2.Risk Factors and Hygiene Hypothesis of Inflammatory Bowel Disease
Chinese Journal of Gastroenterology 2014;(8):496-499
Till now,the etiology of inflammatory bowel disease( IBD)is still not very clear. Some genetic risk loci have been identified that predispose people to IBD,however, they increase the risk of IBD by only a small magnitude. Therefore, environmental risk factors have been the focus of recent researches. This article reviewed the association of environmental factors( hygiene and dietary factors etc. ),especially childhood hygiene with IBD,and concluded that rural environment, higher number of siblings and having pets decreased the risk of IBD,while urban environment and small household size/sibship were risk factors for IBD. Currently,population-based study focusing on hygiene and IBD is deficient domestically, further epidemiological surveys are warranted to confirm their associations.
3.Stability of femoral prosthesis after total hip replacement
Chinese Journal of Tissue Engineering Research 2015;(39):6281-6285
BACKGROUND:There are many experimental studies about compression mechanics of femur after hip replacement at home and aboard. Therefore, it is very important to study the torque, torsion angle, load-displacement relationship of femur after hip replacement. Comparing and analyzing the properties of compression and torsional mechanics of traditional-type prosthesis and anatomical-type prosthesis is of important significance for studying hip replacement and the stability of artificial prosthesis.
OBJECTIVE:To compare and analyze the stability of traditional-type prosthesis and anatomical-type prosthesis by stimulating femoral axial compression and torsion tests after hip replacement, so as to provide biomechanical parameters for clinic practice.
METHODS: Twelve specimens of left- and/or right-side femur were selected. The femoral necks of 6 left-side femur specimens were retained as the anatomical-type titanium artificial joint prosthesis group, and the femoral necks of the 6 right-side femur specimens were removed as the traditional-type cobalt-chromium-molybdenum artificial joint prosthesis group. The femur specimens in these two groups were respectively placed onto the electronic universal testing machine workbench and were imposed compression stress at the experimental velocity of 5 mm/min. The corresponding displacement values were read under the force of 20, 40, 60, 80 and 100 N. And then, both ends of the femur specimens from these two groups were placed within the chuck of torsion testing machine, and were imposed torque at the experimental velocity of 1(°)/s. The corresponding torsion angle values were read under the torque force of 5, 10, 15 and 20 N?m.
RESULTS AND CONCLUSION: Under the external force of 100 N, the displacement was (2.03±0.06) mm in the traditional-type prosthesis group, and (1.83±0.05) mm in the anatomical-type prosthesis group. Under the torque force of 20 N?m, the torsion angle values of traditional-type prosthesis was (21.7±0.7)°, and that of anatomical-type prosthesis was (13.2±0.4)°. The displacement under the external force of 100 N and the torsion angle values under the torque force of 20 N?m in the anatomical-type prosthesis group were al significantly less than those in the traditional-type prosthesis group (P < 0.05). These results suggest that traditional-type and anatomical-type prostheses have different compression and torsion mechanical properties. The anatomical-type femur prosthesis has a better stability than traditional-type femur prosthesis.

OBJECTIVETo investigate diagnostic efficacy of transvaginal three-dimensional hysterosalpingo-contrast sonography (3D-HyCoSy) in assessing tubal patency with chromolaporoscopy.
METHODSA total of 157 infertile women underwent 3D-HyCoSy to evaluate tubal patency. Among these patients, 39 patients were also examined by chromolaporoscopy. The concordance of the two clinical assessment methods was analyzed by the Kappa coefficient test.
RESULTSAmong the 306 oviducts examined by 3D-HyCoSy, 99 (32.4%) were patent, 126 (41.2%) partially obstructed, and 81 (26.5%) completely obstructed. Diagnostic results with 3D-HyCoSy were not statistically different from those obtained in the 39 women (78 oviducts) who also underwent chromolaporoscopy, and the two methods showed a high concordance (k=0.747, P=0.000). The 3D-HyCoSy procedure had a sensitivity of 84.8% (28/33), a specificity of 96.2% (25/26), and positive and negative predictive values of 93.3% (28/30) and 86.2% (25/29) respectively.
CONCLUSIONTransvaginal 3D-HyCoSy can accurately reveal the spatial path and morphology of the oviduct and is a safe and effective method to evaluate tubal patency.

BACKGROUND:Cytotrophoblasts in placental cell components plays an important role in fetal immunological tolerance.Placental mesenchymal stem cells(pMSCs)have potential of multiple differentiation and inhibition of lymphocyte proliferation. However, conventional methods cannot acquire a large amount of purified human cytotrophoblasts or pMSCs.OBJECTIVE:To establish a method to obtain large placenta tissue, and harvest plenty of cytotrophoblasts and pMSCs with high purity and activity.METHODS:Human placenta tissues were dissected, minced, and dissociated in trypsin and DNAse I.The dissociation was performed in three stages of incubation at 180 r/min for 20 minutes at 37 ℃ The digesting suspension was filtered using a 200 mesh strainer before separated by Percoll gradients. The cytotrophoblast cells and pMSCs fractions were collected respectively.Fibroblasts of cytotrophoblast cells fraction were removed by differential adhesion. The pMSCs were seeds on 75-cm2 flask directly for culture. The dissociation of placenta tissue was observed. The number of harvesting cytotrophoblasts was quantified and Cytokeratin 7 expression was tested. The pMSCs primary culture time, cell passage, induced osteoblast differentiation were observed. The cell surface makers were also detected.RESULTS AND CONCLUSION:After digesting in trypsin and DNAse I,there was only little residue left.(5.48±1,98)×10~8 cytotrophoblasts were obtained after differential adhesion.(90±4.36)% of these cells were positive for Cytokeratin 7.At 19-21 days after pMSCs reached approximately 90% confluency, the cell number was(1.96±0.24)×10~6.The subcultre cells could be passaged again in 4 or 5 days.Flow cytometric analysis of pMSCs showed that the cells expressed CD29,CD44 and HLA-ABC intensively and were negative for CD34,CD45,CD14 and HLA-DR.pMSCs differentiated into osteoblast-like cells after induction, which stained bright salmon pink by Alizarin Red.Dissociating the placenta tissue in trypsin and DNAse I in combination with discontinuous Percoll gradient separation obtained a large number of cytotrophoblasts and pMSCs recovered from placenta tissue, with high purity and activity.
