1.Simultaneous three-trocar laparoscopic cholecystectomy and appendectomy in 92 cases

Objective: To summarize the clinical experiences and effect of laparoscopic cholecystectomy(LC)combined with laparoscopic appendectomy(LA). Methods: The clinical data of 92 cases which successfully underwent three-trocar LC and LA was retrospectively analyzed. Results: All operations were performed successfully without complications. The mean operation time was 70 min. The mean hospital stay was 5 days. No complications occurred in the 85 cases who were followed up in 6 months. Conclusion: The simultaneous approach to LC and LA is a safe and effective treatment.
2.Protective effects of ischemic postconditioning on injuries induced by intestinal ischemia reperfu-sion

Objective To observe the effects and mechanism of ischemic postconditioning(IPo) on alleviating injuries of small intestine and remote organs induced by intestinal ischemia-reperfusion (IR).Methods A total of 48 rabbits were divided randomly into three groups(n =16 in each):the Sham group,the IR group and the IPo group.After 2 hours of reperfusion,arterial and venous blood and tissues of intestine,liver and lung were harvested.The levels of TNF-α,IL-1β,IL-6 and IL-10 in arterial blood and levels of ALT,AST,BUN,Cr,LDH,CK-MB activity and endotoxin in venous blood were tested.MDA, CAT,SOD and MPO levels in serum,small intestine,liver and lung were also measured.HE staining was performed for observation of injured intestinal mucosa.Bacterial translocation rate was calculated after bac-teria culture.Results Compared with the IR group,MDA and MPO levels in serum,small intestine,liver and lung decreased in the IPo group,while CAT and SOD levels increased obviously.Compared with the IR group,ALT,AST,LDH,CK-MB and BUN levels in the venous blood,and TNF-α,IL-1β,IL-6 and endotox-in levels in arterial blood were all decreased.IL-10 increased and the intestinal mucosal injury score de-creased obviously.Conclusion Ischemic post conditioning can decrease the damage of intestinal mucosa, reduce endotoxin translocation,promote activation of anti-inflammatory cytokines,inhibit excessive release of inflammatory mediators,enhance the antioxidant capacity of intestine and remote organs,reduce injuries to the small intestine and remote organs.
3.Effects of CO_2 Pneumoperitoneum on Pancreatic Function in Diabetic Rabbits

Objective To explore the effects of CO2 pneumoperitoneum on pancreatic function in diabetic rabbits. Methods Forty-eight rabbits were divided into 4 groups: control group (the group of N0, n=4), the group of T0 (n=4), the group of T10 (n=20), and the group of T15 (n=20). The animal used in the groups of T0, T10 and T15 was diabetic rabbit, and the pressures of pneumoperitoneum of the three groups were 0 mm Hg, 10 mm Hg and 15 mm Hg respectively.The model of diabetic rabbits were made through intrvenous administration of Allxon. Arterial blood samples were collected before the onset of CO2 pneumoperitoneum, 0, 2, 6, 12 hours after deflation for measuring blood glucose, amylase, insulin and C-peptid. Then the rabbits were sacrificed and their pancreases were removed for measuring SOD activity and MDA content. 4.Laparoscopic cholecystectomy in elderly patients: A clinical analysis of 182 cases

Objective To investigate the prevention and countermeasures of complications following laparoscopic cholecystectomy(LC) in aged patients.Methods The study was based on a retrospective view of clinical date of 182 aged patients(60~85 years old) who had been treated with LC from January 2000 to October 2006 in this hospital.Co-morbidities existed in 87 patients.The operation was performed under general anesthesia and endotracheal intubation,by using 3-port technique in 137 patients and 4-port technique in 45 patients. Results The LC was successfully completed in 174 patients,with an operation time of 30~100 min and a postoperative hospital stay of 3~10 d.Conversions to open surgery were required in 8 patients due to a difficult dissection(6 patients) or a diagnosis of gallbladder carcinoma(2 patients).No death or serious complications occurred.There were 3 patients with port-site infection and 2 patients with pulmonary infection.Follow-up observations in the patients for 3~12 months revealed no abdominal pain,fever,or jaundice.Conclusions Strictly following indications, properly treating the accompanying diseases,and promptly converting to open surgery are key factors to prevent and reduce complications of LC in aged patients.
5.Role and prognostic impact of E-cadherin expression on invasion and metastasis of gastric carcino-ma

Objective To investigate the role and prognostic impact of E-cadherin( E-cad)on in-vasion and metastasis of gastric carcinoma by analyzing E-cadherin expression at DNA,RNA and protein levels. Methods RNA and protein expression of E-cad were tested in 60 cases of gastric carcinoma and adjacent tissues by streptavidin peroxidase( SP )immunohistochemistry and in situ hybridization. DNA methylation of E-cad was tested with methylation-specific polymerase chain reaction( MSP). Results The positive rates of E-cadherin protein and mRNA in gastric carcinoma were significantly lower than those in adjacent tissues. DNA methylation of E-cad negatively correlated with the E-cad protein and RNA expres-sion in gastric carcinoma respectively(r= -0. 355,r = -0. 271). DNA methylation of E-cad negatively correlated with the 5-year survival rate of gastric carcinoma(r = -0. 381,P ﹤0. 01). However,E-cad protein positively correlated with the 5-year survival rate of gastric carcinoma(r=0. 378,P﹤0. 01). Con-clusion The decreased expression of E-cad is one of the most important biological features in gastric car-cinoma. Methylation of E-cad DNA may be an important agent causing low expression of E-cad protein and mRNA and it can be used as an important indicator in predicting invasion and metastasis of gastric cancer.
6.Antioxidases Activities of Normal and Senile Cataractous Lens

A large series of lens were collected from normal and senile cataractous human eyes, and were investigated according to the theories of oxyradical and lipid peroxide (LPO). The findings showed that the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase in the senile cataractous lens were very significantly lower than those in the normal ones, while the content of malondialdehyde (MDA) in the lens of senile cataractous eyes was very significantly higher than that of normal eyes. The results suggest that the oxyradical and LPO might be one of the direct causes leading to senile cataract formation.
7.Effect of Wrist-ankle Acupuncture on Post Traumatic Stress Disorder after Work-related Injury

Objective To study the clinical efficacy of wrist-ankle acupuncture on post-traumatic stress disorder (PTSD) after work-related injury. Methods 62 patients were divided into observation group (n=30) and control group (n=32). The observation group accepted wrist-ankle acupuncture combined with Paroxetine, and the control group accepted Paroxetine only. They were assessed with Clinical Administered PTSD Scale (CAPS), Hamilton Depression Rating Scale for Depression (HAMD), and Hamilton Anxiety Scale (HAMA) before, 6 weeks and 12 weeks after treatment. Results The scores of CAPS, HAMD and HAMA reduced in both groups (P<0.01) after treatment, and the reduction rate was greater in the observation group than in the control group (P<0.05). Conclusion Wrist-ankle acupuncture may further relieve PTSD after work-related injury
8.Preparation and Identification of High Immunogenic A/PR/8/34 Maternal Strain HA Protein for Influenza Virus Classical Reassortment.

Preparation of maternal strain A/PR/8/34 HA antiserum for influenza virus classical reassortment. A/PR/8/34 virus was digested by bromelain after inactivation and purification. 5%-20% sucrose continuous density gradient centrifugation method was used to purify HA protein. SIRD method was used to select the target protein. SDS-PAGE method was used to identified HA protein. High Immunogenic A/PR/8/34 HA protein was successfully prepared and HI titer reached 10240. High purity HA antiserum was identified by SIRD method. The key reagent in the classical reassortment of influenza virus was prepared, and the complete set of technical methods were explored, which laid the foundation for the independent research and development of seasonal influenza vaccine strains of China.

9.Hyperproliferation of human colon cancer cells in co-cultures of rat regenerating hepatocytes

Objective To investigate the stimulated proliferation of colon cancer cells in co-cultures of regenerating hepatocytes. Methods Regenerating hepatocytes(24 hours after partial hepatectomy)were obtained by collagenase perfusion of models of rats undergoing 70% liver resection. To determine whether the ratio of human colon cell line SW480 cells to hepatocytes in co-cultures has influence on their interaction,these cells were cultured in ratios of 1: 101:1, or 10: 1. Proliferation capacity was assessed by the percentage of 3 H-TdR incorporation. Expression of epidermal growth factor receptor(EGFR), insulin-like growth factor1 receptor(IGF-1R)and hepatocyte growth factor receptor(c-met) were analyzed by western blot. Results For co-cultured SW480 and hepatocytes in the ratios of 1: 1 and 1: 10, an increase of disintegrations per minute(dpm) occurred after 72 hours' culture, and lasted at 120 hours' culture(P < 0.05). No difference was found between the group with ratio of 10:1 and control group. Protein levels of EGFR and IGF-1R, but not c-met, were significantly increased between culture of 24 hours and 120 hours; however, no change of these receptors was found in the ratio of 10: 1. Conclusions These results imply that co-culturing human colon cancer cells with regenerating hepatocytes leads to increased expression of EGFR and IGF-1R. We conclude that this effect is probably dependent on paracrine stimulation, by which numerous signals from the hepatocytes contribute to the hyperproliferative state of colon cancer cells via up-regulating the responding receptors.
10.Complete nucleotide sequence of a human echovirus 30 strain KM/A363/09 isolated in Yunnan, China in 2009

Objective To analyze the genetic characteristics of the complete genome of a human echovirus 30 (Echo30) KM/A363/09 strain isolated in Yunnan, China in 2009.Methods Primers specif-ic for Echo30 were designed .The extracted RNA was amplified by using RT-PCR.Seven fragments covering the complete viral genome were sequenced and the complete sequences were aligned with other sequences of enterovirus reference strains downloaded from Genbank . By using Mega5.1, Geneious, RDP3 and SimPlot3.5.1 softwares, the phylogenetic and recombination analysis were carried out .Results The com-plete nucleotide sequence of KM/A363/09 isolate was 7425 bp in length, encoding 2194 amino acids.KM/A363/09 isolate was highly similar with Bastianni prototype strain showing the homology of 81.2%in nucle-otide and 95.8%in amino acid.The eight Echo30 isolates shared 81.2%-88.6%homologies in nucleotide sequences and 95.8%-97.8%in amino acid sequences .Phylogenetic analysis showed that the KM/A363/09 strain belonged to one clade of Echo 30 in China.The genetic recombination of KM/A363/09 isolate was detected in the non-structural region .Conclusion KM/A363/09 isolate belongs to one clade of Echo 30 in China indicating that the evolution of Echo 30 has occurred in China .
