1.The Image Analysis of Two-Dimensional Gel Electrophoresis

Proteome research has become a new hot spot in the post-genome era. High-resolution two-dimensional gel electrophoresis (2-DE), which provides the most comprehensive analysis system of the whole proteome, was highly improved in recent years. With the development of computer techniques, the powerful and user-friendly image analysis systems appeared to help high-throughput, large-scale proteomic studies. Using new generation two-dimensional image analysis software, ImageMaster 2D Elite, the 2D gels of proteins extracted from cultured Schwann’s cells were processed. The analysis procedure, including image acquirement, spot detection, match, background subtraction, pI/Mr calibration, analysis results report and database query, were reported and discussed.
2.Association between the Level of LDH in Gastric Cancer Patient’s Serum, the Relative Expression of LDH in Tissues and the Patient’s Prognosis

Objective To explore the association between the level of lactate dehydrogenase (LDH)in gastric cancer patients’ serum or the relative expression of LDH in their tissues and the patients’prognosis.Methods 60 specimens of gastric canc-er patients who confirmed by pathological diagnosis were collected from 2012 to 2013,include the serum specimens and tis-sue specimens.Their serum level of LDH were detected,and the relative expressions level of LDH in the sample of normal tissues,gastric cancer tissues and metastatic lymph nodes were quantify by real-time fluorescence quantitative reverse tran-scription-PCR (Realtime-PCR).Finally,the relationship between the expression of LDH and clinical pathologic features were analyzed by independent t-test or anova statistics.Results The serum level of LDH was highly increased than control group (340.89±10.67 IU/ml,t=24.7,P<0.01);the relative gene expression of LDH in normal tissue,human gastric cancer and metastatic lymph node were 1.0,3.39 and 2.35.The result suggests the serum level of LDH were associated with pTNM stage and lymph node metastases (t=5.2,4.8,P<0.01).The relative gene expression of LDH in gastric canc-er tissues were associated with tumor sizes,pTNM stage and lymph node metastases (t=18.2,15.3,P<0.01 and F=7.2, P<0.01).Conclusion The serum LDH level and the expression of LDH in cancer tissue were significantly increased,and their expression were correlated with the patient’s prognosis.The serum level of LDH and the expression of LDH in gastric cancer tissue may be potential indicator to evaluated the prognosis of the patient with gastric cancer.
3.Review of HPV Infection Status of the Women in Shantou Region

Objective To review the HPV infection status of the females in Shantou in the past years.Methods 3 664 speci-mens of cervical exfoliated cells of the female patients with gynecological disease from 2013 to 2014 were selected to ampli-fied HPV gene and conducted genetypes detection.The infection rate and various subtypes of HPV were analyzed.All these patients were divided into 5 groups according to their ages and the infection status of each group were also discussed.Results 805 patients were infected with HPV (22%,805/3 664).The single infection type and the mixed infection type were both visible,HPV-16 was the most common single infection type,followed by subtypes HPV-52,43,58 and 56.HPV16+52 was the most common mixed infection type,followed by HPV16+33 and HPV43+52.The infection rates of the each group were differences (χ2 =27.08,P < 0.01),No.1 (> 25),No.2 (25 ~ 34)and No.5 group (< 55)were the high-risk groups of HPV infection.Conclusion The main genotypes of these women whose infection with HPV were HPV-16,52,43,58 and 56 in Shantou region,respectively.The females aged less than 35 years old and more than 55 years old both had high-risk for HPV infection.It has important significance to developed the vaccines which can efficiently prevent HPV-16,52,43,58,56 and reduce the cervical cancer incidence.
4.Effect of sphingosine-1-phosphate receptor 2 on coronary endothelial cells proliferation

Objective To explore the role of sphingosine-1-phosphate receptor (S1PR2) in human coronary artery endothelial cell proliferation in vitro. Methods MTT assay was used to detect cell proliferation in human coronary artery endothelial after treatment of S1P and S1PR2 antagonist JTE-013. Phosphor-ERK and total- ERK level were measured by western blot in endothelial after treatment of S1P and JTE-013. Results 1 μmol/L S1P significantly increased endothelial cells proliferation. S1PR2 antagonist JTE-013 inhibited S1P-induced endothelial cell proliferation in dose-dependent manner. S1PR2 antagonist JTE-013 significantly inhibited S1P-induced phosphor-ERK level in endothelial cells. Conclusion S1PR2 may involve in S1P-induced endothelial cell proliferation through activation of ERK pathway.
5.Treatment of Tumors in the Pineal Region by X- Knife Radiosurgery

Within three years ,86 cases of tumors in the pineal region were treated by X- Knife radiosurgical technique,using the standard axial serial CT scan for stereotactic localization,the target were localized by the BRW coordinate system.Radionics RSA- 3 X- Knife treatment planning system were used to make plan,Philips SL- 75- 14 Linac was used to produce X- ray.After treatment, follow- up ranged 6~ 42 months (meanly 24 months),most of the patients showed improvements within 1` 6 months. The results of this report proved that the X- Knife treatment for tumors in the pineal region might be an effective,economical and reliable method.
6.Comparative study of nasopharyngeal carcinoma staging system between the chinese 2008 and'92 Fuzhou

Objective To carry out a comparative study between the Chinese 2008 and'92 staging system of nasopharyngeal carcinoma (NPC). Methods A total of 777 patients presented with untreated nondisseminated NPC who had received MRI scan of nasopharynx and neck were studied retrospectively. The clinical materials and information of imaging were collected. All patients were restaged according to the Chinese 2008 and 92 staging system of nasopharyngeal carcinoma. Distribution of T, N stage, survival and prognostic value were compared. 513 patients of the 777 cases were treated with conventional radiotherapy,264 cases with intensity modulated radiation therapy. Results The 3-year follow-up rate was 97.6%. The consistency of T stages was 95.0%. T, N and clinical stage distributions in two systems were similar ( Kappa = 0. 93, P = 0. 000; Kappa = 0. 58, P = 0. 000; Kappa = 0. 74, P = 0. 000). Local failure-free survival and disease specific survival were also similar. There was no difference of distant metastasis between N0 and N1(x2 = 1.94,P=0. 164), and a marginal difference between N1 and N2(x2 =3.83,P=0.051) in the Chinese'92 staging system. However, although there was also no difference of distant metastasis-free survival between No and N1a(x2 =0. 07,P =0. 797), ) the difference of overall survival among N1b, N2, and N3 were significant ( x2 = 4. 95, P = 0. 026; x2 = 6. 74, P = 0. 009) in the Chinese 2008 staging system. Conventional radiotherapy or intensity modulated radiation therapy was not a prognostic factor for survival ( x2 = 3.60,P =0. 058). It is reasonable for the Chinese 2008 staging system integrated lymph node characteristics such as laterality, level and extranodal neoplastic spread into the N staging criteria ( x2 = 6. 59, P = 0. 010; x2 =4.78,P=0. 029;x2=9.32,P=0. 002). Conclusions For the Chinese 2008 staging system, it was reasonable to simplify the previous T stage. The N stage showed a better predictive role of distant failure.The role of retropharyngeal lymph node in stage system needs further investigation.

Object To define suitable gathering season of Ginkgo biloba L. leaves. Methods The content of total flavonoids and total terpene lactones in G. biloba leaves were determined by HPLC DAD and HPLC ELSD. Results The content of total flavonoids and total terpene lactones in G. biloba were distinct in different ages of tree and collecting seasons. Conclusion The content of total flavonoids and total terpene lactones in G. biloba are the highest in 2 3 ages of tree. The total flavonoid is the highest in May and total terpene lactone is the highest in September.
8.The use of percutaneous endoscopic gastrostomy in advanced nasopharyngeal carcinoma

Background and purpose:Nasopharyngeal carcinoma usually occurs in people of states of the Southern China. Chemoradiotherapy plays an important role in the therapy of advanced nasopharyngeal carcinoma. However, chemoradiotherapy causes more toxic side effects than radiation therapy alone, which affects the therapy. Now symptomatic treatment and nutrition supports are common ways in the clinic in order to improve the tolerance of patients for the therapy, but with little effect. To evaluate the clinical significance of percutaneous endoscopic gastrostomy (PEG) in advanced nasopharyngeal carcinoma. Methods: From Oct. 2010 to Jun. 2012, a total of 71 patients with advanced nasopharyngeal carcinoma who received PEG before chemoradiotherapy were enrolled. During chemoradiotherapy, gastrostomy diet and nursing were supplied, adverse events, tolerance and nutrition indicators including weight and alcohol of human albumin were detected as well. Results:PEG were performed successfully on 68 patients, but failed in the other 3 patients. Of the 68 patients, 4 have not received concurrent chemotherapy because of liver metastases and liver function damage, 51 of the remaining 64 patients could completely ifnished 3 cycles of concurrent chemotherapy, with the completion rate of concurrent chemotherapy at 79.69%. The weight change ranged from-11.86%to 0.83%, with a mean value of-5.32%±2.99%. Paired-sample t test of human albumin before and after the treatment showed no significant difference (P=0.742). Grade Ⅱ radioactive oral cavity mucositis appeared in 60.29%patients (41/68), and 22.06%patients (15/68) suffered gradeⅢradioactive oral cavity mucositis.Conclusion:For patients with advanced nasopharyngeal carcinoma, preventative PEG improved the tolerance of chemoradiotherapy, reduce the incidence of adverse events. The period of therapy interruption caused by sever adverse event were shortened as well, PEG also increased the completion rate of concurrent chemotherapy. Nutritional status and living quality of patients are improved. In addition, PEG is a safe, economic and simple method.
9.MicroRNA-130a Increases and Predicts Cardiotoxicity during Adjuvant Chemotherapy in Human Epidermal Growth Factor Receptor-2-Positive Breast Cancer

Purpose:
This study aimed to investigate the changes in microRNA-130a (miR-130a) and its correlation with cardiotoxicity during epirubicin/cyclophosphamide followed by docetaxel plus trastuzumab (EC-D+T) adjuvant chemotherapy in human epidermal growth factor receptor-2-positive (HER2+) breast cancer patients.
Methods:
A total of 72 HER2+ breast cancer patients who underwent resection and were scheduled to receive EC-D+T adjuvant therapy were consecutively enrolled. The expression of miR-130a and cardiotoxicity (defined as any of the following situations: 1) absolute decline of left ventricular ejection fraction (LVEF) ≥ 10% and LVEF < 53%; 2) heart failure; 3) acute coronary artery syndromes; and 4) fatal arrhythmia) were assessed every 3 months throughout the 15-month EC-D+T treatment.
Results:
The accumulating cardiotoxicity rate was 12 (16.7%), of which the incidence of heart failure, acute coronary syndrome, life-threatening arrhythmias, ΔLVEF ≥ 10%, and LVEF < 53% was 0 (0.0%), 1 (1.4%), 0 (0.0%), and 12 (16.7%), respectively. Baseline miR-130a expression was negatively correlated with LVEF (%) and positively correlated with cardiac troponin I. The expression of miR-130a gradually increased in both cardiotoxicity and noncardiotoxicity patients during EC-D+T treatment, while the increment of miR-130a was more obvious in cardiotoxicity patients compared with non-cardiotoxicity patients. Further logistic regression and receiver operating characteristic curve analysis indicated that miR-130a was an independent predictive factor for increased cardiotoxicity risk.
Conclusion
MiR-130a increases constantly and predicts high cardiotoxicity risk during ECD+T adjuvant chemotherapy in HER2+ breast cancer patients.
