1.Clinicopathalogical Differences of Immunofluorescent Staining Results in Lupus Nephritis Patients with Various Syndrome Patterns

Objective To discuss the differences of immunofluorescence pathological test results in four types of syndromes for lupus nephritis(LN) patients. Methods A retrospective study was carried out in 116 LN patients who had performed renal biopsy. The patients were differentiated into four syndrome patterns of excessive heat-toxicity, liver-kidney yin deficiency, qi-yin deficiency, and spleen-kidney deficiency. The results of immunofluorescence pathological tests were recorded for the analysis of the differences among the four patterns. Results (1)Of 116 LN patients, 59 were classified into spleen-kidney deficiency, 30 into excessive heat-toxicity, 14 into liver-kidney yin deficiency, and 13 into qi-yin deficiency. (2)The positive expression rate of IgA, IgG, IgM, C3, and Fi fluorescen ce antibody differed in the 4 syndrome patterns of LN patients(P<0.05), IgA and C3 had higher positive expression rate, while Fi had the lowest expression rate. (3)The percentage of“full-house” immunofluorescence staining(5 kinds of antibodies simultaneously showing positive immunofluorescence staining) differed in the 4 syndrome patterns of LN patients(P<0.05), and excessive heat-toxicity syndrome had the highest percentage. (4)The differences of positive rate of immunofluorescence staining were insignificant in the 4 syndrome patterns of LN patients(P>0.05). Conclusion The immune complex deposition in LN patients is dominated by IgA and C3 antibodies deposition, and the“full-house” immunofluorescence staining is commonly seen in the syndrome of excessive heat-toxin type. But the results of immunofluorescence pathological tests can not be used as the evidence to tell the differences in the 4 syndrome patterns of LN patients.

3.Prevalence of Depressive Disorder among Inpatients in 40 General Hospitals in Beijing

Objective:Describe the prevalence of depressive disorder and the care-seeking of depressed individuals among inpatients in general hospitals in Beijing.Methods:2 925 inpatients ≥15 years of age from 40 general hospitals (selected by stratified random sampling) were first screened by psychiatric nurses and then psychiatrists used DSM-IV criteria to generate a 'gold standard' diagnosis in subjects screened positive and 10% of those screened negative. Results:Adjusted point prevalence, one-year prevalence, and lifetime prevalence of any type of depressive disorder were 7.04%, 7.47% and 9.96%, respectively; corresponding prevalences of Major Depressive Disorder were 3.83%, 4.33%, and 6.11%, respectively. Of the 123 depressed patients only 12 (9.8%) had ever received treatment. The prevalence was significantly higher in females than in males(5.18%vs 3.59%,RR=1.44 95%CI:1.01~2.05), in patients from Level-2 hospitals than in those from Level-3 hospitals(6.28%vs 3.31%,RR=1.90 95%CI:1.31~2.75), and in patients on internal medicine wards than in those from surgery (6.08%vs3.60%,RR=1.69 95%CI:1.15~2.49)or other wards(6.08%vs 3.01%,RR=2.02 95%CI:1.21~3.37).Conclusions:The prevalence of depressive disorder among inpatients in Beijing's general hospitals is high but very few are diagnosed or treated. Different programs to increase the recognition and treatment of depression in general hospitals need to be developed and regularly monitored.
4.Experimental Study of ex vivo Liposome-mediated Gene Transfer in Orthotopic Rat Left Lung Transplantation

Objective To determine the feasibility of ex vivo liposome-mediated gene transfer ( Lac-Z gene ) to lung isografts and to get expression. Methods An orthotopic rat left lung transplant model was developed with the use of a modification of the “cuff” technique. 18 SD rats were divided into three groups randomly. Gene transfer group ( group 1 ): PSV-?-gal/ liposome ( Lipofectin R○ ) complex was transfered into the donor left lung via the left pulmonary vein route at the time of lung harvesting. Liposome group ( group 2 ) and Control group ( group 3 ) : Grafts underwent the same procedure but received liposome and 0 9% saline solution respectively. Donor lungs were harvested one day after transplantation. The transgene expression of Lac-Z gene was detected by histochemical staining. Results Transgene expression of ?-gal in group 1 was detected in bronchial epithelial cells and alveolar epithelial cells. But no expression of ?-gal was found in groups 2 and 3. Conclusion Ex vivo plasmid vector/liposome complex-mediated gene transfer to lung isografts via the left pulmonary vein route is feasible. This study provides the scientific proof to the feasibility of functional gene therapy of complications such as ischemia-reperfusion injury, rejection and infection after rat lung transplantation.
5.Evaluation of left ventricular global systolic function using velocity vector imaging in patients with coronary heart disease before and after percutaneous coronary intervention therapy

Objective To analyze left ventricular (LV) global systolic function in patients with coronary artery disease(CAD) and their changes after percutaneous coronary intervention(PCI) using velocity vector imaging(VVI). Methods Eighteen healthy adults and twenty two patients with CAD were enrolled in this study. Two-dimensional dynamic images of standard apical four-chambers, two-chambers and parasternal short-axis views at the level of mitral valve and apex were obtained in VVI condition. All patients were examined 1 day before PCI, 1 week, 1 month and 3 months after PCI respectively. LV rotation degree, rotation velocity at baseline and apex were measured using the off-line syngo US workplace software and LV twist, torsion were calculated as global motion condition. Results LV ejection fraction(LVEF) and peak torsion at myocardial infarction group were lower than those at normal control and myocardial ischemia group. After PCI, LVEF were gradually improved in both myocardial infarction group and myocardial ischemia group,while changes of LV apex rotation degree and velocity, peak twist and peak torsion between two groups were opposite: those parameters of the former increased 1 week or 1 month after PCI but decreased 3 months after PCI,while those of the latter decreased 1 week after PCI but increased gradually 1 month and 3 months after PCI. Conclusions LV twist and torsion can be evaluated exactly by VVI.Compared with LVEF, LV twist and torsion can preferably reflect the different characteristic of LV global systolic function after coronary artery recanalization in myocardial infarction group and myocardial ischemia group.
6.Improving ergosterol production from molasses by Saccharomyces cerevisiae.

Ergosterol is an economically important metabolite produced by yeast. To improve the production of ergosterol by Saccharomyces cerevisiae YEH56 (pHXA42) from molasses, a cheap and regenerative material, different strategies were applied. First, Plackett-Burman design and central composite design were applied to screen the significant factors in fermentation medium using ergosterol yield (g/L) as the response value. Ergosterol yield reached 371.56 mg/L by using the optimal fermentation medium in shake-flask culture (total sugar in molasses 40 g/L, KH2PO4 1 g/L, K2HPO4 1.86 g/L, CuSO4 x 5H2O 17.5 mg/L, FeSO4 x 7H2O 13.9 mg/L, MgSO4 x 5H2O 12.3 mg/L, corn steep liquor 10 mL/L), which was increased by 29.5% compared with the initial culture. Second, ergosterol yield was increased by 62.1% using a pH-control strategy in a 5-L bioreactor. Third, ergosterol production was improved further by using molasses feeding strategy. After 38 h fermentation, ergosterol yield reached 1 953.85 mg/L, which was 3.2 times of that in batch fermentation. Meanwhile, ergosterol production rate was increased by 42.7% compared with that in the batch culture.

7.Expression of fibronectin protein levels in sinoatrial node tissues of hearts following electrocution

Objective To study the protein levels of fibronectin (Fn) in the sinoatrial node tissues of hearts caused by electric shock death ,and to provide a new theoretical basis for the diagnosis of electric shock death and the identification of death time.MethodsUsing immunohistochemical technique(S-P method) to detect protein levels of 15 cases of human sinoatrial node following electrocution(human experimental group),15 cases of human sinoatrial node with severe traumatic cerebral injury(human control group),35 cases of rabbit sinoatrial node following electrocution(rabbit experimental group ,divided into7 groups according to different sampling time: 0 h ,1 h ,3 h ,6 h ,12 h,24 h.48 h),and 35 cases of rabbit sinoatrial node with broken neck executed(rabbit control group ,divided int0 7 groups according to different sampling time :0 h,1 h.3 h,6 h.12 h.24 h,48 h).ResultsFn positive expression rate in human experimental group was 100% while it was 6.67% in the human control group,and the differences were statistically significant (P < 0.01 ).The Fn expression can be seen after electric shock immediately in the rabbit experimental group,and there was statistical difference compared with the rabbit control group (P < 0.05).Distribution of Fn expression became more and more widely after 3 hours t0 12 hours in the rabbit experimental group ,and there was significant difference compared with the rabbit control group(P < 0.01).The Fn expression began to decrease 24 hours after electrocution,ancl it still had statistical significance 48 hours after electrocution compared with the rabbit control group (P < 0.05).Conclusion Electric shocks significantly enhanced the expression of Fn protein levels in the sinoatrial node tissues , and optical density of Fn positive expression has a regularity change along with the extended time of death.These results could provide a theoretical reference basis for forensic diagnosis of electrocution and identification of death time.
8.Characteristic changes of sacral foramina in adolescents and its clinical significance

Objective To study the developmental patterns and aging characteristics of anterior sacral foramina and posterior sacral forami-na in adolescents aging from 12 to 18 years old through CT imaging scanning and three-dimensional reconstruction measurement,and to provide the anatomical basis for the diagnosis and treatment of sacral vertebra diseases.Methods The 30 adolescents without any the problems of nerv-ous system were selected to conduct 64-slice spiral CT in the pelvic cavity.The original data were input into three-dimensional reconstruction software Mimics with the form of DICOM,and the relevant indicators were measured and analyzed.Results There was no significant difference in the height,width and distance of inner edge of anterior sacral foramina and the posterior sacral foramina in different gender and sides (P >0.05).The physical development of human increases gradually with age and there were significant differences.Conclusion There is a regular change process in the relevant indicators of anterior sacral foramina and posterior sacral foramina in adolescents.But the clinical appli-cation must be combined with the results of individual imaging,the technology of reverse engineering and the support of rapid prototype manu-facturing so as to meet the satisfactory results of individualized treatment.
9.Determination of Hydroxysafflor Yellow A Content in Drug Pair of Flos Carthami and Semen Persicae in Different Ratios by HPLC

Objective To study the effect of compatibility of Flos Carthami and Semen Persicae in different ratios on the contents of hydroxysafflor yellow A(HSYA) in their water decoction.Methods Reverse-phase HPLC was performed on a Luna C18 column(4.6 mm?150 mm,5?m) at 30℃.The mobile phase consisted of methanol-0.12 mmol?L-1 phosphate solution(28∶72,V/V) at the flow rate of 1.0 mL? min-1.The detector was operated at wavelength of 403nm.Results A good linearity of HSYA was obtained in the range of 4.0~160.0?g?mL-1(Y=25 396X+40 247,r=0.999 7),the average recovery was 94.76% and the RSD(n=6) was 2.93%.The amount of HSYA increased with the ratio of Flos Carthami,but had no correlation with the ratio of Semen Persicae.Conclusion The HPLC method for determination of HSYA is simple,fast and accurate.It can be used for the quality control of Flos Carthami and its compound preparations.Semen Persicae has no effect on the extraction of HSYA.
10.THE PARANODAL REGION AND AXO-GLIAL JUNCTION OF MYELINATED FIBERS

By using electron microscopy, the paranodal region and axo-glial junction were examined in optic nerves of rats aged 14 days. The paranodal region was characterized in longitudinal sections by the sequential termination of the myelin lamellae, beginning proximally with the innermost and ending, at the Ranvier node, with the outermost lamella. The termination of each lamella was accom- panied by a separation of the major dense line of the compact myelin and the consequent formation of a "loop" of glial cytoplasm. Each paranodal loop inde- nted the axonal surface as it became junctionally apposed to the axolemma. The periaxonal extracellular space, 10-20nm in width in the internodal region and reduced at the paranodal junction to approximately 3nm, forming an axo-glial junction, which was thought to be held together by dense structure. The parano- dal junction seems to serve strong adhesion between the apposed axonal and glial membranes. Conduction of the nerve impulses in myelinated axons was saltatory. Axons and sheath cells probably maintain vital communication with one another, presumably at the paranodal junctional complex. This communication was viewed as vital to the stability and maintenance of myelin. We found some clear vesi- cles in axoplasm near the Ranvier node and speculated that there were endocyto- sis and exocytosis in paranodal region. This was a direct morphological evidence supporting metabolic coupling between axons and sheeth cells.
