Journal of Medical Research 2006;0(01):-
Objective To investigate the expression of large-conductance calcium-sensitive potassium channel (BKCa) in myometrium. Methods The expression levels of BKCa ? subunit protein in human myometrium tissues in term of non-labor(TNL) and term labor(TL)were measured by Western blot analysis. Results Relative abundance of BKCa channel ? subunit protein was 1.18+0.20 for TNL group(n=9),and 0.60+0.08 for TL group(n=11).The statistic difference was significant (P=0.009). Conclusion Expression of BKCa channel protein in lower segment myometrium of TNL group was higher than that of TL group. BKCa may play a certain role in modulating uterine function.

Objective Discbuss the hypertension prevalence rate in the Wuhan male taxi drivers and the dangerous factor to provide the basis for its professional publie health work. Methods The Wuhan male taxi drivers were carried on the healthy physical examination who in the Hubei province medicine medical examination center during 2005-2007 and were carried on the epidemiology statistical analysis. Results The Wuhan male taxi drivers hypertension prevalence rate is 28.28%. That is higher than the common crowd. Hypertension leaves outside the heredity factor and be closely correlated with the age of harnesses, the body weight index, and so on. Conclusions The Wuhan male taxi drivers'hypertension prevalence rate is unoptimistic. We should take the synthesis preventing and controlling measure to reduce this crowd hypertension prevalence rate.

Objective_To investigate whether hUCMSCs undergo malignant transformation when exposed to MCF-7B breast cancer microenvironment and whether the abnormal activation and over expression of STAT3 play an impor-tant role in this transformation.Methods_The experiment was divided into three groups:blank group ( hUCMSCs were separately cultured),experimental group(hUCMSCs were indirectly co-cultured with MCF-7B breast cancer cells),positive control group(MCF-7B breast cancer cells were separately cultured).Morphology of cells was detec-ted by invertedmicroscope.Cell cycle was detected by flow cytometry. The mRNA expression of STAT3, c-Myc and Bcl-xL was tested by real-time PCR.The protein expression and location of p-STAT3,c-Myc and Bcl-xL were detected by immunofluorescence. The protein expressions of p-STAT3,STAT3,c-Myc and Bcl-xL were also meas-ured by Western blot.Results_The experimental group cells showed typical morphology of the tumor cells. The cells proportion of experiment group in G1 phase was significantly lower than that of the blank group(P<0.05),but which in S and G2 phase were significantly higher than those of the blank group(P<0.05).The mRNA expression levels of STAT3 ,c-Myc and Bcl-xL in experimental group was significantly higher than those in blank group ( P<0.05).p-STAT3,STAT3,c-Myc and Bcl-xL protein were significantly higher than those of the blank group(P<0.05),and they were mainly located in the nuclei. The protein expression of STATS also showed significant changes in experimental group.Conclusions_hUCMSCs trends to malignant transformations when exposed to MCF-7B breast cancer microenvironment. The abnormal activation and over expression of STAT3 are of important factors leading to the malignant transformation of hUCMSCs.

Aim To study the anti-itching and anti-hypersensitivity effects of Doxepin hydrochloride cream. Methods The effects of Doxepin hydrochloride cream on the pruritus of skin in mice due to 4-AP and guineapigs due to histamine phosphate were observed. The inhibitory actions of it on the increase of abdominal cavity capillary permeability in mice due to acetic acid and the angiectasis of skin in rats due to histamine were also observed. Its effects on the delayed-type hypersensitivity (DTH) of mice due to 2, 4-dinitrochlorobenzol (DNCB) and the homologous passive anaphylaxis of skin in rats were used to investigate its anti-hypersensitivity actions. Results The application of Doxepin hydrochloride cream (50, 125, 250 mg?kg -1) on the skin of mice had significant inhibitory actions on the pruritus of skin caused by 4-AP, and the application of Doxepin hydrochloride cream (10, 25, 50 mg?kg -1) on the skin of guinea pigs also had significant inhibitory actions on the pruritus of skin caused by histamine phosphate. The application of Doxepin hydrochloride cream (10,25,50 mg?kg -1 and 50,125,250 mg?kg -1) on skin inhibited the increase of skin capillary permeability in rats caused by histamine phosphate and the increase of abdominal cavity capillary permeability in mice caused by acetic acid. The application of Doxepin hydrochloride cream (50, 125, 250 mg?kg -1 and 10, 25, 50 mg?kg -1) on skin also significantly inhibited the delayed-type hypersensitivity in mice due to 2, 4-dinitrochlorobenzol and the homogeneous cutaneous anaphylaxis in rats. Conclusions Doxepin hydrochloride cream can relieve itching, inhibit the increase of capillary permeability caused by inflammatory substances. It also has significant inhibitory actions on TypeⅠ and Type Ⅳ allergic reactions.

The paeonol proniosomes ointment and ordinary ointment were administered to rats. Physiological saline served as perfused solution. The perfusion rate was 5 mL x L(-1) and the microdialysis samples were collected every 20 min intervals. The paeonol concentration in perfused solution was determined by HPLC. Investigation of the pharmacokinetics of paeonol proniosomes ointment and ordinary ointment by the skin-blood synchronous microdialysis coupled with HPLC is reported in this study. The results show that the recovery was (54.80 +/- 1.50)% in vitro and (54.58 +/- 4.61)% in vivo. The results showed that paeonol proniosomes ointment significantly raised the drug concentrations in skin more than the paeonol ordinary ointment. The paeono proniosomes ointment has less drugs into the blood as the ordinary ointments in blood, but its blood drug concentrations were steadier. The paeonol proniosomes ointment may be developed into a new preparation.

Journal of Huazhong University of Science and Technology (Medical Sciences) 2014;34(4):491-6
This study aimed to explore Semaphrin4D (Sema4D) expression and clinical significance in non-small cell lung cancer (NSCLC), and to define the roles and mechanisms of Sema4D in regulating the malignant behaviors of A549 cells by small interfering RNA (siRNA). Firstly, immunohistochemistry revealed that Sema4D was more frequently expressed in NSCLC than in lung benign lesion (P<0.05) and its overexprssion was associated with low differentiation (P<0.05), poor pTNM staging (P<0.05) and occurrence of lymph node (LN) metastasis (P<0.05). Endogenous Sema4D expression was suppressed by Sema4D siRNA in A549 cells overexpressing Sema4D. Protein levels of Sema4D, total Akt and p-Akt were examined by Western blotting. Cell proliferation, migration and invasion abilities were measured by MTT assay and Transwell assay respectively. Results showed that Sema4D siRNA significantly suppressed phosphorylation of AKT in A549 cells, but it did not alter total AKT expression. In addition, efficient down-regulation of SemaD significantly inhibit cell proliferation (P<0.05), migration (P<0.05) and invasion (P<0.05) in A549 cells. These findings suggest that Sema4D might serve as a reliable tool for early prediction of NSCLC poor prognosis. Sema4D could play an important role in promoting tumor proliferation, migration and metastasis in the NSCLC, by influencing the Akt protein phosphorylation. Inhibition of Sema4D may be a useful approach for the treatment of NSCLC.
