1.An Investigation on the Impression of the Chinese People on Chinese Urban Children

Objective: To study the impression of Chinese people on Chinese urban children, their main source of information about the Chinese people, and the infvuence of age, school, and gender upon their evaluation. Methods: 383 Chinese urban children ranging from 5 to 12 years were interviewed individually or investigated with the questionnaire. Results: Most of the children reported that the Chinese were happy, clean, peaceful, clever, hardworking, kind and nice. However, the urban children's positive impression of the Chinese people was decreased with their growing up. The study also found that children's information on the Chinese people was mainly in order from television, newspapers, books and cartoons, parents, classmates, friends and teachers, and radio, and that the information source became increasingly diversificed with age. No significant difference in gender and between two schools was found. Conclusion: Chinese urban children had a positive impression of Chinese people, and they had a preference and identity for Chinese people . Mass media were the main source of information.
2.Intraperitoneal chemotherapy for malignant ascites of gastric cancer based on the results of in vitro drug sensitivity test of anticancer agents

Objective To estimate the sensitivity of malignant cells from ascites of gastric cancer to anticancer agents by in vitro sensitivity test, and to study its value in individualized intraperitoneal chemotherapy. Methods Forty-seven cases of gastric cancers with malignant ascites were selected. The gastric cancer cells were isolated from the ascites of 19 patients with gastric cancers, and in vitro sensitivity tests to carboplatin, taxol, fluorouracil, cisplatin, adriamycin, hydroxycamptothecine, methotrexate, mitomycin and dacarbazine were performed by ATP bioluminescence assay. One of the most sensitive drugs was applied to intraperitoneal chemotherapy. The rates of the patients with complete remission of ascites and with disappearance of intraperitoneal cancer cells in the sensitivity test group were compared to those of 28 patients in control group who all were treated with intraperitoneal cisplatin. Results In sensitivity test group, the sensitive cases to carboplatin, taxol, fluorouracil, cisplatin, adriamycin, hydroxycamptothecine, and mitomycin were 7,6,6,6,5,5 and 5 respectively. No case was sensitive to methotrexate or dacarbazine. 3.Laparoscopic cholecystectomy in elderly patients: A clinical analysis of 182 cases

Objective To investigate the prevention and countermeasures of complications following laparoscopic cholecystectomy(LC) in aged patients.Methods The study was based on a retrospective view of clinical date of 182 aged patients(60~85 years old) who had been treated with LC from January 2000 to October 2006 in this hospital.Co-morbidities existed in 87 patients.The operation was performed under general anesthesia and endotracheal intubation,by using 3-port technique in 137 patients and 4-port technique in 45 patients. Results The LC was successfully completed in 174 patients,with an operation time of 30~100 min and a postoperative hospital stay of 3~10 d.Conversions to open surgery were required in 8 patients due to a difficult dissection(6 patients) or a diagnosis of gallbladder carcinoma(2 patients).No death or serious complications occurred.There were 3 patients with port-site infection and 2 patients with pulmonary infection.Follow-up observations in the patients for 3~12 months revealed no abdominal pain,fever,or jaundice.Conclusions Strictly following indications, properly treating the accompanying diseases,and promptly converting to open surgery are key factors to prevent and reduce complications of LC in aged patients.
4.Alterations of p14~(ARF) and p53 genes in human primary colorectal carcinomas

AIM: To investigate the genetic and epigenetic alterations of p14~ ARF gene and mutation status of p53 gene in human primary colorectal carcinomas and to analyze the relationship between the two gene changes and the role of abrogation of the p14~ ARF -p53 pathway in colorectal carcinogenesis. METHODS: The homozygous deletions, mutations, methylation of 5′ CpG islands, mRNA expression of p14~ ARF gene and mutations of p53 gene were assessed by PCR, direct sequencing, methylation-specific PCR, and RT-PCR in the tumorous and matched adjacent normal colorectal tissues from 56 patients with colorectal carcinoma. RESULTS: ① p14~ ARF alterations were detected in 27% (15/56) of colorectal carcinoma tissues studied, of which 1 case showed homozygous deletion, 14 cases showed 5′ CpG island methylation, and no mutation was found in any tumor. ②15 colorectal carcinomas with p14~ ARF alterations indicated lack of (13 cases) or at low level of expression (2 cases) of p14~ ARF mRNA, while expression of the p14~ ARF transcript was detected in the remaining 41 colorectal carcinomas and any matched adjacent normal colorectal tissues. ③ The mutations of p53 gene were detected in 48% (27/56) of colorectal carcinomas investigated. ④ Of these 56 cases, 12 had p14~ ARF alterations alone, 24 had p53 mutations alone, 3 had both p53 mutations and p14~ ARF methylation, and 17 had neither. 70% (39/56) of the samples had either or both abnormalities of the two genes, and p14~ ARF hypermethylation was related to wildtype p53 (P
5.Effect of Wrist-ankle Acupuncture on Post Traumatic Stress Disorder after Work-related Injury

Objective To study the clinical efficacy of wrist-ankle acupuncture on post-traumatic stress disorder (PTSD) after work-related injury. Methods 62 patients were divided into observation group (n=30) and control group (n=32). The observation group accepted wrist-ankle acupuncture combined with Paroxetine, and the control group accepted Paroxetine only. They were assessed with Clinical Administered PTSD Scale (CAPS), Hamilton Depression Rating Scale for Depression (HAMD), and Hamilton Anxiety Scale (HAMA) before, 6 weeks and 12 weeks after treatment. Results The scores of CAPS, HAMD and HAMA reduced in both groups (P<0.01) after treatment, and the reduction rate was greater in the observation group than in the control group (P<0.05). Conclusion Wrist-ankle acupuncture may further relieve PTSD after work-related injury
6.Construction of the vector for fusion protein gene driven by IGF - Ⅱ P3 promoter and its expression in hepatocellular carcinoma cells

AIM: To construct the shuttle plasmid vector for thymidine kinase (tk) and EGFP fusion protein gene driven by IGF - Ⅱ P3 promoter, and investigate the specific killing effect of the HSV - tk/GCV system on hepatocellular carcinoma(HCC) cells in vitro. METHODS: Recombinant shuttle plasmid vector was constructed by techniques of genetic recombination and screening, and identified by restriction digestion and sequencing analysis. Then the recombinant shuttle plasmid was transfected into HepG2 and HeLa cells by techniques of lipofectamine transfection and its expression was detected by fluorescence microscope and RT -PCR. Cell killing after ganciclovir(GCV) application was determined by MTT. RESULTS: Identification of pDC316 -tkEGFP- P3 by enzyme digestion and sequencing analysis showed that the length, inserted location and direction of the target genes which were inserted into the recombinant were correct. It was found that enhanced green fluorescence protein could only be seen in HepG2 cells, but not in HeLa cells. The results of RT -PCR showed that only two bands could be seen in the samples of pDC316 -tkEGFP- P3 transfected HepG2 cells. The MTT test showed the selective cytotoxicity of GCV to the transfected HepG2 cells. CONCLUSION: The shuttle plasmid vector carrying the tkEGFP fusion protein gene driven by IGF - Ⅱ P3 promoter has been constructed successfully and its specific expression in HepG2 cells provided a sound basis for targeted gene therapy for HCC.
7.Changes of the Resistance of Common Pathogenic Bacteria Isolated During the Past 5 Consecutive Years in Our Hospital to Quinolones

OBJECTIVE: To investigate the change of the resistance of common pathogenic bacteria isolated in the First Clinical Hospital of Jilin University during the past 5 years to quinolones.METHODS: Disk diffusion test was used to study the susceptibility test,with the results evaluated based on US Clinical and Laboratory Standards Institute(CLSI).WHONET-5 software was used to analyze the drug resistance data.RESULTS: During 2003~2007,the detection rates of extended spectrum beta-lactamases(ESBLs)-producing Escherichia coli and Klebsiella.spp were 43.6%~60.1% and 51.0%~65.1%,respectively;the detection rates of methicillin-resistant Staphylococcus aureus(MRSA) and methicillin-resistant coagulase negative Staphylococcus(MRSCN) were 50.8%~83.3% and 80.0%~81.5%,respectively;the resistance of common pathogenic bacteria to quinolones showed an increasing tendency,much as in MRSA,MRSCN,feces enterococci,Enterococcus faecalis,and Escherichia coli.CONCLUSION: Monitoring of the resistance of common pathogenic bacteria to quinolones should be strengthened.The change of the antimicrobial resistance should be investigated in order to direct rational drug use in the clinic and prevent the propagation of drug-resistant strains.
8.Affinity maturation of a single-chain antibody for hepatocellular carcinoma by error-prone PCR and DNA shuffling

AIM: To obtain a single-chain antibody with high affinity for hepatocellular carcinoma (HCC). METHODS: A second single-chain antibody mutant library was established by using error-prone PCR and DNA shuffling. Single-chain antibodies with high affinity for hepatocellular carcinoma were selected from phage antibody library by using ELISA. RESULTS: The content of the second single-chain antibody mutant library was about 4.5?10~7. Two selected mutants M25 and M36 were obtained after 3 rounds of panning and ELISA. Immunoassay showed that M25 and M36 bound to human HCC cells specifically. The relative affinity of M25 was 2.0 folds higher than that of the original antibody, and M36 was 2.4 folds higher than the original antibody. CONCLUSION: Error-prone PCR combined with DNA shuffling is an effective method to improve affinity of antibodies isolated from phage antibody library.
9.Retrospective study on treatment of intrathoracic fistula after esophageal cancer surgery

Objective:By comparing the clinical outcome of the modified triple tube method and the traditional three-tube method in the treatment of thoracic internal fistula after esophageal cancer surgery, in order to provide a basis for the clinical treatment of thoracic fistula after esophageal cancer surgery.Methods:The traditional three-tube method for the treatment of thoracic fistula after esophageal cancer surgery was the traditional group, and the modified three-tube method was the modified group. There was no spillover of the contrast agent during the above gastrointestinal angiography and the patient could eat normally as the standard of cure. In this study, we collected the data of thoracic fistula patients after esophageal cancer surgery who were treated with two kinds of treatment methods in our hospital in different time period(the traditional group: 10 patients from February 2008 to June 2014; the modified group: 36 patients from January 2012 to December 2019). Retrospective analysis of the two groups of patients in terms of general data(sex, age, etiology and other factors), time to cure and other indicators, compare the pros and cons of these two methods in the treatment of postoperative thoracic fistula.Results:There was no significant difference in general data such as sex, age, and cure time between the two groups, but there was significant difference in the cure time( P<0.01). The average cure time of the modified group was(38.08±11.97) days, which was significantly better than that of the traditional group(95.60 ± 7.79) days. Conclusion:Compared with the traditional three-tube method, the modified three-tube method can significantly shorten the treatment time when treating patients with thoracic fistula after esophageal cancer surgery.
10.Experimental research of shielding the patients during electron beam radiotherapy

Objective To explore the shielding effects of 1-4 layers of lead aprons (LPs) and body shielding devices (BSDs) against stray radiation (SR) outside the electron beam field of 6-15 MeV.Methods JR-115B LiF TLDs were used to measure the stray radiation doses (SRDs) to the patient undergoing treatment,before and after being shielding,for different distances,different energies,different applicators,variable layers of LPs,and different thickness of body shielding devices (BSDs),respectively,along long calculating and comparing the shielding ratios of LPs and BSDs against SR.Results When the applicator (10 cm × 10 cm) is unchanged,the shielding ratio increased with the increased distance from measuring point (r =0.717,P < 0.05) and decreased with the increased electron energy (r =-0.678,P < 0.05);when the energy was constant,there was no correlation between the shielding ratio and the size of applicator (P > 0.05);For lower energy electron beam of 6 and 9 MeV,the shielding ratio for 1 mm Pb-BSD was slightly higher than that for 2 layers of LA (t =2.519,2 662,P < 0.05),ranging from 81.5% to 95.3% and 55.4% to 84.6%,respectively.For 12 and 15 MeV higher energy electron beam,the shielding ratio for 2 mm Pb-BSD was slightly higher than that for 4 layers of LA (t=3.768,7.934,P<0.05),ranging from 64.6% to 93.4% and 51.1% to 92.4%,respectively.Conclusions LAs or BSDs are availavle for effectively reducing the doses from stray radiation,and may help reduce the secondary risks from stray radiation.BSDs have more obvious advantages than LPs with regard to shielding effect.
