1.Detection and analysis of different genotypes of cervical cancer-associated miR-17-5p target loci

Objective To detect the different genotypes of cervical cancer-associated miR-17-5p target loci,and to analyze the influence of target site polymorphism on the risk of cervical cancer.Methods A total of 250 cervical cancer patients and 250 healthy females were selected in Hanzhong Central Hospital from June 2014 to June 2016.The blood samples were collected from the subjects.The genotypes of the three target loci rs3741216,rs217727 and rs2839702 in miR-17-5p were determined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP).The associativity between genetic polymorphisms and the risk of cervical cancer was calculated by SPSS 21.0 online software.Results The three candidate SNP loci fitted the Hardy-Weinberg equilibrium law.In the allele model,the rs217727 locus on the H19 gene significantly increased the risk of cervical cancer [OR =1.55,95% CI(1.21,2.32),P =0.001].Genetic model analysis showed that rs217727 locus in the best model (dominant model),the risk of cervical cancer in the individuals carrying A/G and A/A genotypes increased significantly,and the risk of cervical cancer in the individuals carrying A/G and A/A genotypes was 1.65 times higher than that in the individuals carrying G/G genotype [OR =1.65,95% CI (1.14,2.28),P =0.006].Conclusion The polymorphism of miR-17-5p target site rs217727 is associated with the risk of cervical cancer,the risk of cervical cancer in the individuals carrying A/G and A/A genotypes is significantly increased.
2.Effect and safety analysis of proximal femur locking compression plate for treatment of intertrochanteric frac-ture in 52 cases of old patients

Objective To investigate the effect and safety of proximal femur locking compression plate for treatment of intertrochanteric fracture in old patients. Methods Clinical data and visit records of 98 cases of intertrochanteric fracture old patients were analysed retro-spectively. According to the treatment of operative methods, these patients were divided into two groups: 46 patients in the control group were given the dynamic hip screw fixation; the other 52 cases in the observation group were given the proximal femur locking compression plate . Compared the therapeutic effect and safety of the two groups. Results The short-term and long-term therapeutic effect of observation group were significantly better than the control group,and the safety of observation group was higher than that of the control group,the differ-ences were statistically significant (P<0. 05). Conclusion Proximal femur locking compression plates is an effective surgical treatment for old patients with intertrochanteric fracture. It can significantly improve the function of hip joint and reduce the risk of postoperative complica-tions. As it is of significant effect and high safety, it is worthy of clinical popularization.
3.Association between the Level of LDH in Gastric Cancer Patient’s Serum, the Relative Expression of LDH in Tissues and the Patient’s Prognosis

Objective To explore the association between the level of lactate dehydrogenase (LDH)in gastric cancer patients’ serum or the relative expression of LDH in their tissues and the patients’prognosis.Methods 60 specimens of gastric canc-er patients who confirmed by pathological diagnosis were collected from 2012 to 2013,include the serum specimens and tis-sue specimens.Their serum level of LDH were detected,and the relative expressions level of LDH in the sample of normal tissues,gastric cancer tissues and metastatic lymph nodes were quantify by real-time fluorescence quantitative reverse tran-scription-PCR (Realtime-PCR).Finally,the relationship between the expression of LDH and clinical pathologic features were analyzed by independent t-test or anova statistics.Results The serum level of LDH was highly increased than control group (340.89±10.67 IU/ml,t=24.7,P<0.01);the relative gene expression of LDH in normal tissue,human gastric cancer and metastatic lymph node were 1.0,3.39 and 2.35.The result suggests the serum level of LDH were associated with pTNM stage and lymph node metastases (t=5.2,4.8,P<0.01).The relative gene expression of LDH in gastric canc-er tissues were associated with tumor sizes,pTNM stage and lymph node metastases (t=18.2,15.3,P<0.01 and F=7.2, P<0.01).Conclusion The serum LDH level and the expression of LDH in cancer tissue were significantly increased,and their expression were correlated with the patient’s prognosis.The serum level of LDH and the expression of LDH in gastric cancer tissue may be potential indicator to evaluated the prognosis of the patient with gastric cancer.
4.Effect of Pingwei Xiaodao Capsule on Gastric Motility of Functional Dyspepsia Model Rats

Objective To observe the effect and mechanism of Pingwei Xiaodao Capsule (Capsule for functional dyspepsia) on the gastric motility of functional dyspepsia (FD) model rats.Methods Totally 60 wistar rats were randomized into a blank control group (normal group),model group,domperidone group,Pingwei Xiaodao Capsule large,medium,and small dosage groups.Except for the normal group,the rats of all other groups were irritated by clamping tails for making FD models.Then,the treatment was given for 7 days.The comparative observation was carried out on the gastric interdigestive myoelectric complex (IMC),slow wave frequency variation coefficient,abnormal rhythm index,and gastric emptying rate of all groups.Results Compared with the model group,the IMC period of domperidone group and all Pingwei Xiaodao Capsule groups was shortened,phase Ⅲ prolonged,incidence of phase Ⅲ increased(P
5.Primary experience of intraperitoneal laparoscopic partial nephrectomy with zero ischemia time

Objective To evaluate the feasibility and clinical efficacy of intraperitoneal laparoscopic partial nephrectomy for T1a peripheral renal neoplasms. Methods Intraperitoneal laparoscopic partial nephrectomy was performed without renal artery occlusion for T1a peripheral peripheral renal neoplasms. The operative time, bleeding volume and complications were observed and the clinical experience was summarized. Results From October 2014 to January 2017 ,there were 10 patients:7 males and 3 females. All patients had T1a peripheral renal tumors. 10 patients underwent operation successfully ,of which 1 case developed temporarily blocked renal artery in the surgery due to hemorrhage. There was no referral during surgeries. The operative duration was 108 to 210 min,with a median of 135 min. The estimated blood loss was 100 to 750 mL,with a median of 320 mL. Followed up duration was 2 to 24 months (median 12 months),there were not postoperative renal secondary bleeding , leakage and other complications. No recurrence of tumor was found. Conclusion It is feasible and safe to exercise intraperitoneal laparoscopic partial nephrectomy without renal artery occlusion in the treatment of T 1a peripheral renal tumors,which can protect renal function to the greatest extent.
6.Behavior Observation on Learning and Memory Ability of Brain Aging Mice Intervened by Exercise and Diet

Objective To observe effects for exercise and diet on learning and memory ability in mice with encephalon aging induced by D-galactose(D-gal).Methods The model of mice with encephalon aging was made by D-gal.The learning and memory ability of mice was determined by Morris water maze.Results There was significant difference between high fat feed encephalon aging group and restrict food on normal feed encephalon aging group,normal feed and exercise encephalon aging group,high fat feed and exercise encephalon aging group,high fat feed normal group,normal feed and exercise group,normal feed group(all P<0.05).In spatial probe test,there was significant difference between restrict food on normal feed encephalon aging group and normal feed encephalon aging group(all P<0.05).Conclusion Exercise and restrict food can improve the learning and memory function in the mice;feed with high fat can promote encephalon aging.
7.Diagnosis and treatment of vascular complications of external iliac arteries after kidney transplantation:a report of 6 cases

Objective: To explore the characteristics of external iliac artery vascular complications atfer renal transplantation and the diagnosis and treatment. Methods: We reviewed the clinical data of 6 patients with of external iliac artery vascular complications atfer renal transplantation from more than 2000 renal transplantation patients in the Transplantation Center of the Third Xiangya Hospital of Central South University from 2001 to 2013, and analyzed the clinical characteristics, diagnosis and treatment. Results: hTe renal allogratf was removed in 5 of the 6 patients due to repeated external iliac arteryhemorrhage: 2 patients were replaced the external iliac artery with reversed autogenous great saphenous vein, 2 patients underwent the bilateral femoral artery bypass surgery, and 1 was repaired the external iliac artery directly. The other 1 was resected the renal allograft and the involved external iliac arteries due to fungal mass in the external iliac artery. Among the 6 patients, except 1 patient died atfer the surgery of the repair of the external iliac artery, the other 5 are all alive. Conclusion: Vascular replacement and artery bypass are effective methods for patients with external iliac artery vascular complications atfer kidney transplantation.
8.Development of a clinical nomogram for predicting bladder outlet obstruction in male LUTS/BPH

Objective To develop a clinical nomogram for predicting the probability of bladder outlet obstruction (BOO) in male LUTS/BPH using the most common and noninvasive parameters in clinical practice , with the hope of detecting BOO individually and precisely. Methods Retrospectively analyze the outpatients and inpatients of male LUTS/BPH from November 2003 to November 2015 in Guangzhou First People′s Hospital. Collect the Pressure-flow study parameters and other clinical parameters including Qmax , PV, TZV, TZI, PSA, and PVR. Find out the best independent predictors on the diagnosis of BOO and develop the nomogram for pre-dicting BOO. Results The data from 1 599 patients were analyzed. The areas under the ROC curve (AUCs)of PV, TZV, TZI, PSA, Qmax, and PVR were 0.803, 0.807, 0.698, 0.775, 0.742, and 0.641, respectively. Qmax, PV, and PSA were selected as the best clinical parameters to predict BOO. The Logistic regression equa-tion is Log(p)=0.332 4 - 0.201 8*Qmax + 0.026 6*PV + 1.135 1*PSA. Finally, a nomogram model was developed by R statistical software. This nomogram showed a concordance index of 0.854 according to the inter-nal validation of the model. Conclusions The clinical nomogram presented a high accuracy (85.4%) in de-tecting BOO, which would help predicting BOO in male LUTS/BPH noninvasively, individually, accurately, and providing valuable reference and guidance in clinical decision.
9.Application of molecular absorbent recycling system in auto liver transplantation

Objective To investigate the application of molecular absorbent recycling system (MARS) in auto liver transplantation (ALT).Methods The clinical data of 46 patients who underwent ALT in Zhongnan Hospital of Wuhan University from September 2014 to August 2015 were retrospectively studied.The patients were randomly divided into the MARS group (n =30) and the hemodialysis group (n =16).In the MARS group,20 patients were male and 10 were female with an average age of (15 ±67) years,and the median age was 46.In the hemodialysis group,8 were male and 8 were female with an average age of (22 ± 54) years.A heparinized left iliac vein was linked to a centrifugal pump of a MARS and an output tube was linked to a left jugular vein.Before the anhepatic phase,physical pretreatment was carried out by ligating the porta hepatis and inferior vena cava (IVC) to adapt the patient to the anhepatic phase.Subsequently,a bypass was established followed by ligation of the IVC.The duration of operation,length of stay in ICU after operation,one-year survival rate and hepatorenal function were studied.Results The duration of operation in the MRAS group and in the hemodialysis group was not significantly different (P >0.05).The length of stay in ICU after operation in the MARS group was (12.0 ±3.0) d,which was significantly shorter than that of the hemodialysis group (20.0 ± 2.0) d (P < 0.05).The one-year survival rate in the MARS group was (100%),which was significantly higher than that of the hemodialysis group (93.8%,P <0.05).Conclusion The application of MARS in ALT shortened the length of stay in hospital and improved one-year survival rate,resulting in better outcomes than the conventional method.
10.The efficacy and safety of intradetrusor injection of botulinum toxin type A in treatment of ketamine-related cystitis

Objective To evaluate the efficacy and safety of intradetrusor injection of botulinum toxin type A in the treatment of ketamine-related cystitis.Methods A retrospective analysis of clinical data of 36 ketamine-related cystitis patients with intradetrusor injection of botulinum toxin type A treatment in our hospital during August 2010 to December 2015 was conducted, including 31 males and 5 females with a mean age of 25.5 years.All patients had failed to conventional treatment options including cessation of ketamine, antibiotics, M-blockers.At the time of the first injection, patients were injected with 200 U botulinum toxin type A diluted in 15 ml of 0.9% saline into the detrusor muscle at 30 sites, sparing the trigone, under cystoscopic guidance.3-d voiding diary, interstitial cystitis symptom index (ICSI),interstitial cystitis problem index (ICPI), pelvic pain and urinary frequency/urgency symptom score (PUF) were recorded to evaluate the efficacy.The treatment-related complications were recorded.When the efficacy of botulinum toxin type A decreased and the patient's symptoms returned to baseline before treatment, the patient received repeated injections of botulinum toxin type A with the same dose and method as the first injection.Results Thirty-six patients with ketamine-related cystitis were treated with intradetrusor injection of botulinum toxin type A.Sixteen patients received two injection treatments and two patients received three injection treatments.During the follow-up, 3-d urinary diaries, ICSI, ICPI, and PUF showed a significant improvement in outcome at 4 weeks after the first injection.The efficacy of the second and third injection treatment was also remarkable.Three patients developed urinary tract infection after the first injection, and two patients developed urinary tract infection after the second injection.Mild hematuria occurred in 15 patients after the first injection, and mild hematuria occurred in 7 patients after the second injection, which was improved in 1 to 2 days.All patients did not appear acute urinary retention and other adverse drug reactions.Conclusions Intradetrusor injection of botulinum toxin type A could be a safe and effective method for the treatment of ketamine-related cystitis.Repeated injection therapy is still safe and effective.
