1.Gene expression profile of multiple myeloma cell line treated by arsenic trioxide.

cDNA microarray was used to compare the gene expression profiles of multiple myeloma cell line RPMI8226 24 h before and after treatment with arsenic trioxide. Two cDNA probes were prepared by mRNA reverse transcription of both arsenic trioxide-treated and untreated RPMI8226 cells. The probes were labeled with Cy3 and Cy5 fluorescence dyes separately, hybridized with cDNA microarray representing 4096 different human genes, and scanned for fluorescence intensity. The differences in gene expression were calculated on the basis of the ratios of signal intensity of treated and untreated samples. The up-and down-regulated genes were screened through the analysis of gene expression ratios. The results showed that 273 genes were differentially altered at mRNA level, 121 genes were up-regulated and 152 were down-regulated. It is concluded that the treatment with arsenic trioxide can induce a variety of gene changes in RPMI8226 cell line. Many genes may be involved in the pathogenesis of multiple myeloma. ALK-1 and TXNIP genes may play an important role in the apoptosis and partial differentiation of RPMI8226 cells.

2.INVESTIGATION OF REALGAR IN THE TREATMENT FOR MULTIPLE MYELOMA

Objective To observe the clinical effect of realgar on multiple myeloma and to investigate its mechanism. Methods MTT and double antibody cramped ELISA assay were used to detect the activity of interleukin-6(IL-6) and level of soluble interleukin-6 receptor(sIL-6R) of the bone marrow supernant in 15 multiple myeloma patients treated by realgar or not. Results The activity of interleukin-6 and level of soluble interleukin-6 receptor of multiple myeloma patients were significantly higher than that of control group(P<0.01). They were not apparently decreased after realgar was used for 50 days(P>0.05).The interleukin-6 activity of stage Ⅲ patients were much higher than that of stage Ⅰ or Ⅱ(P<0.05). There was no difference of the level of soluble interleukin-6 receptor between two groups(P>0.05). For the 15 patients who were treated by using realgar, 3 got complete remission(CR), 5 got partial remission(PR), 7 came to not remission(NR). Conclusion Realgar could not decrease the activity of interleukin-6 and level of soluble interleukin-6 receptor so as to inhibit the proliferation of tumor cells.
3. Study on rat small intestinal absorption features of puerarin loaded O-carboxymethyl chitosan microspheres based on single pass perfusion method

Objective: To study the rat intestinal absorption characteristic of puerarin loaded O-carboxymethyl chitosan (CMCS) microspheres in situ. Methods: Single-pass intestinal perfusion (SPIP) model was used for rat in situ and HPLC to determine the concentration of puerarin. The effects of different perfusion rate, drug concentration, and intestinal segments on intestinal absorption were investigated, and the absorption characteristics of the puerarin and puerarin loaded O-CMCS microspheres were compared. Results: Perfusion rate had significant effect on the absorption rate constants (Ka) and the apparent absorption coefficient (Papp) (P < 0.05); The concentration of puerarin in perfusate had no significant effect on Ka and Papp (P > 0.05); Ka and Papp of drug loaded microspheres in jejunum and ileum showed no significant difference, but they were significantly higher than that in duodenum (P < 0.05); The drug loaded microspheres and puerarin had the significant difference in Ka and Papp in jejunum (P < 0.05). Conclusion: The absorption mechanism of puerarin in the microspheres is passive diffusion, puerarin is absorbed well in jejunum and ileum, and puerarin loaded O-CMCS microspheres can significantly improve the absorption of puerarin.
4. Progress in research on prevention and treatment of hemophilia inhibitors

Hemophilia F/F inhibitor is a kind of specific neutralizing antibody, making treatment invalid, and increasing bleeding and the incidence of death and disability. It is a serious complication of hemophilia and also a new challenge. Risk factors associated with the production of inhibitors are related to genetic factors and the environment, providing a direction for prevention. The existing effective treatments of hemophilia inhibitor include bypass drugs for homeostasis and immune tolerance induction therapy to clear inhibitor and new homeostatic prophylaxis. However, these treatment still cannot meet the needs. Recently, a variety of new hemostatic drugs have been developed, which have a long half-life and can be injected subcutaneously once every four weeks for prevention and treatment. The annual bleeding rate is 0 and no inhibitor is produced, which has made a breakthrough.
5.Effect of Expanding Training Orthosis on Webbed-Finger Adhesion after Burns

Objective To observe the effect of the expanding training orthosis on webbed-finger adhesion post burn. Methods From May, 2014 to April, 2016, 37 hands of 33 patients with webbed-finger adhesion post burn were divided into two groups. 21 hands from 18 patients were as experimental group, 16 hands from 15 patients were as control group. The control group accepted routine care, while the experimental group accepted the expanding training orthosis in addition. Their angles between the fingers were measured six months after treatment. Results The angles between the fingers increased in the experimental group after treatment (t>3.060, P<0.01), and were more than those of the control group (t>2.273, P<0.05). Conclusion Application of expanding training orthosis can improve the compliance of the patients with webbed-finger adhesion post burn, and promote hand function recovery.
6.Tianma Goutengyin and Its Single Herb in Treatment of Parkinson's Disease: A Review

Parkinson's disease (PD) is a kind of chronic progressive neurodegenerative disease that has a high prevalence rate in recent years, especially in the elderly. PD belongs to the category of "tremor disease" and "tremor" in traditional Chinese medicine, and Tianma Goutengyin is a classic prescription contained in the Synopsis of The New Significance of Patterns and Treatment in Miscellaneous Diseases(《中医内科杂病证治新义》). This article explored the theory of Tianma Goutengyin in the treatment of PD, and based on network pharmacological research, the article summarized relevant research on Tianma Goutengyin and its single herb in the treatment of PD. Moreover, it discussed the clinical applications and mechanisms of Tianma Goutengyin and its single herb in the treatment of PD. It is found that the mechanisms of Tianma Goutengyin in treating PD may be related to resisting oxidative stress, inhibiting inflammatory response, regulating neurotransmitters, and protecting dopamine (DA) neurons. Besides, the main components of the single herb in Tianma Goutengyin for treating PD are gastrodin, rhynchophylline, geniposide, gardenial alcohol, eucommitol glycoside, motherwort alkaloid, baicalin, pachyman, and achyranthes bidentata sterol. They can improve the related symptoms of PD patients by inhibiting inflammatory response, resisting oxidative stress, affecting calcium ion concentration, restoring mitochondrial function, and and protecting DA neurons. This article summarized the research progress of Tianma Goutengyin and its single herb in treating PD, so as to provide a reference for the prescription and medication of Tianma Goutengyin in the treatment of PD and subsequent research on the mechanisms of Tianma Goutengyin and its single herb in the treatment of PD and give play to the advantages of traditional Chinese medicine in the treatment of PD.
7.Comparison of Diosgenin Content in Wild Dioscorea zingiberensis from Different Habitats

OBJECTIVE:To establish a method for the content determination of diosgenin in Dioscorea zingiberensis,and to compare the contents of diosgenin in wild D. zingiberensis from different habitats. METHODS:HPLC method was adopted. The determination was performed on Inertsil ODS-3 column with mobile phase consisted of acetonitrile-water (50 ∶ 50,V/V) at the flow rate of 1.0 mL/min. The column temperature was set at 30 ℃. The detection wavelength was set at 203 nm,and sample size was 10 μL. Established method was used to determine the contents of diosgenin in wild D. zingiberensis from different habitats (Shaanxi Shangluo,Shaanxi Ankang,Henan Neixiang,Yunnan Xuanwei,Sichuan Deyang,Hubei Shiyan,Hunan Zhangjiajie,Hunan Changde). The differences of diosgenin content were compared. RESULTS:The linear range of diosgenin were 4.16-165.6 μg/mL (r= 0.999 9). RSDs of precision,reproducible and stability tests were all lower than 2% (n=5 or n=6). The average recovery of diosgenin was 101.18% (RSD=1.27%,n=6). The content of diosgenin in wild D. zingiberensis from different habitats were in descending order,i.e. Sichuan Deyang (1 405.36 μg/g)>Shaanxi Shangluo (1 201.79 μg/g)>Hunan Zhangjiajie (1 035.18 μg/g)>Shaanxi Ankang (632.64 μg/g)>Hunan Changde (598.64 μg/g)>Yunnan Xuanwei (425.34 μg/g)>Henan Neixiang (350.13 μg/g)>Hubei Shiyan (338.39 μg/g). CONCLUSIONS:Established method is simple,accurate and suitable for the content determination of diosgenin in D. zingiberensis. The contents of diosgenin in wild D. zingiberensis from different habitats are different significantly,and the content of diosgenin in the samples from Sichuan Deyang is the highest.
8.Research progress of Hedgehog signaling pathway inhibitors in anti-tumor field

Hedgehog (Hh) signaling pathway plays an important regulatory role in the process of cell proliferation, differentiation and tissue formation. Proper intensity and action time of Hh signal are crucial for the normal development of various tissues of the body, and its abnormal activation will lead to the occurrence and development of most malignant tumors, including breast cancer, liver cancer, pancreatic cancer, and lung cancer, which makes Hh signaling pathway an ideal target for anti-tumor drug research and development. At present, the main targets of Hh signaling pathway inhibitors include Hh ligand, receptor Smoothened (Smo) and transcription factor Gli. Among them, the compounds that depend on the Hh ligand pathway still remain at the stage of laboratory research because they cannot act on the non-classical Hh signaling pathway. The special structure of Smo protein enables it to combine with drugs efficiently and selectively, which is a powerful and effective drug target. Therefore, Smo selective inhibitors have been an active field of related research, and many Smo inhibitors have entered the clinical use or trial stage. Gli can regulate multiple carcinogenic genes, promote abnormal cell proliferation and lead to tumor, and can also cause feedback inhibition to Hh signaling pathway. Therefore, the development of drugs that can inhibit the activity of Gli has broad prospects. In the future, a combination of multiple pathway inhibitors can be designed to avoid drug resistance and other side effects.
9. Effect and mechanism of Qifu Yin regulation of JAK2/STAT3 pathway to improve type 2 diabetic cognitive impairment

Objective To explore the regulatory effeet of Qifu Yin ( QFY) on JAK2/STAT3 pathway in rats with type 2 diabetic cognitive impairment.Methods A small dose of STZ combined with high-fat and high- sugar feed was used to build the model.After success, they were divided into model group, QFY low-dose, high-dose group, and metformin group.After four weeks of intervention, fasting blood glucose ( FBG ) was measured; Morris water maze was used to detect spatial learning and memory ability in rats; Nissl staining and immunofluorescence staining were respectively used to detect the degree of brain injury and the expression of lba-1 , a marker of microglia .EL1SA was used to detect the expression of TNF-cx, IL6, ILK) and BDNF.Western blot was employed to detect the expression of JAK2/STAT3 pathway.Results Compared with the control group, the model group showed significant increase in blood glucose, decreased spatial learning and memory capacity, severely damaged hipp-ocampal neurons, increased activated microglia, significantly higher levels of TNF-cx, 1L6, p-JAK2/JAK2 and p-STAT3/STAT3, and signif icantly lower levels of ILK) and BDNF.Compared with the model group, QFY group effectively reduced FBG, inhibited the con-tinuous rise of FBG, improved learning and memory a- bility, improved hippocampal neuronal damage, reduced activated microglia, reduced TNF-cx, 1L6, p- JAK2/JAK2 and p-STAT3/STAT3 levels, and increased ILK) and BDNF levels.Conclusion QFY has been shown to improve type 2 diabetic cognitive impairment , and the mechanism may be related to the inhibition of blood glucose rise and regulation of the JAK2/STAT3 pathway, thereby inhibiting microglia activation.
10.Identification and analysis of the rare HLA-A/B/DRB1 allele genes of 10165 bone marrow registry donors in Shaanxi region.

This study was purposed to analyze the detected status of rare alleles from HLA-A/B/DRB1 typing of 10165 unrelated hematopoietic stem cell donors in Shaanxi region during 2009-2012. The rare allele distributions of HLA-A/B/DRB1 gene typing of 10165 unrelated-donors from Shaanxi sub-registry of Chinese National Marrow Donor Project (CMDP) were detected and analyzed by PCR-SBT. The results showed that there were 40 rare alleles from 48 donors identified by PCR-SBT in 10165 unrelated-donors of Shaanxi sub-registry. Among them, 10 rare alleles of A*02:04, B*07:10, B*27:09, B*35:11, B*44:29, DRB1*03:04, DRB1*08:18, DRB1*13:05, DRB1*13:14 and DRB1*14:11 from 15 donors were not included in the common alleles and well documented alleles (CWD) of China, but were included in the CWD of American Society for Histocompatibility and Immunogenetics (ASHI). The alleles of A*68:24, B*35:11, B*44:29, DRB1*03:04, DRB1*08:18 and DRB1*13:05 were confirmed in more than two samples. There were totally 21 novel HLA alleles identified by our laboratory and officially assigned by the WHO Nomenclature Committee from 2005 to 2012, and some of them were also detected from multiple donors in other HLA typing laboratories of China. Now the novel alleles of HLA-A*02:90, HLA-B*48:14 and HLA-DRB1*01:14 were added into the Chinese CWD list. It is concluded that to ensure the polymorphism integrity and accurate population distribution of HLA genes and its constant accumulations on CMDP, it is necessary to recognize and submit timely the potential novel alleles in our practical work, as well as to record and statistics the identified rare alleles, which can provide the basis for the modification of Chinese CWD. When CWD list is referred, it should be careful for ambiguous results containing the identified rare alleles in order to avoid the occurrence of false or undiscovered detection, and ensure that the patients carrying rare alleles could find a matching donor with the maximum opportunity.
