1.Preparation of gastric adenocarcinoma related cDNA microarray

Objectives To prepare the gastric adenocarcinoma related cDNA microarray and to screen differentially expressed genes between human gastric adenocarcinoma and normal gastric mucosa. Methods A set of cDNA microarray slides containing a duplicate set of 576 cDNA spots. These selected clones have been verified in sequence. The PCR products of human genes were spotted onto a chemical-material-coated glass plates in array. DNAs were fixed on the glass plate after series of treatment. Both RNAs from the gastric adenocarcinoma and normal gastric mucosa were reversely transcribed to the cDNAs with the incorporation of fluorescent dUTP to prepare the hybridization probes. The mixed probes were hybridized to the cDNA microarray. After high-stringent washing, the cDNA microarray was scanned for fluorescent signals to display differences between two tissues. Results Gastric adenocarcinoma related cDNA microarray slides contained 111 differentially expressed genes of gastric adenocarcinoma, 37 known genes related to gastric carcinogenesis (such as p21ras, p53, etc.) and control genes. The results of 10 couples of hybridizations to the gastric adenocarcinoma related cDNA microarray slides with Cy3-labeled cDNA probe from normal gastric tissue and Cy5-labeled cDNA probe from gastric adenocarcinoma showed analyzed spots uniformity background and clear signal. Among the 576 target genes, the expression level of 48 genes differed between gastric adenocarcinoma and normal mucosa in 5 or more samples (eight genes differed in all samples). There were 39 down-regulated and 9 up-regulated genes in gastric adenocarcinoma. Conclusion Gastric adenocarcinoma related cDNA microarray slides were successfully developed. The results of the experiments showed that gastric adenocarcinoma related cDNA microarray slides are reliable, sensitive and reproducible.
2.Expressions of light chain immunoglobulins kappa and lambda in multiple primary malignant tumors in senile patients

Objective To investigate the abnormal expressions of light chain immunoglobulins kappa (Ig?) and lambda ( Ig?) in multiple primary malignant tumors (MPMT) in elderly patients. Methods The expressions of Ig? and Ig? in specimens obtained from multiple malignant tumors in 21 elderly patients were investigated. Immunohistochemical staining was performed with mouse antihuman Ig? and Ig? monoclonal antibodies. Among 21 patients with multiple primary malignant tumors, the first tumor which occurred in the digestive tract was detected in 20 patients. In 17 patients first and second tumors were found to occur in the digestive tract. In 21 patients with MPMT the relationship between the expressions of light chain immunogloblin and survival time was analyzed. Results Positive staining of Ig? and/or Ig? in specimens from the first and/or second tumor tissues was found in 16 patients(76 2%). 3.Urodynamic analysis after operative treatment of traumatic posterior urethral disruption

12 ml/s was used as Group A and Qmax ≤12 ml/s as Group B. Results One week after removal of urinary catheter, dysury appeared in five cases with Qmax for (9.45?2.62) ml/s, of which two cases were with posterior urethral stricture, two with benign prostatic hyperplasia and one with weak detrusor contraction. 4.Roles of P53 and P21 in the differentiation of PC12 cells

Objective To investigate the roles of P53/P21 during neuronal differentiation with a differentiated model of PC12 cells. Methods A new cell line PC12(P53/m175) was created by stable transfection of a retrovirus plasmid pBabe-P53/m175, which contains a dominant-negative P53 gene mutant. After NGF treatment, observing with phase-contrast microscopy, flow cytometric analysis and western blotting of P53 and P21 were performed. Results Expression of P53 and P21 was obviously increased in NGF-induced PC12 cells. The appearance of cell cycle G1 phase arrest paralleled the increased expressions of P53 and P21. The level of P21 protein did not change after treatment with NGF in PC12(P53/m175) cells and the extent of G1 phase arrest markedly decreased. However, we did find the normal neurite outgrowth in NGF-treated PC12(P53/m175) cells. Conclusion NGF-induced an increased protein levels of P53 and its transcriptional element, P21 is essential for cell cycle G1 phase arrest, but does not necessarily correlate with the neurite outgrowth of PC12 cells.

6.Investigation of the efficacy and safety of tirofiban in elderly patients with acute myocardial infarction referred for percutaneous coronary intervention

Objective To evaluate the efficacy and safety of use of platelet membrane glycoprotein (GP) Ⅱ b/Ⅲ a inhibitor tirofiban for percutaneous coronary interventional (PCI) in elderly patients with acute ST segment elevation myocardial infarction (STEMI).Methods A total of 120 elderly patients who suffered from STEMI and underwent PCI were selected from July 2007 to November 2009.The patients were randomly assigned to control group, standard-dose tirofiban group and low-dose tirofiban group.We observed coronary blood reflow, the bleeding complication, coagulation factor(TF,vWF) and cell adhesion molecular(sICAM-1 and sVCAM-1).Results Total 116 patients accomplished this study (control group: 38 cases; standard-dose group: 39 cases; low-dose: 39 cases).The percentages of TIMI 3 flow after PCI were higher in the two tirofiban groups than in control group (P<0.05).The incidences of bleeding complications in both tirofiban groups (12.8%,5.1%) were higher than that in control group (2.6%, P<0.05).The concentration of TF, vWF,sICAM-1 and sVCAM-1 were lower in both tirofiban groups than in control group(P< 0.05).Conclusions GP Ⅱ b/Ⅲ a inhibitor tirofiban can benefit the elderly patients with STEMI referred for PCI therapy, low-dose tirofiban may offer almost the same level of efficacy as standard-dose, with less associated bleeding.
7.18F-FDG PET/CT in Synchronous Multiple Primary Esophageal Carcinomas

Purpose Synchronous multiple primary esophageal carcinomas may be undetected by gastroscopy. This study aims to assess the role of 18F-fluorodeoxyglucose (18F-FDG) PET/CT in detecting synchronous multiple primary esophageal carcinomas. Materials and Methods Thirteen patients with synchronous multiple primary esophageal carcinoma who underwent gastroscopy and 18F-FDG PET/CT within two weeks before treatment were retrospectively analyzed. Results Gastroscopy and 18F-FDG PET/CT detected 22 and 29 esophageal carcinomatous foci respectively, and the difference was statistically significant (Z=-2.333, P<0.05). All the 29 esophageal carcinomatous foci showed high 18F-FDG uptake with a SUVmax ranging from 3.1 to 20.1 (mean, 10.7±5.7). 18F-FDG PET/CT also detected surrounding organ invasion in 3 patients, lymphatic metastasis in 10 patients and distant metastasis in 3 patients. Conclusion 18F-FDG PET/CT detects more esophageal carcinomatous foci compared with gastroscopy in patients with synchronous multiple primary esophageal carcinomas and also performs the staging diagnosis, which may help in improving clinical management and prognosis.
8.Surgical Treatment of Female Stress Urinary Incontinence by Tension-free Vaginal Tap

Objective To describe the clinical curative effects of surgical treatment of female stress urinary incontinence by tension-free vaginal tap.Methods Surgical treatment of 17 women suffering from stress urinary incontinence whose average course are11years by tension-free vaginal tap.Then,observe the complications and how symptome improved after operation.In this 17 cases,one had a failure experience of bladder neck ventrofixation,another had an operation of pelvic cavity in the past.Results All operations were successed,the mean operative time was about 30 minutes,the mean amount of bleeding was 40ml.No case had the complication of bladder perforated and dysuria after operation.The urinary incontinence symptomes were disappered within 5 days after operation.During a follow up of 3~12 months,no SUI recurred.Conclusions Our result demonstrates that TVT has the virtues of small trauma、simplely manipulate and less complications.Patients recover well and have a high rate of curing.It can also be use in such patients who had operations of pelvic cavity and failure operation for SUI in the past.
9.Echocardiographic diagnosis of juxtaposition of atrial appendage

Objective To study the value of echoeardiography for diagnosis of juxtaposition of atrial appendage(JAA) and to discuss its features. Methods Eehocardiographic characteristics of JAA in nine cases were compared with results of cardiac catheterization and operation. The diagnostic features of echocardiography were summarized. Results Seven cases had juxtaposition of left atrial appendage and two had juxtaposition of right atrial appendage. The nine cases were all associated with severe congenital heart disease and the most frequent malformations observed with JAA were double outlet right ventricle, transposition of great artery, single ventricle and so on. Direct visualization of the JAA in the parasternal short-axis view at the base of the heart and visualization of an unusual transverse orientation of the atrial septum were the most features of JAA. Echocardiographie characteristics of JAA cases were correspondent to the results of cardiac catheterization and operation. Conclusions There is high accuracy by echocardiography to diagnosis JAA. JAA should be alerted in severe congenital heart disease.
10.Application of CT values in calculation of proton incident energy in proton treatment planning system

Objective To explore the application of CT value in calculating the proton incident energy in proton treatment planning system. Methods Bethe-Block formula and the formula for calculating the proton range were analyzed to study the correlation of the range of proton beam ( 70-250 MeV ) between a variety of radiation equivalent material and water. Procedure of Monte Carlo SRIM2008 was used to verify the possibility of a constant proportional coefficient of range ( Ci ). The proportional coefficient ( Ci ) of range in radiation-equivalent material and the CT value were fitted by using Origin 8.0 software to study the functional relation of CT value and Ci. The actual range of proton was equivalent to a range of water and incident proton energy could be calculated. Results There was a constant range of Ci of proton beam (70-250 MeV) between a variety of radiation equivalent material and water. There was a functional relation between CT value and Ci ( r = 0.999). The actual range of proton in radiation equivalent material can be equivalent to a range of the water. Conclusions CT values and a range of proportional coefficient ( Ci ),and the actual required range of the tumor could be used to accurately calculate the water equivalent range,and the incident proton energy to the position of Bragg peak. A new exploration for using CT technology in proton treatment planning system could be obtained.
