ObjectiveTo explore the mechanism of hypericin against liver cancer using network pharmacology. MethodThe traditional Chinese medicine systems pharmacology database and analysis platform (TCMSP), Drug Gene Interaction Database (DGIdb), Comparative Toxicogenomics Database (CTD) and SwissTargetPrediction were used to predict the targets of hypericin. Five databases including GeneCards and Online Mendelian Inheritance in Man (OMIM) were employed to obtain liver cancer-related targets. The intersection was performed to obtain the targets of hypericin against liver cancer. The Database for Annotation, Visualization and Integrated Discovery (DAVID) v2021q4 was used for Gene Ontology (GO) function annotation and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis. The protein-protein interaction (PPI) network of the targets was constructed by Cytoscape 3.6.1 to screen the core targets, and the affinity between hypericin and the core targets was verified by molecular docking. The effects of hypericin on liver cancer and the migration of liver cancer cells were observed by cell viability assay and would healing assay, respectively, and its effects on the mRNA and protein expression of key targets cysteinyl aspartate-specific protease-3(Caspase-3) and mitogen-activated protein kinase 3 (MAPK3) were detected by real-time polymerase chain reaction(Real-time PCR) and Western blot, respectively. ResultA total of 45 genes related to the anti-liver cancer effect of hypericin were obtained, and six core target genes were screened. The signal pathways enriched by KEGG pathway analysis included apoptosis, tumor necrosis factor (TNF) and cancer signal pathways. Molecular docking showed that the core target genes Caspase-3,TNF,estrogen receptor 1 (ESR1),MAPK3,catalase (CAT) and cyclooxygenase 2 (PTGS2) had good affinity with hypericin, especially Caspase-3 and MAPK3. In addition, compared with the conditions in control group, cell experiments demonstrated that hypericin could reduce the viability of liver cancer cells (P<0.05),inhibit their migration, increase the mRNA expression of Caspase-3 (P<0.05) and decrease that of MAPK3 (P<0.05). ConclusionHypericin exerted the anti-liver cancer effect by affecting the core targets such as Caspase-3,TNF,ESR1,MAPK3,CAT and PTGS2 and jointly interfering with apoptosis,TNF and cancer signal pathways.

Objective To investigate the effect of acitretin on the histopathology and ultrastructure of lesions from patients with bullous ichthyosiform erythrodermia (BIE), and to explore mechanisms underlying the modulation of keratinization process by acitretin. Methods Lesional tissue was obtained from the back of 4 patients with BIE before and after the treatment with acitretin. Light microscopy and transmission electron microscopy were performed to observe histopathological and ultrastructural changes in these lesions. Results After treatment, the improvement in clinical manifestations was more than 75% in all the 4 patients, and reached 90% in 1 of the 4 patients. As histopathology and ultrastructural study showed, there was an obvious improvement in hyperkeratosis and continuity of extra cellular lamellar membrane, and a decrease in keratin deposition in prickle and granular layer, but no remarkable changes were observed for the proliferation of prickle cells or acantholysis. Conclusions Acitretin shows a favorable efficacy in clinical treatment of BIE,with histopathological and ultrastructural improvement mainly located in the stratum corneum. The modulation of keratinization process in keratinocytes by acitretin appears more apparent in granular and corneum layers.

In this study, we conducted an investigation among medical workers, patients and college students concerning their acceptability of breast palpation performed by male doctors (hereinafter referred to as "acceptability", or "the examination", respectively, if not otherwise indicated), to get the information about their acceptability and reasons for accepting or declining the examination among the three population. A questionnaire investigation was conducted in 500 patients with breast diseases, 700 students of medical colleges, and 280 medical workers working in hospitals. The subjects were asked to choose between two options: accept or do not accept (the examination). The subjects were asked to fill out the questionnaire forms on free and anonymous basis and the questionnaire forms were collected on spot, immediately after completion. The questionnaires collected were coded, sorted out and checked. Data of the eligible questionnaires were input into Epidata software and analyzed by SPSS. Upon the establishment of the database, the intra-group data were tested by utilizing χ(2) test. Among 1480 questionnaires, 1293 (90.41%) questionnaires were retrieved. Our results showed that 56.78% of patients reported that they could accept breast palpation by male doctors. About 59.66% of medical staff expressed their acceptance of the examination, but only 35.03% of students said the examination. On the basis of this study, we were led to conclude that the examination is not well accepted by different populations, and therefore, (1) medical professionals and administrators should pay attention to the gender-related ethics in their practice and the feeling of patients should be respected when medical examinations involve private or sensitive body parts; (2) to this end, related departments should be properly staffed with doctors of both sexes, and this is especially true of the departments involving the examination or treatment of private or sensitive body parts; (3) health education should, among other things, include helping female patients to overcome the fear and anxiety in such examinations. This is of great importance since some women may miss the opportunity to get timely diagnosis.
