3.Human epidermal thickness and skin aging
Chinese Journal of Medical Aesthetics and Cosmetology 2011;17(1):34-36
Objective To clarify the association of age and sun exposure with human epidermal thickness, and to probe deeply into the changes of skin histology and epidermal thickness with skin aging. Methods Two hundreds and eleven full thickness skin samples were collected from 3 different age groups of healthy volunteers and then stained with haematoxylin and eosin to study the changes of epidermal thickness using image analysis software ( Image pro plus 6.0). The obtained values were analyzed with factorial design ANOVA and correlation analysis to detect any significant effects of age and sun exposure on epidermal thickness. Results With increasing age, the junction between epidermis and dermis became flattened, and stratum comeum got looser. There was a notable attrition of the epidermal thickness that could be correlated with age. The at-trition of epidermal thickness attributed to age did not vary between female and male volunteers. Epidermal cell layers thickness increased with cumulated sun exposure, but epidermal total thickness was not associated with sun exposure. There was no difference for epidermal thickness of different sun exposure between female and male volunteers. Conclusion During chronological aging, epidermal cells decrease in thickness with lowered metabolism. With cumulated sun exposure, epidermal cell layer thickness becomes thicker. There is no interaction between the effect of age and sun exposure and the epidermal thickness.
4.Early diagnosis of colonic anastomotic leakage by detecting bacterial DNAs in rots with PCR
Journal of Chinese Physician 2008;10(6):749-751
Objective To asses the value of detecting bacterial DNAs in rat's blood with PCR for early diagnosis of colonic anastemotic leakage. Methods 48 healthy female Wistar rats were random divided into three groups: Group A(n=8,sham operation group),Group B(n=20,colonic anastomosis group),Group C(n=20,colonic anastomotic leakage group).Group B and C rats underwent standardized colon resection 3cm away from the ileocecal junction 10cm,Group B rats were done with a complete anastomosis(end-to-end single layer anastomosis with 0# silk sutures) while Group C rats were done with an anastomosis leaving a 5mm opening in colonic anterior wall.lml and 3ml venous blood samples were collected from Group A,B and C.DNAs were extracted from these blood samples and PCR techniques were used to amplify lacZ genes from Escherichia coli and 16S ribosomal RNA genes(16SrRNA genes) 3 days after operation. The data were analyzed by chi square test.Specimens of the experimental intestine were HE stained for pathological studies. Results The positive ratios of expressing lacZ genes in peripheral blood(PB) with PCR in Group C were significantly higher than that in Group B(P<0.05),hut there were no differences between the two groups in expressing 16SrRNA genes(P>0.05).Conclusions It could be a useful way to detect lacZ genes of Escherichia coli from PB by PCR but not 16SrRNA genes for diagnosis of colonic anastomotic leakages.
5.Relationship between reduced folate carrier gene methylation and methotrexate resistance
International Journal of Pediatrics 2014;41(3):267-270
Tumor recurrence and drug resistance impact patients'therapeutic effect and prognosis seriously. In recent years,it has been found that epigenetic changes are involved in drug resistance.DNA methylation is one of the ways of epigenetic.DNA methylation of drug metabolism enzyme gene can affect gene's expression, leading to drug resistance.Methotrexate(MTX),a folic acid antagonist,is the most widely used drug in the treatment of malignant tumors, such as leukemia, osteosarcoma, head and neck cancer, and so on.Reduced folate carrier(RFC) is the main transporter cartier of MTX,and gene methylation is related to tumor cells'resistance to MTX.This paper reviews the role of RFC,relation of RFC methylation and methotrexate resistance and the factors of RFC methylation.

10.The Analysis of Postoperative Curative Effects of Facial Paralysis Caused by Middle Ear Cholesteatoma
Journal of Audiology and Speech Pathology 2017;25(3):246-249
Objective To study the effects of the duration and degree of the peripheral facial paralysis due to middle ear cholesteatoma on the recovery of postoperative nerve functions. Methods The clinical data of 30 cases (single ear) of patients with cholesteatoma complicated with facial paralysis were retrospectively analyzed. The mean age was 54.7±8.9 years old (range 21~77years old) including 17 males and 13 females. The duration of facial paralysis, degree and location of facial nerve injury, surgical timing of facial nerve decompression, recovery of facial paralysis after surgery were followed up.According to the course of paralysis, the patients were divided into ≤2 months group (14 cases) and >2 months group (16 cases).According to the degree of paralysis, the patients were divided into the incomplete facial paralysis group (III-IV grade,14 cases) and the complete facial paralysis group (V-VI grade,16 cases). The influence of the course and degree of paralysis on the postoperative recovery of neurological function were analyzed by the Fisher exact probability test. Results Three cases underwent open radical surgery with no facial nerve damage confirmed by intraoperative exploration, and their postoperative facial nerve functions were fully restored.Facial nerve canal damage was found in 27 cases, accompanied by facial nerve congestion, edema or granulation formation.Among them, the facial nerve damage location was the tympanic segment in 20 cases. The open radical operation and local facial nerve decompression were carried out, and the postoperative facial paralysis recovery rate was 46.67%(14/30).The facial nerve function recovery rate in less than 2 months group was higher than the >2 months group (P<0.05).The facial nerve function recovery rate of the incomplete paralysis group was higher than the complete facial paralysis group (P<0.05).Conclusion This study suggests that facial nerve damage most occurred in the tympanic segment of the facial nerve.Radical mastoidectomy and local facial nerve decompression are effective for the treatment of middle ear cholesteatoma complicated with facial paralysis. The shorter course and lighter degree of paralysis lead to the better postoperative recovery of neurological function.
