Objective:Toprepare poly-DL-lactide-poly (PELA) microspheres encapsulating recombinant tissue inhibitors of metalloproteinase-1 (TIMP-1) adenovirus, and to investigate their effects on the proliferation of hepatocellular carcinoma HepG2 cells. Methods: The microsphere was constructed by encapsulating recombinant adenovirus containing TIMP-1 in biodegradable PELA. The diameter of the microsphere, quantity of virus encapsulated, loading rate, and releasing kinetics were measured. HepG2 cells were infected with the microspheres; the infection efficiency was examined by fluorescent microscope; and the ultrastructure was observed by TEM. The expression of TIMP-1 mRNA in HepG2 cells was examined by semi-quantitative RT-PCR, and the proliferation of HepG2 cells was detected by MTT assay. Results: The microsphere encapsulating recombinant TIMP-1 adenovirus was successfully constructed, with its diameter, entrapment efficiency, and virus loading rate being 1.965, 60.0%, and 10.5×10~8/mg, respectively. About 60% of the viruses were released within 120 h, and the total releasing time was longer than 240 h. Infection with rAdTIMP-1 PELA microsphere efficiently induced TIMP-1 expression in HepG2 cells, and significantly inhibited the proliferation of HepG2 cells, with the inhibitory rate being 47%. Conclusion: PELA microsphere encapsulating recombinant TIMP-1 adenovirus can markedly inhibit the proliferation of HepG2 cells, which provides an experimental basis for the combining macromolecular chemistry and gene therapy for treatment of hepatocellular carcinoma.

Objective To study community morbidity of gastroesophageal reflux cough (GERC),and then establish its procedure of diagnosis. Methods For the patients with chronic cough, on the basis of inquiring medical history and physical examination, chest radiograph, induced sputum, pulmonary ventilation function and airway hyperresponsiveness, blood eosinophilic, IgE,allergens skin test, nose, throat examination,sinus X-ray or pharyngoscope etc was managed. On the basis of the results,patients suspecting GERC was screened out, and then RDQ was performed, extra-esophageal symptoms related performance was inquired and scores were recorded. The patients with scores ≥ 12 were diagnosed as GERC; the patients with scores ＜ 12 were performed diagnosing treatment with proton pump inhibitor (PPI),and patients with positive results were diagnosed as GERC. The materials was analyzed statistically. Results The patients with chronic cough were 430 cases,and the community morbidity of GERC was 22.79%(98/430). RDQ ≥12 scores was 68.37%(67/98),and RDQ ＜ 12 scores was 31.63%(31/98).There was significant difference in extra-esophagus manifestation (P ＜0.01);26 cases with RDQ ＜12 scores were diagnosed as GERC (83.87%),and the rate of GERC in chronic cough was 21.63% (93/430). Conclusions RDQ is a favourable screening test in diagnosing GERC. PPI diagnosing treatment has high sensibility in GERC. It is necessary to establish community diagnosis procedure.

BACKGROUND:Inflammation plays an important role in vessel proliferation after balloon injury.Reducing inflammatory reaction may lighten the ocurrence and development of the restenosis after angioplasty.Studies have demonstrated that PPAR? excitomotor has inhibitory effects on inflammation development.OBJECTIVE:To observe the changes in inflammatory factors after carotid artery balloon injury in rats and the intervention of PPAR? excitomotor rosiglitazone.DESIGN,TIME AND SETTING:The randomized,controlled animal experiment was performed at the Central Laboratory of Shenzhen People's Hospital from January to June 2009.MATERIALS:Male SPF SD rats weighing about 350 g were selected to generate models of carotid balloon injury.METHODS:SD rats were equally and randomly divided into 3 groups:the control group,the balloon injury group and the rosiglitazone group.The left common carotid arteries were injured by balloon in the balloon injury group and the rosiglitazone group.The control group received sham operation.The rosiglitazone group was administered rosiglitazone daily by gavage,which began 4 days before operation and continued until harvesting.Accordingly,the control group and the balloon injury group were administered normal saline daily by gavage.MAIN OUTCOME MEASURES:All rats were executed under anesthesia at 14 days after operation,respectively to harvest left common carotid artery samples.The vessels were stained by hematoxylin-eosin,and Neointimal area(NIA) and media area(MA) as well as NIA/MA were calculated.Real time RT-PCR and Western Blot method were used to assay the expression of interleukin(IL)-6,IL-10,IL-17A mRNA and the distribution of nuclear factor(NF)-?B protein.RESULTS:Of the 36 rats,5 were excluded due to failed modeling or death,and 31 rats were included in final analysis.①The expression levels of IL-6 and IL-17A mRNA in the rosiglitazone group were significantly lower than the balloon injury group,but higher than the control group(P

Objective To establish a method for the determination of tetrahydropalmatine in Fuyantai Capsule. Methods TLC Scanning was used. with n-hexane ∶chloroform ∶methanol(7 ∶3 ∶0.5)as developer,scanned by double-wavelength reflection,?S=275 nm,?R=370 nm,slit 1.2?1.2 mm,SX=3.Results The linear range of tetrahydropalmatine concentration was 0.2～1.0 ?g and r=0.9992. The average recovery was 96.7 %and RSD was 1.92 %.Conclusion This method is accurate with a good reproducibility and can be used for the quality control of Fuyantai capsule.

CONCLUSION: Rosiglitazone can regulate the expression of II-6 IL-10 IL-17A mRNA and the balance of inflammatory factors via NF-kB,inhibit the inflammatory reaction of injured vessels and may contribute to lighten the restenosis of injured vessels.

BACKGROUND:Skeletal muscle satelite cels are muscle-derived stem cels with proliferation and differentiation potential distributing between the muscle cel membrane and the base film. Studies have shown that skeletal muscle satelite cels are of efficacy and safety, but the survival rate of the transplanted stem cels is very low, which greatly limits the application of skeletal muscle satelite cels. OBJECTIVE: To observe the effects of Fasudil on apoptosis of skeletal muscle satelite cels induced by H2O2. METHODS: Skeletal muscle satelite cels cultured in vitro were randomly divided into three groups including H2O2group, H2O2+Fasudil group (Fasudil group) and control group. Apoptosis rates were observed by flow cytometry. The concentrations of interleukin-4 and tumor necrosis factor-a in each group were detected by ELISA. Western blot was employed to measure the protein level of Bax in each group. RESULTS AND CONCLUSION: Compared with the H2O2group, a significant decrease was found in the apoptosis rate of cels, protein level of Bax, and concentrations of interleukin-4 and tumor necrosis factor-a in the Fasudil group (alP < 0.05). These findings indicate that Fasudil can play anti-apoptosis protection by inhibiting Rho-kinase signaling pathway, which may be related to the reduced expression of Bax.

AIM: To establish a method for the determination of ephedrine hydrochloride in Juyuanzhike Tablets (Radix Platycodonis,Radix Polygalae,ephedrinelydrochloride,etc.). METHODS :Ephedrine hydrochloride in Juyuanzhike Tablets were determined by HPLC. RESULTS : The linear range was 0.2～1.6 ?g,r =0.9999.The average recovery was 97.7% and RSD was 0.58%,respectively. CONCLUSION : The method is simple,feasible and reproducible. It can be used for the quality control of Juyuanzhike Tablets.

Objective To investigate the expression and immune function of NOD2 signal in MyD88-/- mice. Methods MyD88-/- mice and wild-type C57 BL/6 mice were characterized by PCR. Mice model of pulmonary infection was constructed by tracheal instillation of BCG vaccine strain(attenuated strain of Mycobacterium). PBS tracheal instillation was used as negative control.Peripheral blood sample and lung tissue were collected aseptically 24 h after Mycobacterium challenge. Real-time PCR and Western blot were used to detect the expression of NOD2 gene and protein. IL-6 level in the peripheral blood was determined by enzymelinked immunosorbent assay. Results The expression of NOD2 protein in BCG infected mice was significantly higher than PBS negative control group. NOD2 protein expression in MyD88-/- mice was higher than in wild-type mice. BCG infection was associated with higher NOD2 protein expression than infection-free PBS control in both groups of animals. The IL-6 level in peripheral blood was significantly higher after BCG infection than PBS group in both MyD88-/- mice and wild type mice. Conclusions BCG can activate the NOD2 signaling pathway when MyD88-dependent pathway is deficient.
