Objective To study the effects of the effective composition of Angelica sinensis injection on the functional state of platelet in patients with ulcerative colitis (UC),and its significance of therapy.Methods In 64 patients with UC and 30 healthy people, a-granule membrane protein (GMP-140), thromboxane B 2(TXB 2), 6-keto-PGF 1a were detected by means of ELISA.Platelet adhesion rate was detected by the platelet adhesion meter, von willebrand faetor related antigen (vWF:Ag) was also detected by the means of monoclonal -ELISA.Results The indexes of platelet activation and vWF:Ag in patients with UC were significanrly higher than those in the control group (P
2.INVESTIGATION OF RELATIONSHIP BETWEEN CLINICAL MANIFESTATIONS AND LIVER PATHOLOGY OF PATIENTS WITH ADVANCED SCHISTOSO-MIASIS COMPLICATED WITH HEPATITIS B VIRUS INFECTION

88 patients with advanced schistosomiasis japonica complicated with hepatitis B, which was confirmed by positive HBsAg and/or HBcAg in liver with double perioxidase-anti-perioxidase (PAP) method, were analyzed clinically and pathologically. It was found that there were four pathological types in the livers of these patientss simple schistosomial liver fibrosis (SSLF, 16.3%), SSLF complicated with chronic hepatitis (18.2%), SSLF complicated with inactive cirrhosis (29.6%) and SSLF complicated with active cirrhosis(38.6%). Among the 88 patients, 43(48.9%) cases had no clinical manifestation associated with the damage of liver function and only 23(26.1%) cases had the history of acute hepatitis. The relationship between clinical manifestations and liver pathological changes was also discussed.
3.Clinical observation on improved three-dimensional conformal radiotherapy in the treatment of massive type locally advanced non-small-cell lung cancer

Objective To analyze the effection of improved three-dimensional conformal radiotherapy (3D-CRT) to the dose of target area of local advanced non-small-cell lung cancer (NSCLC) and observe the therapeutic efficacy and toxity.Methods 81 patients with local advanced NSCLC were collected and treated.The diameter of tumor exceed 5 cm.52 patients were squamous carcinoma.24 patients were adencarcinoma.5 patients were adenosquamous carcinoma.63 patients were onstage of Ⅲ A,18 patients were Ⅲ B.The patients were randomized into two groups,the first group was unmodified planning of 3D-CRT group (T1 group,39 patients),the second group was modified planning of 3D-CRT group (T2 group,42 patients).The 31 patients of T1 group (79.5 %) received radiotherapy and chemotherapy.Toties quoties was 2 Gy.The fractions were 26-30.The total dose was 52-60 Gy.The 31 patients of T1 group (73.8 %) received radiotherapy and chemotherapy.Toties quoties was 2 Gy.The fractions were 30-35.The total dose was 60-70 Gy.Results The 1-,2-,3-year overall survive rates of T1 group were 56.4 %,33.3 %,28.4 %,the 1-,2-,3-year local control survive rates were 38.4 %,28.2 %,20.5 %,and the median survive time was 17 months.The 1-,2-,3-year overall survive rates of T2 group were 61.9 %,35.7 %,28.5 %,the 1-,2-,3-year local control survive rates were 47.6 %,40.4 %,30.9 %,and the median survive time was 19 months.The significant difference was found for the local control survive rates between T1 and T2 group (P < 0.05).No significant difference was found for the overall survive rates and the median survive time between T1 and T2 group (P > 0.05).Conclution Improved 3D-CRT can advance local control survive rate and living quality to local advanced NSCLC.Meanwhile,it also increases survive rate of 1-year,but can not increase long-time survival rate.
4.Ultrasonic Diagnosis of Fetal Hypospadias

Purpose To investigate the prenatal ultrasonic features of fetal hypospadias and to compare the diagnostic accuracy.Materials and Methods The clinical data of 28 patients with suspected or confirmed hypospadias were retrospectively analyzed.The prenatal ultrasonic features,missed diagnosis and misdiagnosis of the patients were analyzed,and the diagnostic accuracy of different ultrasonic features for hypospadias was compared.Results In 28 cases,21 cases of hypospadias were confirmed by examination after birth or induced abortion with 2 cases of missed diagnosis by ultrasound,and 7 cases of non hypospadias were misdiagnosed by prenatal ultrasound.The diagnostic accuracy rate of 2D ultrasound was 67.9% (19/28),and that of 3D ultrasound combined with 2D ultrasound was 92.9% (13/14).The diagnostic accuracy rates of ultrasonic features such as abnormal urinary jet,abnormal urethra,tulip sign,abnormal penis tip,scrotum separation with or without penoscrotal transposition and short penis were 100.0% (6/6),92.9% (13/14),88.2% (15/17),83.3% (5/6),25.0% (1/4) and 33.3% (1/3),respectively.Conclusion The ultrasonic features including abnormal urinary jet,abnormal urethra,tulip sign and abnormal penis tip have great value for prenatal diagnosis of fetal hypospadias.3D ultrasound is more intuitive than 2D ultrasound and provides more details.Comprehensive analysis of all ultrasonic features is helpful for prenatal diagnosis.
5.Inducement effect of tetrandrine on the apoptosis of cultured human Tenon's capsule fibroblasts

BackgroundExcessive conjunctival scar formation is a main cause of filtering surgical failure in glaucoma.It has been reported that the failure rate of filtering surgery is a tough problem in the research of glaucoma.Research showed that tetrandrine (Tet) suppress the proliferation of cultured human fibroblasts of Tenons capsule (TCFs) in vitro,but its possible mechanism is still unclear up to now.ObjectiveThe aim of this study is to investigate the inducement effects of Tet on the apoptosis of fibroblasts of Tenons capsule and mechanism.MethodsThe Tenon's capsule was obtained from donor eyes of Zhongshan Eye Center Eye Bank for further use.Human Tenon's capsule fibroblasts were cultured by explant culture method in mixed medium in vitro and the third to seventh generations of cells were collected for the experiment.The subcultured cells were identified by morphology observation and Vimentin staining.The apoptosis of cultured fibroblasts in Tet-treated group and control group was studied by using TUNEL (TdT- mediated dUTP nick end labling,TUNEL),flow cytometry (FCM) and transmission electron microscope.The cell number in different cellular cycle was calculated in Ted-treated group and control group.Results The cultured cells reached confluence in two weeks after cultured and presented the spindle or triangle shape with the radial-like or vortexin-like arrangement .The cells showed the positive staining for Vimentin.Apoptosis changes of cultured cell and apoptosis bodies were seen under the transmission electron microscope.Apoptotic cell nuclei were observed in Tet group by TUNEL.FCM result showed that the cells at G_0/G_1 phase decreased by 22.2%,and the cells at S and G_2/M phases increased by 20.53% and 1.6% in Ted-treated group.Significant differences in the numbers of cells in different cellular cycles and cell numbers of apoptosis were found between Tet-treated group and control group(P=0.000).ConclusionTet can inhibit the proliferation of human TCFs by inducing apoptosis in vitro.
6.Cloning, Expression and Identification of Membrane Protein Tetraspanin 2-A of Schistosoma japonicum

Objective To clone and express a membrane protein(Tetraspanin 2) gene of Schistosoma japonicum (SjTsp2). Methods A pair of primers was designed to amplify the SjTsp2 gene which was subcloned into prokaryotic plasmid pET28a(+). The recombinant plasmid was transformed into E. coli BL21(D3) and followed by expression of the protein induced by IPTG. The protein was purified by affinity chromatography and used to immunize BALB/c mice. Dilution of antibody against SjTsp2 was determined by ELISA. The protein was also identified by Western blotting. Results Big loop of SjTsp2-A, 228 bp, was amplified in vitro by PCR. Its deduced amino sequence shared 52% similarity with SmTsp2. The soluble recombinant SjTsp2-A was expressed in the experiment and high dilution antibody against the recombinant (1 ∶ 32 000 in maximum) was produced in immunized mice. SjTsp2-A reacted positively with sera of acute and chronic schistosomiasis patients but not with sera from healthy persons by Western blotting. Conclusion SjTsp2 has been expressed and shows certain antigenicity.
7.Reactivity of Human Preformed Natural Antibodies with Various Porcine Pancreatic Cells

The reactivity of human preformed natural antibodies (PNAbs) with various porcine pancreatic cells and its isotypes was investigated. Eighteen serum samples from patients with insulin-dependent diabetes mellitus (IDDM) and 20 serum samples from healthy human subjects were collected. The frozen sections of the pig pancreas were incubated with these sera, and subsequently incubated with FITC-conjugated goat antihuman IgG and IgM monoclonal antibodies. The reactivity of human PNAbs with various porcine pancreatic cells was determined by indirect immunofluorescence staining technique. The results showed that 55.6 % of IDDM patients and 55.0 % of healthy human individuals contained PNAbs against porcine endocrine cells. However, the percentage of strongly reacting sera in the patient group was significantly increased as compared with that in the control group. All used sera from IDDM patients and 95 % of sera from healthy controls could react to one or more of the various pancreatic cell types, including: endocrine cells, exocrine cells, vascular endothelial cells, ductal epithelial cells and macrophages. The isotypes of PNAbs contained both IgG and IgM. In view of strongly positive reactivity of PNAbs with various porcine pancreatic cells, pretransplantly cross-matching test and graft pretreatment may be necessary for survival of islet transplants.

9.The Molecular Biological Study of Hepatolenticular Degeneration

【Objective】 To investigate the molecular basis of hepatolenticular degeneration (Wilson disease, WD) and to attempt to construct the feasibility of gene diagnosis in the disease. 【Methods】 We have performed the molecular biological study on this disease for 10 years by molecular geneti c techniques. 【Results】 ①Location of WD gene in Ch inese: Using pairwise linkage analysis and multipoint linkage analysis method, w e constructed a genetic map of DNA markers within D13q14.2-3 which refined the location of WD gene by restriction fragment length polymorphism(RFL P) and microsatellite polymorphism analysis; ②Screen for mutations of WD gene in Chinese people: we detected the structure of 21 exons of WD ge ne in 45 patients from 39 pedigrees by PCR-SSCP(Single strand conformation poly morphism) and PCR-DNA sequencing technology, found a new mutation in exon 5 and nuclcotide sequence analysis showed it is a T insertion. We also conformed the Arg778Leu in exon 8, the highest frequence mutation point in Chinese people, wit h mutation rate 22.8% in total;③Carrier detection and presymptomatic diagnosi s of WD: Based on DNA recombination technology, we peformed successfully the gen e diagnosis in all individuals of 79 families with WD and built up a helpful spe cific enzyme cut method (PCR-Msp1) to detect the carrier and presympomatic patients in Chinese pe ople with WD. 【Conclusion】 These results showed that the location of WD gene within D13q14.2-3 is the same in Chinese as in Caucasians, but the g ene high m utation point,the gene diagnosis method and its pathogenesis are markly different.
10.Value of procalcitonin, matrix metalloproteinase-9 in the differential diagnosis of intracranial infection in children

Objective To observe the serum and cerebrospinal fluid (CSF) levels of procalcitonin (PCT),matrix metalloproteinase-9 (MMP-9) in children with purulent meningitis and viral encephalitis,and evaluat the differential diagnosis value of PCT and MMP-9.Methods The clinical data of 73 patients with intracranial infection admitted to neurology of Qingdao Women and Children's Hospital from September 2014 to December 2015.Twenty-two patients with pumlent meningitis were se1ected as purulent meningitis group,51 patients with viral encephalitis as viral encephalitis group,and another 20 non-infectious convulsion children as a control group.Samples of 2 ml CSF and 3 ml venous blood from all the subjects were collected within 24 hours after admission.The levels of MMP-9 were detected by Enzymelinked immunosorbent assay (ELISA),and the PCT 1evels were measured by electrochemiluminescence immunoassay.The data in multiple groups were compared with analysis of variance,and by SNK-q test to carry on pairwise comparison among groups.The positive rate significance compared by Chi-square test.Linear correlation analysis was used for correlation analysis.Results Mean serum and CSF levels of PCT in the pumlent meningitis patients group were significantly higher than those in the viral encephalitis group as well as the control group (P < 0.01),but the levels of serum or CSF there were no significant difference between the viral encephalitis group and the control group (P > 0.05);Mean serum and CSF levels of MMP-9 in the pumlent meningitis patients group were significantly higher than those in the viral encephalitis group and control group (P < 0.01),and the levels in patients with viral encephalitis were significantly higher than those in control group (P < 0.01);The increased percentage of the serum and CSF PCT in pumlent meningitis group were significantly higher than in viral encephalitis group (P < 0.01).,but there were no significant difference for MMP-9 (P > 0.05);The PCT level of serum or CSF in pumlent meningitis patients was positively correlated with MMP-9 (r =0.498,P < 0.01),but there was no significant correlation in viral encephalitis group (P > 0.05).Conclusion Detection of PCT and MMP-9 level is valuable in differential diagnosis for the pumlent meningitis or viral encephalitis.
