1.Survey of first permanent molars of 7 - 12-year-old students in Huairou district, Beijing in 2010

The first permanent molar caries and current status of sealant of 7 - 12-year-old students were surveyed in Huairou district of Beijing.Based on the World Health Organization and national oral health survey standards,cluster and stratified random sampling methods were used.The prevalence rate of deciduous tooth caries was 17.34% with a decayed/missing/filled teeth (dmft) index of 0.30.The caries rate of females was higher than that of males (20.60% vs.14.08% ).The incidence of mandible was higher than that of maxillary.And there was no significant difference around the jaw.The rate of fissure sealant was 51.36%.The current status of first permanent molar caries in this district was in accordance with the characteristics of general epidemiological caries disease.An overall policy of early discovery,early diagnosis and early treatment should be practiced.
2.Heterogenous acellular dermal matrix seeded with adipose derived stem cells for urethral reconstruction in a rabbit model

Objective To assess the feasibility of heterogeneous acellular dermal matrix(ADM)seeded with adipose derived stem cells(ADSC)for urethroplasty in a rabbit model.Methods ADSC were isolated from a rabbit and expanded in vitro,then identified by flow cytometry.We seeded ADSC onto the ADM and made it into tissue-engineered urethra.12 male rabbits were removed 1 cm urethra and divided into experiment group and control group.There were 6 rabbits in each group.Reconstructed urethra with tissueengineered urethra was used in experiment group,while unseeded ADM were used in control group.Urethrography was performed at 6 months after surgery.The animals were scarified at 3 and 6 months after surgery and the repaired urethra were harvested.H&E staining and immunohistochemical staining were performed with cytokeratin AE1/AE3 and smooth muscle desmin makers.Results The morphology of isolated ADSC was with long spindle cross-links,and had multicentral growth.Flow cytometry showed that the ADSC expressed CD166,CD105,CD90 and CD44,but not expressed CD45 and CD13.The cells could growth well on the ADM and showed good biocompatibility with it.All animals could void normally,urethrography showed there was no significant stenosis.3 months after surgery,the experiment group appeared regenerated smooth muscle but not in the control group,both groups did not regenerate urothelium.At 6 months urothelium and smooth muscle cells could be observed in the experiment group,but only the smooth muscle was evident in the control group.Conclusions By applying tissue engineering methods,we can seed the ADSC onto the heterogeneous ADM and make it into tissue-engineered urethra,which can help improve the reconstructive effect of urethra.
3.Awareness and administration of antidiabetic drugs in patients with diabetes in Huairou district of Beijing

Objective To investigate the awareness and administration of antidiabetic drugs in patients with diabetes in Huairou district of Beijing.Methods A questionnaire survey on knowledge and administration of antidiabetic drugs were conducted in 900 diabetic patients from 16 communities in Huairou district of Beijing from November 2014 to February 2015.The related factors influencing antidiabetic drug use were analyzed with logistic regression method.Results A total of 899 questionnaires were recovered. Results showed that the rate of metformin single use was the highest (23.7%, 213/899), the rate of combination use of metformin and acarbose was 11.8%(106/899), and that of metformin and glipizide was 17.1% (154/899).Survey showed that 65.9% (592/899) patients were compliant immediately for antidiabetic agents, 32.2% (290/899) were not immediately compliant, 1.9% (17/899) did not use antidiabetic agents.Logistic regression analysis showed that the factors affecting the use of antidiabetic agents were the types of medication (t=-4.351, P=0.000) , adverse effects of drugs (t=6.999, P=0.000), awareness of diabetes to health hazard (t=-10.969, P=0.000) and the doctor′s recommendation (t=-3.352, P=0.001).Conclusion The results showed that the administration of antidiabetic drugs in diabetic patients from Huairou district is basically followed the recommendation by 2013 edition of type 2 diabetes guidelines and the compliance rate of medication need to be further raised.
4.Effect of acupuncture along affected meridian on the MME gene expression of migraine patients without aura of gan-yang hyperactivity syndrome.

OBJECTIVETo observe the effect of acupuncture along affected meridian on the mem- brane metallo-endopeptidase (MME) gene expression of migraine patients without aura (MO) of Gan-yang hyperactivity syndrome (GYHS).
METHODSTotally 20 MO patients of GYHS were randomly assigned to the acupoint group (acupuncture along affected meridian) and the non-acupoint group, 10 cases in each group. Needling was performed once per day for 10 consecutive days. Gene chip technology was used to obtain two sets of gene expression profiles and analyzed using Gene Ontology (GO).
RESULTSIn the acupoint group, MME gene expression decreased after needling (P = 0.0023).That gene was rich in the beta-amyloid metabolic process (P = 3.16E-05) and the peptide metabolic process (P = 0.009612). Its expression was not seen in the non-acupoint group.
CONCLUSIONThe effect of point selection along affected meridian could be achieved possibly by regulating the MME gene expression.

5. Effects of endothelin-1 on the expression of ClC-3 chloride channel in bovine cerebrovascular smooth muscle cells

AIM: To study the effects of endothelin-1 (ET-1) on ClC-3 chloride channel protein expression in cultured bovine cerebrovascular smooth muscle cells (CSMC). METHODS: Cell culture and Western blot. RESULTS: 1 The endogenous ClC-3 expression was found in basilar artery, middle cerebral artery, and microvessel; 2 The molecular weight of expressed ClC-3 chloride channel protein was about 95 ku; 3 ET-1 enhanced ClC-3 protein expression which was inhibited by nifedipine and SK&F96365. Cyclopiazonic acid(CPA) increased the expression of ClC-3 protein in a concentration-dependant manner, and enhanced ET-1 effect on this protein expression. CONCLUSION: ET-1 stimulated ClC-3 chloride channel protein expression in cultured bovine cerebrovascular smooth muscle cells. The intracellular Ca2+ plays an important role on signal transduction pathway in ClC-3 protein expression process.

Objective:To study the antisense oligonucleotide mediated inhibition on telomerase activity and cell proliferation of GBC-SD cell.Methods:We design the antisense,sense,and random oligonucleotide with phosphoric acid modification for the hTR(Human Telomerase RNA)template sequence.MTT and PCR methods were used to observe the inhibition on telomerase activity and cell proliferation of GBC-SD cell ,and fibroblast cells were used as control group.Results:PS-ODN can lead to the reduction of cell survival rate of GBC-SD cell,wich dosage dependence.Tne experimental group cell detected by scanning electron appeared apoptotic feature.Conclusion:PS-ODN can inhibit telomerase activity of GBC-SD cell effectively and induce the cell apoptosis.

Objective To establish a comprehensive method combining physical model experiment and numerical simulation for studying airflow state of upper respiratory tract. Methods Based on CT medical images published online, a three-dimensional (3D) model of human upper respiratory tract was reconstructed. Based on 3D printing technology, an experimental model of the upper respiratory tract was established and the flow process of respiration was measured. A numerical simulation model was created based on the meshing of upper respiratory tract model and the turbulent Realizable k-ε model. Results Firstly, the result of numerical simulation was compared with the experimental conditions, and good agreement was achieved. The numerical simulation results showed that the airflow in respiratory process was in a parabolic shape; the distribution of flow field, pressure on wall and vortex structure were different between inspiratory and expiratory phases; there were air residues in the upper and lower nasal passages during the respiratory exchange process. In addition, the effects of airflow on physiological environment of the upper respiratory tract were preliminarily analyzed through the steak line, pressure field and vortex structure distribution. Conclusions The method proposed in this paper has the characteristics of pertinence, rapidity and accuracy, which gives full play to the advantages of reliable physical experiments and fine numerical simulation, and is applicable for studying different problems of the upper respiratory tract in different cases, with a high value for personalized diagnosis and treatment in clinic.
10.Risk Factors for in-Hospital Mortality in Patients with Takotsubo Syndrome

ObjectiveTo analyze the risk factors for in-hospital mortality in patients with Takotsubo Syndrome (TTS). MethodsHospitalization data of consecutive patients with TTS from February 2009 to January 2022 were retrospectively collected and analyzed. Patients were divided into survival group and death group according to outcomes. The basic clinical information, triggering factors, laboratory examinations, electrocardiogram, echocardiography, complications and treatments of the two groups were compared. Univariable logistic regression analysis was used to screen the possible risk factors for in-hospital mortality in TTS patients, and multivariable logistic regression analysis was used to determine the independent risk factors for in-hospital mortality in TTS patients. ResultsA total of 62 TTS patients were included in our study, including 21 males (33.9%), 41 females (66.1%) and 26 postmenopausal women (41.9%), with the mean age of (55.6±16.2) years, and physical triggers were found in 50 patients (80.6%). 17 patients (27.4%) died while 45 patients (72.6%) survived during hospitalization. The death group had lower systolic blood pressure and left ventricular ejection fraction (LVEF), higher incidence rate of syncope, higher level of N-terminal pro-B natriuretic peptide (NT-proBNP) and hypersensitive troponin T (hs TnT) when compared with survival group (all P value <0.05). As for the triggering factors, the proportion of TTS induced by neurologic disorders in the death group was higher than that in the survival group (P<0.05). The death group had higher rates of cardiogenic shock, malignant ventricular arrhythmia, atrial fibrillation, and respiratory failure (all P value <0.05). Compared with the survival group, therapeutic dopamine, therapeutic norepinephrine, hemodialysis and mechanical ventilation were higher in the death group (all P value <0.05). Univariable logistic regression analysis suggested that syncope, NT-proBNP, LVEF, neurologic disorders, cardiogenic shock, malignant ventricular arrhythmia, atrial fibrillation, respiratory failure, therapeutic dopamine, therapeutic norepinephrine, hemodialysis and mechanical ventilation were potential risk factors for in-hospital mortality in TTS patients (all P value <0.05). Multivariable logistic regression analysis indicated that neurologic disorders [OR(95%CI)=5.651(1.195,26.715),P=0.029], atrial fibrillation [OR(95%CI)=6.217(1.276,30.298), P=0.024)] and therapeutic norepinephrin [OR(95%CI)=8.847(1.912,40.949), P=0.005] were independent risk factors for in-hospital mortality in TTS patients. ConclusionsNeurologic disorders, atrial fibrillation and therapeutic norepinephrin are independent risk factors for in-hospital mortality in patients with Takotsubo Syndrome. Clinically, attention should be paid to the prevention and treatment of neurologic disorders and atrial fibrillation; norepinephrine should be carefully used in patients with diagnosed TTS complicated with hemodynamic instability.
