1.Simultaneous determination of 11 mycotoxins in malt by isotope internal standard-UPLC-MS/MS.

A suitable ultra-high performance liquid chromatography coupled with tandem mass spectrometry (UPLC-MS/MS) method was developed for the determination of 11 mycotoxins with isotope internal standard in malt. The mycotoxins in malt were extracted and purified by one-step ultrasonic extraction procedure using acetonitrile/water/acetic acid (80 : 19 : 1), and then detected and confirmed by UPLC-MS/MS, and quantified by isotope labeled AFB1 ([13C17]-AFB1) and ZEN ([13C18]-ZEN) internal standards. Rapid separation of the 11 mycotoxins was successfully achieved on a Phenomenex Kinetex C18 column (100 mm x 2.1 mm, 2.6 μm) with gradient elution using the mobile phase of methanol containing 0.1% formic acid and 2 mmol x L(-1) ammonium acetate in water. Simultaneous acquisition was performed in multiple reaction monitoring (MRM) mode with electrospray ionization (ESI) source operated in both positive and negative ionization modes. The established method provided a good linearity for the 11 mycotoxins within their respective linear ranges with correlation coefficients all higher than 0.999 1. The average recoveries ranged from 75.0% to 117.0% with relative standard deviations (RSDs) below 5.1%. The limits of detection (LODs) and quantitation (LOQs) ranged from 0.05 to 30 μg x kg(-1) and 0.15 to 87.5 μg x kg(-1), respectively, which were below the maximum residue levels (MRLs) set by the European Union. Twenty malt samples were analyzed and nine samples were detected with mycotoxins, which were confirmed according to the same fragment ions found in positive samples and the standards at the same retention time. This study has demonstrated that the one-step extraction procedure of mycotoxins from complex matrices coupled to UPLC-MS/MS method is simple, quick, accurate and sensitive for quantitative and qualitative analysis of multiple mycotoxins in malt.
2.Diagnosis and treatment of 40 cases of hyperthyroidism in pregnancy

Objective: To study the diagnosis, treatm ent and outcome of infants in patients with hyperthyroidism in pregnancy. Methods: The levels of T3,T4,TSH,FT3,FT4 in 40 cases of hyperthyroid gravida and control group were measured before delivery, the r oute of delivery and the outcome of infants were studied, and the neonatal thyroid f unctions were evaluated by the levels of T3,T4,TSH in cord serum samples . Results: The levels of T3,T4,FT3,FT4 in non-treatm ent group were higher than that of treatment group and control(P<0.01); an i ncreased rate of forceps and cesarean in hyperthyroid gravida was seen compared with that in control group, about 90%(36/40); the body weights of infants in no n-treatment group were lower than that in treatment and control group. There wa s a case of neonatal hyperthyroidism in non-treatment group. Conclusion : Hyperthyroidism should be diagnosed and treated earlier and there is little side effects on the outcome of infants.
3.Effect of Combined Acupuncture-medication Anesthesia on Visceral Function in Laparoscopic Cholecystectomy

ObjectiveTo observe the effect of combined acupuncture-medication anesthesia on visceral function of patients undergone laparoscopic cholecystectomy, and to furtherexplore its clinical and theoretical significance.MethodSixty patients who were going to receive laparoscopic cholecystectomy were enrolled and randomized into an observation group (combined acupuncture-medication anesthesia) and a control group (conventional general anesthesia) by the random number table, 30 subjects in each group. The total bilirubin (TBIL), alanine aminotransferase (ALT), aspertate aminotransferase (AST), urea nitrogen, creatinine, and Mini-Mental State Examination (MMSE) were observed to evaluate the effect of combined acupuncture-medication anesthesia on visceral function of patients undergone laparoscopic cholecystectomy.ResultBefore operation, there were no significant differences in TBIL, ALT, urea nitrogen, creatinine, and MMSE between the observation group and the control group (P>0.05), while the AST level of the observation group was significantly higher than that of the control group before operation(P<0.05). On post-operation day 1, there were no significant differencesin comparing ALT and AST levels between the two groups (P>0.05); the TBIL level in the observation group was significantly lower than that in the control group (P<0.01); there were no significant differences in comparing the urea nitrogen and creatinine levels between the two groups (P>0.05). The MMSE score of the observation group was significantly higher than that of the control group 6 h after the operation (P<0.01). on post-operation day 1, there was no significant difference in comparing MMSE score between the two groups (P>0.05).ConclusionThe combined acupuncture-medication anesthesia and conventional general anesthesia play equal effects on liver and kidney functions in laparoscopic cholecystectomy, thus the two anesthesia methods are both safe andeffective. The combined acupuncture-medication anesthesia has certain protective effects on brain function in early stage, and possibly produces certain protective effectson liver function.
4.An Analysis on the Mental Health of Adolescent with Cochlear Implants

Objective To analyse the mental health of adolescents with cochlear implants and provide the ref‐erence for programming the personalize rehabilitation schedule of audio - speech -psychological .Methods The mental health test(MHT) was used to evaluate the mental health status of 60 adolescent with cochlear implants ,and compared with those of adolescent with normal hearing .Results The incidence of psychological problems was 3 .33% in adolescent with cochlear implants .The hypersusceptilility scores of the group of cochlear implants (5 .63 ± 2 .22) was higher than that of in normal hearing group(4 .05) in ,which was significantly different(P<0 .01) . The body syndrome scores in the cochlear implants group (4 .03 ± 2 .72) was lower than that of in normal hearing group(4 .99) ,which was significantly different(P<0 .01) .The phobic tendency scores in cochlear implants group (1 .95 ± 2 .01)was significantly lower than that of in normal hearing group (3 .06)(P<0 .01) .Conclusion The over‐all mental health of adolescent with cochlear implants is good .Cochlear implants can help them improve hearing and speech function ,and improve quality of life ,but they still need parents ,teachers and the community to pay more at‐tention and care to help them get through adolescence ,and return to mainstream society .
5.The Mental Health and Affecting Factors on Cochlear Implanted Children

Objective To investigate the mental health and the affecting factors on children cochlear implants and to provide the reference of comprehensive rehabilitation for them.Methods We used the Psychological Ques-tionnaire for 3~6 Years Children to evaluate the mental health status of 1 6 6 children of 3~4 years old with cochlear implants,and compare those with the standard values of children with normal hearing.We studied the affecting fac-tors on their psychological development.Results The pass rates in action,emotion and will,cognitive ability,sociali-ty,living habits and speech ability of children with cochlear implants decreased as their age increased.The scores of action,emotion and will,cognitive ability,sociality,living habits and speech ability of children with cochlear im-plants increased as the use time increased.The affecting factors on the psychological development of children with cochlear implants were the time length of usage and their residences before operation.Conclusion There were differ-ences between the children with cochlear implants and normal hearing in psychological development.We should fo-cus on their development of hearing and speech ability,and the psychological development.

7.The research of rehabilitation effect of cochlear implantation for deaf children with gene mutation.

OBJECTIVE:
To assess the evaluation on auditory rehabilitation effect for 42 deaf children with GJB2 gene mutation after cochlear implantation to provide a reference for the cochlear implant effect evaluation of such patients.
METHOD:
To conduct the detection on common genetic deafness gene mutation hotspots of hearing impaired children with cochlear implantation. To conduct auditory rehabilitation effect evaluation on 42 cases of patients with GJB2 genetic deafness after 3 months, 6 months and 12 months of the operation respectively. The single factor repeated measure ANOVA was applied to analyze whether there were significant difference among the results of initial consonant of a Chinese syllable recognition at 3 different stages after the operation, the results of vowel of a Chinese syllable recognition at 3 different stages after the operation, and the results of two-syllable recognition at 3 different stages after the operation.
RESULT:
235delC is the high-incidence mutational site in 42 cases of patients with GJB2 genetic deafness, the total detection rate is up to 90.48%. There were significant differences in the initial consonant of a Chinese syllable recognition rate, the vowel of a Chinese syllable recognition rate, the two-syllable recognition rate as well as the vowel of a Chinese syllable recognition rate after 3 months, 6 months and 12 months of the operation (P < 0.01).
CONCLUSION
Cochlear implantation is a safe and effective measure for auditory reconstruction, it can help patients with GJB2 hereditary severe sensorineural deafness to improve auditory speech recognition.

9.Shikonin Suppresses Osteoclastogenesis in Vitro and Ameliorates Ovariectomized-induced Osteoporosis in Mouse Model

Objective:To investigate the effects of shikonin on osteoclastogenesis in vitro and amelioration of bone loss in ovariectomized-induced osteoporosis in mouse model.Methods:The optimal concentration of shikonin treating were evaluated in vitro depending on its effect on the viability of C57BL/6J mouse bone-marrow-derived macrophages by CCK-8 method.To establish the osteoclastogenesis cell model,macrophages were cultured with RANKL and M-CSF treatment,and TRAP staining was used to observe the generation of osteoclasts after treating with different concentration of shikonin solution.Expressions of osteoclast marker genes,including TRAP,c-Fos and NFATclwere detected with real-time PCR.Fifthteen mice were randomly allocated into sham operation group,ovariectomized model group and shikonin treatment group.After the modeling,mice in treatment group were received the intraperitoneal injection of shikonin,while the other two groups treated with normal saline.After thirty days treatments,all animals' tibias were dissected for micro-CT analysis.Results:①The macrophages viability was significantly inhibited when the concentration of shikonin was higher than 250 nmol/(P<0.01).②The osteoclastogenesis was significantly suppressed by differemt dose of shikonin(P<0.01).③ The expression of the osteoclastic marker genes (TRAP,c-Fos and NFATc 1) were suppressed by addition of shikonin comparing to control group (P<0.01).④ Shikonin effectively prevented ovariectomy-induced bone loss (P<0.05).Conclusion:Shikonin suppresses osteoclastogenesis in vitro and ameliorates ovariectomized-induced osteoporosis in mouse model.
10.Effects of Pre-implant Hearing Aid Fitting on the Open-set Word Recognition in Children with Cochlear Implants during Infants

Objective To explore the impact of pre-implant hearing aid fitting on the early open-set word recognition in children who received a cochlear implant(CI) when they were infants .Methods A total of 289 chil‐dren who received a cochlear implant between 0 .9 and 3 .0 years of age were included in this study .According to pre-implant hearing aid fitting ,participants were divided into hearing aid group and non-hearing aid group .The open-set word identification abilities of the cochlear implant children were evaluated via Mandarin Lexical Neighborhood test(M -LNT) .Results Comparisons of the scores of M -LNT between two groups displayed no significant differ‐ence at the same identification age or duration of CI use .Conclusion The abilities of the early open-set word rec‐ognition between hearing aid group and non-hearing aid group didn’t make significant differences for the CI chil‐dren w ho received a cochlear implant betw een 0 .9 and 3 .0 years old .
