1.Coexistence of Gitelman's syndrome and thyroid disease: SLC12A3 gene analysis in two patients

Two patients with coexistence of thyroid disease and suspected Gitelman's syndrome underwent SLC12A3 gene analysis. The results confirmed that both patients were compound heterozygotes of SLC12A3 gene mutation. Three novel variants of SLC12A3 were found in this study. This report suggests that Gitelman's syndrome may coexist with other disorders associated with hypokalemia, such as Graves' disease.
2.TRANSFORMATION OF BACILLUS THURINGIENSIS RECIPIENT BMB171 BY ELECTROPORATION

This paper reports the optimized electro-transformation parameters of Bacillus thuringiensis plasmid-free mutant strain BMB171 by electroporation, and expressing effect of several cry genes introduced in this recipient. It showed that a highest electro-transformation frequency could be obtained, when SG solution was used as the buffer, and a 10.0V/cm of field strength, one time of pulse as well as a growth phase of recipient cells at the exponential phase (OD650nm value was 0.2~0.3) were selected. The highest of electro-transformation frequency with pHT3101 could reach at 8 ?107 hansformants/?g DNA. The transformation frequencies increased at linear velocity as the concentration increase of pHT3101 from 54.69pg/ml to 3.50?g/mL, then reached saturation afterwards. All plasmids introduced in BMB171 could produce characteristic insecticidal crystal proteins through expression of relevent cry genes carried by them. Meanwhile, these insecticidal crystal proteins could form parasporal crystals, which have characteristic geometric shapes.
3.Analysis and identification of a novel CD36 allele, 1142 T>G

BACKGROUND:As a main antigen of platelet, CD36 antigen is also known as platelet glycoprotein IV (GPIV). The mutation of CD36 gene may result in deficiency of the antigen.
OBJECTIVE:To identify a novel CD36 alele.
METHODS: DNA was isolated from peripheral blood sample, and 12 coding regions of CD36 gene were amplified by PCR. Sequencing-based typing was used to analyze the sequence of the target regions. The derived sequences were aligned with the standard sequence of NG_008192 in GenBank to identify the novel alele.
RESULTS AND CONCLUSION: 1142 T>G mutation was detected in exon 12 of CD36 gene of the proband, and the other regions were consistent with the standard sequence. No data or report about 1142 T>G was found in GenBank or National Center for Biotechnology Information (NCBI), and thus it was reported to GenBank and received by number KM275213. 1142 T>G results in amino acid 381 Leu>Ser of the CD36 protein. There is a big difference in hydrophilia and polarity of the two amino acids. Also the 381 amino acid locates in highly conserved region. Thus it is speculated that 1142 T>G may reduce or vanish the activity of the protein.

5.Heavy silicon oil tamponade in vitreoretinal surgery for traumatic proliferative vitreoretinopathy

Objective To study clinical application and complication of heavy silicon oil (Densiron68) in the treatment of traumat- ic proliferative vitreoretinopathy.Design Non-comparatives,retrospective case series.Participants Twenty patients with proliferative vitreoretinopathy resulting from ocular trauma were recruited,whose retinal detachment arising from inferior or posterior retinal breaks. Methods Heavy silicon oil was applied to patients during vitrectomy.Silicone oil or gas was applied to patients with redetachment after heavy silicon oil was removed.Main Outcome Measures The rate of retinal attachment,vision,intraocular pressure,inflammatory re- action of anterior segment and silicone oil emulsification period.Results The rate of retinal attachment with one operation using heavy silicon oil was 50%(10/20 eyes)and 15%(3/20 eyes)with further surgery.The average follow-up time was 3.90?1.41 months.At the end of the follow-up,all tamponade agents were removed in 50% patients.Patients' logMAR vision after the surgery was 2.19?0.86,which was better than before the surgery (2.63?1.00) (P=0.037).There was little evidence of high intraocular pressure,excessive inflammatory reaction of anterior segment and cornea endothelial cell damage,but cataract became more serious without exception.Emulsification rate was 100% and the average emulsification period was 2.18?0.87 months.Conclusions Heavy silicon oil tamponade in vitreoretinal surgery for traumatic proliferative vitreoretinopathy has good efficacy and relatively few complications.However,its emulsification period is relatively short,which may constraint its application to a certain extent.
7.Qualitative research on the factors affecting compliance of patients with maintenance hemodialysis

Objective To explore the factors affecting compliance of patients with maintenance hemodialysis and to provide reference for the improvement of patients' compliance.Methods Ten patients with maintenance hemodialysis were individually interviewed in this study.The data were analyzed based on phenomenological method.Results Three factors were found,including feasibility of doctor's advice,characteristics of patients such as awareness of disease related knowledge,recognition todisease progression and usual behavioral pattern,as well as social factors such as economic status,life pressure,family support,professional supervision and guidance.Conclusion In order to enhance patients' compliance,it is suggested to improve thefeasibility of doctors' advice and health education,as well as pay close attention to the influence of patients' recognition,psychological status and social factors on the compliance.
8.Design of ultrasound PACS image system based on B/S mode

Objective To construct the ultrasound PACS image system based on B/S mode.Methods The modern internet technology was used to build the PACS image system based on B/S mode.The B/S mode was typical three layer structures,namely,client presentation layer,middle service logic layer and storage layer.Results Client presentation layer was a figure surface to provide application service for clients.The function of service logic layer was performing application strategy and packaging application mode which was shown to client application program.Storage layer was used to define,service,visit and update data.Conclusion The versatility of B/S mode is good.Besides,the software is easy to operate and upgrade with low cost.
9.Comparison of MRI artifacts caused by Ni-Cr alloy fixed prostheses on different field-strength magnets

Objective To evaluate the influence of different field-strength magnets(1.5 T and 3.0 T)on MRI artifacts caused by Ni-Cr alloy fixed prostheses.Methods The crown,bridge and upper denture fixed prostheses with different thickness were produced by Ni-Cr alloy as test samples,and were one by one put on the centre of water phantom for MR scanning with different field-strength magnets(1.5 T and 3.0 T).The artifact areas on these two field-strength magnets were measured and statistically compared.The plastic prostheses with the same shape and thickness as the test samples were served as controls.Results Ni-Cr alloy fixed prostheses could cause MRI artifacts,and the artifact areas increased with the mass of prostheses.However,no artifact area was found in controls.Compared with those on 1.5 T magnet,the MRI artifact areas significantly increased on 3.0 T magnet(P
10. Preparation and quality evaluation of Acori Tatarinowii Rhizoma-volatile oil microemulsion

Objective: Taking the volatile oil of Acori Tatarinowii Rhizoma with the effect of anti-senile dementia as model drug to prepare Acori Tatarinowii Rhizoma-volatile oil microemulsion (AO-ME). Methods: The composition of microemulsion was preliminarily determined according to the solubility of volatile oil from Acori Tatarinowii Rhizoma in various solvents. By drawing pseudo-ternary phase diagram, the optimal microemulsion formulation was selected according to the size of particles and microemulsion region, stability of preparations and drug loading. Results: The optimal microemulsion formulation was that volatile oil of Acori Tatarinowii Rhizoma-Cremophor EL-glycerol-water was 6.25:20.83:10.42:62.5. The average particle size of AO-ME was (30.5 ± 0.2) nm with a polydispersity index of 0.150 ± 0.002. The viscosity value of AO-ME was (2.80 ± 0.21) mPa∙s. In vitro transdermal rate of AO-ME was (1 288.76 ± 16.20) μg/(cm2∙h), measured in β-asarone. Compared with ordinary emulsion, the transdermal rate of AO-ME was increased 14 times. The ciliary toxicity test of toad showed that the preparation had no significant toxicity. Conclusion: The preparation of AO-ME was prepared with stable physicochemical properties, good transdermal properties, and low toxicity to nasal mucosa cilia.
