Nude mice were then random divided into 4 groups ( n = 15) :Normal saline group, 5-Fu group and different dose of PAT groups [ (20 mg/( Kg · d) ,40 mg/( Kg · d) ]. The volume of mass was measured every 3 days. After final-administration for 24 hours, immunohistochemical staining was used to detect the expression of PCNA in colon cancer cells. ResultsAfter the use of PAT, the growth of mass slowed down. PCNA levels [ (63. 63 ±8. 88)% ,(59. 13 ±6. 15)% ,(33. 38 ± 12. 76)% ] in SW480 cells was reduced by PAT( P <0. 05, P <0. 01 ). ConclusionPAT potentially inhibited the growth of colon cell lines and induced apoptosis of SW480 colon cancer cells.
9.Application of retrograde puncture in intravenous chemotherapy for patients with galactophore cancer

Objective To study the application of retrograde puncture in intravenous transfusion for patients with galactophore cancer. Methods Self compare was adopted,30 cases with galactophore cancer which have put up four cycles intravenous chemotherapy in all 120 intravenous punctures were divided into observation group and control group. Observation group adopted retrograde puncture; control group were given traditional operation. Smooth transfu-sion and the rate of phlebitis and comfort in intravenous chemotherapy in two groups were compared. Results There are significandy different in two groups, observation group is higher in smooth transfusion and comfort (P < 0.05), low-er the rate of phlebitis (P < 0.01) than that of control group. Conclusion Retrograde puncture had good effect in in-travenoas chemotherapy of galactophore cancer,it may be generalized.
10.Effects of Hydrophilic Polymers on the Stability of Self-microemulsifying Drug Delivery Systems

OBJECTIVE:To investigate the effects of hydrophilic polymers on the stability of self-microemulsifying drug deliv-ery systems (SMEDDS). METHODS:Taking felodipine (FDP) as model drug,the content of FDP was determined by HPLC method. The effects of pure water,0.5% Kollidon VA64,HPMC E5,HPMC K100LV,HPMC K4M,PVP K30 solution,while 0.1%,0.5% and 1.0% HPMC E5 and Kollidon VA64 on residual content of dissolved FDP were determined in SMEDDS. RE-SULTS:The residual contents of dissolved FDP in SMEDDS placed in Kollidon VA64,HPMC E5,HPMC K100LV,PVP K30, HPMC K4M and pure water for 1 h were 92.7%,63.6%,50.2%,46.2%,36.0%and 24.0%,respectively. The order of maintain-ing the supersaturation state was Kollidon VA64>HPMC E5>HPMC K100LV>PVP K30>HPMC K4M>pure water. The residu-al contents of dissolved FDP in SMEDDS placed in 0.1%,0.5%,1% Kollidon VA64 and HPMC E5 and pure water for 1 h were 93.2%,95.1%,96.0% and 48.4%,62.1%,75.1%. CONCLUSIONS:Kollidon VA64 and HPMC E5 can significantly inhibit drug release in SMEDDS and be used as stabilizer of SMEDDS,wherein Kollidon VA64 was better.
