China Pharmacy 2015;(22):3103-3105
OBJECTIVE:To study the induction effect of paeoniflorin on the apoptosis of lung adenocarcinoma A549 cells. METHODS:MTT method was used to detect the viability of A549 cells 48 h after the cells were cultured with paeoniflorin of 0 (negative control),5,10,20 and 40μmol/L. 24 h after A549 cells were cultured with paeoniflorin at the above-mentioned concen-trations,enzyme-linked immunosorbent assay was employed to detect the viability of cysteinyl aspartate specific proteinase 3(Cas-pase-3),and Western blot to determine the protein expression of Bcl-xl,Bax,nuclear factor κB p65(NF-κB p65)and phosphory-lated nuclear factorκB p65(NF-κB pp65). RESULTS:Compared to the negative control group,A549 cells cultured with paeoniflo-rin of 5-40 μmol/L for 48 h showed lower cell viability;A549 cell cultured with paeoniflorin of 5-40 μmol/L for 24 h showed high-er activity of Caspase-3 and the expression of Bax;A549 cell cultured with paeoniflorin of 10-40 μmol/L for 24 h showed lower protein expression of Bcl-xl and NF-κB pp65. There were statistically difference(P<0.01 or P<0.05). CONCLUSIONS:Paeoni-florin induces the apoptosis of A549 cells by a mechanism which may be related to the activation of NF-κB signaling pathway and regulation related gene expression.

Twenty-four patients with parapharyngeal space tumors were treated from 1964 to 1999.There were 11 males and 13 females, ranging in age from 10 to 71 years old.Among 24 patients, benign tumors were 17(70.8%),malignant tumors 7(29.2%).The diagnosis of these tumors were biopsy and CT scaning .Pathologic results showed that the original tumors were salivary gland tissue in 11(45.8%),cranial nerve in 7(29.2%),vascular tissue in 2(8.3%)and the unidentified in 4(16.7%).The surgical approaches for these tumors were neck side incision in 19,through mouth in 5.Results showed that 4 malignant tumors were died in one year,3 cases did not clear;the prognosis of 17 benign tumors were well.

Comparative investigation of the distribution of galanin-immunoreactive (GAL-ir) nerve fibers in the neural lobe of the human being, monkey, dog, cat and rat was studied immunohistochemically. The density of GAL-ir fibers in the posterior pituitary was highest in the dog and rat, moderate in the monkey and cat; and lowest in the human being. A dense accumulation of positive fibers appeared in the pituitary stalk bordoring the intermediate lobe. The fiber density in the central part of the posterior pituitary was slightly higher than at its periphery in the human and monkey. Moderate amount of fibers were found to spread all over the neural lobe of the dog and rat, with slightly higher density in the posterior part of the dog. In the cat, the highest density of GAL-ir fibers was found along the wall of the recess hypophysis. A slightly increasing fibers or fiber clumps were seen in the peripheral region of posterior pituitary in four species of the animals and human being. Some fibers were found surrounding the blood vessels or located in their walls.

Objective To establish a method for HPLC fingerprint determination of the triterpene acids in poria peel. Methods Ethanol solvent and ultrasound method were used to extract the sample. The chromatographic conditions of RP-HPLC were:acefonitrile-0.5%phosphoric acid(linear gradient elution),the detection wavelength being 242 nm, temperature at 30℃and flow rate being 1.0mL?min-1. Ten batches of poria peel produced in different places were measured. The common model was set up to calculate the correlation coefficient, cos?between every batch vector and the total standard model vector, thus to define the similarity. Results There was much difference in the constituent content of poria peel produced in different places and batches.Conlusion The HPLC fingerprint of triterpene acids is stable and reprodacible, which is useful for standard cultivation and quality evaluation of the crud drug of poria peel.

Object To define suitable gathering season of Ginkgo biloba L. leaves. Methods The content of total flavonoids and total terpene lactones in G. biloba leaves were determined by HPLC DAD and HPLC ELSD. Results The content of total flavonoids and total terpene lactones in G. biloba were distinct in different ages of tree and collecting seasons. Conclusion The content of total flavonoids and total terpene lactones in G. biloba are the highest in 2 3 ages of tree. The total flavonoid is the highest in May and total terpene lactone is the highest in September.

Objective To investigate the impact of whole body vibration training combined with intermittent blood flow restriction on the motor function and community activity in elderly stroke patients with hemiplegia. Methods A total of 80 convalescent hemiplegic patients after stroke who were hospitalized in the Rehabilitation Medicine Department of Yichang Central People's Hospital(Xiling Campus)from October 2021 to December 2023 were included and randomly divided into control group(n=25),vibration group(n=28)and combination group(n=27)using the random number table method. All patients received conventional rehabilitation training,on the basis;the vabration group received whole body vibration training, while the combination group received whole body vibration training combined with intermittent blood flow restriction. Before and after 6 weeks of training, the balance function was evaluated with Berg balance scale(BBS),while the gait function was tested with 6-minute walking test(6MWT)and the Community Balance and Mobility scale(CB&M)was used to assess the community activity ability. The community balance and mobility(CB&M)were evaluated in the first and third month after discharge. Results There was no significant difference in BBS,CB&M scores and 6MWT walking distance between the three groups before training(P>0.05).After 6 weeks of training, the three groups showed significant increases in the BBS,CB&Mscores and the walk distance of 6MWT(P<0.001).Furthermore,compared with the control group, the combination group and the vibration group were significantly beter(P<0.05),and there was no signifi-cant difference between the combination group and the vibration group(P>0.05).At the first and third month of follw-up after discharge, the CB&M scores of the three groups were significantly higher than those of the control group(P<0.05),while there was no significant difference between the CB&M scores of the combination group and the vibration group at the first month of follow-up(P>0.05).But the CB&M scores of the combination group was higher than those of the vibration group at the third month of follow-up,and the difference was statistically signifi-cant(P<0.05).Conclusion On the basis of conventional rehabilitation, whole body vibration training combined with intermittent blood flow restriction can significantly enhance balance function, balance confidence, walking ability and mobility in early stage of community life for elderly stroke patients with hemiplegia, potentially facilitat-ing their return to community life.
