International Journal of Traditional Chinese Medicine 2014;(10):879-882
Objective To research the efficacy of oralYiyuan-Guchangdecoction and enema retention ofYuyangye in treating chronic ulcerative colitis(CUC).Methods 70 patients of CUC were randomly divided into two groups: a control group of 34 patients, treated by mesalazine(etiasa) orally, 1g/twice daily; and a test group of 36 patients, treated byYuyangyedecoction for oral administration andYuyangyeenema retention. The treatment course of both groups was 8 weeks. The curative effect, clinical symptoms, endoscopic sign and pathology, and ESR, CRP, IL-8, and IL-13 after one course of treatment, were analyzed statistically. Results The total effect of the test group and the control group was 94.44%(34/36) and 64.71%(22/34), and the difference was significant statistically(Z=-4.207,P<0.05). The effects of test group were superior than the control group in terms of the clinical symptoms, endoscopic sign, and histopathology(P<0.05). The ESR, CRP, IL-8, IL-13 of the test group were(13.5±3.3)mm/h,(8.26±1.54)mg/L,(0.85±0.17)ng/ml,(37.18± 14.58)pg/ml before the treament, and(7.0±2.4)mm/h, (3.56±1.03)mg/L,(0.38±0.08)ng/ml, (76.33± 23.74)pg/ml after the treatment; The ESR、CRP、IL-8、IL-13 of control group were(14.5±4.8)mm/h, (8.41 ±1.36)mg/L,(0.86±0.15)ng/ml,(40.46±14.47)pg/ml before the treatment and(7.5±3.5)mm/h,(3.63± 1.10)mg/L,(0.36±0.07)ng/ml,(55.84±17.52)pg/ml after the treatment. ESR, CRP and IL-8 of both groups decreased significantly(P<0.05), while IL-13 significantly increased(P<0.05). ESR, CRP, IL-8 had no obvious statistical significance(P>0.05)between the two groups, while IL-13 of the test group was superior than the control group(P<0.05).Conclusion The therapeutic effect of oralYiyuan-Guchangdecoction and Yuyangyeenema retention is good in treating chronic ulcerative colitis.
2.Study on correlation between serum HCV-RNA level and Anti-HCV with liver fibrosis indexes in patients with chronic hepatitis C
International Journal of Laboratory Medicine 2015;(1):47-48
Objective To investigate the correlation between serum HCV-RNA level and anti-HCV with liver fibrosis indexes in the patients with chronic hepatitis C.Methods The serum samples were collected in 96 cases of chronic hepatitis C.The HCV-RNA level was determined by real time fluorescence quantification PCR and Anti-HCV was detected by ELISA.LN,PCⅢ and CⅣlevels were detected by the electrochemical iuminescence analyzer and HA was examined by radioimmunoassay(RIA).The correla-tion between the HCV-RNA and Anti-HCV levels with the liver fibrosis indexes was analyzed with the statistic software. Results Among these 96 cases of Anti-HCV positive chronic hepatitis C,52 cases (54.2%)were HCV-RNA positive. With increasing of the S/CO value of Anti-HCV,the detection rate of HCV-RNA was increased, which were 9.5%,37.1% and 92.5%.The differ-ences in HA,LN,PCⅢ and CⅣ levels between the high viral load group and the low viral load group had no statistical significance (P >0.05).Conclusion The positive detection rate of HCV-RNA is related with the S/CO value of Anti-HCV,the higher the S/CO value, the higher the positive detection rate of HCV-RNA.But the HCV-RNA level has no correlation with the liver fibrosis in-dexes.

Objective To evaluate the long-term results of Chiari osteotomy in the treatment of Legg-Calv?-Perthes disease for the patients of 8 years old or above and discuss the factors that influence the prognosis of this disease. Methods There were 116 hips of 103 patients of Perthes disease treated with Chiari osteotomy between June 1986 and December 2002. All patients were 8 years old or above. 72 hips of these cases acquired follow-up, the meaning follow-up duration was 97 months(range, 38-196 months). According to lateral pillar classification, 30 hips were classified as group B, 22 hips as B/C group, 20 hips as C group. There were 50 hips in male, 22 hips in female, 47 patients between 8 and 10 years old, 22 cases between 10 and 14 years old. X-ray showed 25 hips with subluxation of femoral head preoperatively. Results According to the revised Stulberg classification system: (1)31 hips had Stulberg classⅠ,Ⅱ outcome, in which B group accounted for 19 of 30 hips, the hips in group C had the worst results: of 20 hips, only 3 had a Stulberg class Ⅰ or Ⅱ outcome, the results for the hips in lateral B/C border group were intermediate between those groups B and C. 33 hips had Stulberg class Ⅲ, in which B group had 10 hips, B/C group had 11 hips, C group had 12 hips. 8 hips had Stulberg class Ⅳ or Ⅴ, B group had 1 hip, B/C group had 2 hips, C group had 5 hips. (2)All cases had a satisfied containment, no one had subluxation after Chiari osteotomy. (3)48% of male patients had a Stulberg class Ⅰor Ⅱ outcome, in contrast, only 23% of female had the comparable outcome. (4)55% of the group of 8 to 10 years old patients had a Stulberg class Ⅰ or Ⅱ outcome, in comparison with 17% from the group of 10 to 14 years old cases. Conclusion The patients older than 8 years at the onset of disease and in the lateral pillar B or C group should be treated with surgical containment, Chiari osteotomy can fulfill such task. The gender, the age of onset, especially the lateral pillar classification strongly correlate with the outcome of Perthes disease, and the latter can help us to evaluate the severity degree of the disease and the prognosis.

OBJECTIVETo investigate the expression of chemokine stromal cell-derived factor-1 (SDF-1) receptor CXCR4 in human gingival mesenchymal stem cells (GMSCs) and the migration potential of GMSCs stimulated with SDF-1.
METHODSHuman GMSCs were isolated by single-cell cloning method. Their cell surface markers were characterized by flow cytometry, and the rate of colony formation was evaluated. Differentiation assay was used to detect the differentiation potential of GMSCs. The expression of chemokine SDF-l receptor CXCR4 in GMSCs was detected by immunocytochemical staining. The chemotactic effect of SDF-1 on GMSCs was detected using a 24-multiwell Transwell cell culture chamber. The number of net migrated cells was counted in different microscope fields.
RESULTSHuman GMSCs possessed high self-renewal potential and formed single-cell colonies cultured in vitro. GMSCs expressed mesenchymal stem cells-associated markers CD44, CD73, CD90, CD105, and CD166, and the expression of hemopoietic stem cell surface markers CD14, CD34, and CD45 was negative. GMSCs differentiated into osteoblasts and adipocytes under defined culture conditions. The colony forming unit-fibroblastic for GMSCs was 21.4%/±2.8%. Immunocytochemical staining demonstrated that GMSCs expressed chemokine SDF-1 receptor CXCR4. The number of GMSCs migrating at concentrations of 100 ng.mL-1 and 200 ng.mL-1 of SDF-l in the Transwell cell culture chamber was significantly higher than that of the negative control (189.3±4.4, 164.6±4.9 cells/field vs. 47.8±2.5 cells/field, P<0.01). Treatment with the CXCR4 neutralizing antibody, an antagonist for CXCR4, significantly reduced the migratory effect compared with the negative controls (29.0±2.4 cells/field vs. 47.8±2.5 cells/field, P<0.01).
CONCLUSIONHuman GMSCs express chemokine SDF-l receptor CXCR4. SDF-1 may participate in regulating chemotaxis of human GMSCs. Results suggest that the migration induced by SDF-1 is mediated by CXCR4.
