1.Monitoring Analysis on Medication Orders for Clinical Inpatients by PASS

OBJECTIVE:To evaluate the performances of prescription automatic screening system(PASS).METHODS:40101medication orders of616inpatients in14departments of our hospital were monitored retrospectively by using“Army Health No.1”Hospital Information System Pharmaceutical Workstation that equipped with PASS.RESULTS:There were835irrational drug use medication orders and the incidences of irrational drug use stood at2.08%.Of which,91.02%involved adverse drug interactions,4.55%involved irrational in vitro injection combinations and4.43%were irrational use of drugs for children.CONCLUSION:PASS system is conducive to rational drug use in clinics,yet it is still for from being perfect thus further improvement is needed.
2.Rapid Quantitation of Lamivudine in Mouse Serum and Tissue Homogenates by RP-HPLC

OBJECTIVE:To develop a rapid RP-HPLC quantitative determination of lamivudine(LAM) in mouse serum and tissue homogenates METHODS:0 1ml of serum of tissue homogenates and 0 5ml of methanol were vortexed in a plastic tube for 30s and the mixture was then centrifuged at 15 000r/min for 10min 20?l of the resulting supernatant was directly injected and LAM was chromatographed on a reversed-phase Kromasil C column(150mm?4 6mm,5?m) using a mixture of phosphate buffer(pH6 8)and methanol(91∶9,v/v)and monitored at 270nm RESULTS:The standard curves were linear over a range from 0 25?g/ml to 50?g/ml The minimal detectable drug concentration was 60ng/ml(S/N=3) The average extraction recovery of LAM was more than 88% The average methodological recoveries of LAM ranged from 97% to 100% The within-dayand between-day precisions were less than 10% CONCLUSION:The assay is economic,simple,rapid and specific
3.Concentration Determination of Floxuridine in Serum and Organs of Mice by RP-HPLC

OBJECTIVE: To establish RP-HPLC method for the fast determination of floxuridinein in serum and homogenate of some of the organs of mice. METHODS: The determination was performed on C18 chromatographic column with its mobile phase consisted of phosphate buffer(pH= 7.4) - methanol(95∶5) and with its detection wavelength at 268nm. RESULTS: Linear detection concentration ranges of floxuridinein were all at 15~240?g/ml. The minimum detection concentration was 3?g/mL(S/N≥3). The average extraction recovery was above 95%. The average recovery ranged from 91%to 112%. Both the intra-day and inter-day precisions were less than 10%. CONCLUSION: The method is simple, rapid, reliable and economical, and it can be used for the determination of serum floxuridinein concentration and the study of its body distribution in animals.
4.Optimization of the Preparation Techniques of Floxuridinyl Diacetate Solid Lipid Nanoparticles by Orthogonal Test

OBJECTIVE:To optimize the preparation techniques for floxuridinyl diacetate solid lipid nanoparticles(FUDRA-SLN).METHODS:The FUDRA-SLN was prepared by film-ultrasound dispersion method with soybean phos?pholipids as carrier,and the preparation techniques were optimized by means of an orthogonal test design with entrapment ef?ficiency and appearance as indexes of investigation.RESULTS:The FUDRA-SLN prepared in the optimized techniques was even and regular in appearances,with particle diameter at(215?83)nm,entrapment efficiency at98.27%and drug loading dosage at8.20%.CONCLUSIONS:FUDRA-SLN prepared by the optimized film-ultrasound dispersion method has a high entrapment efficiency and high drug loading dosage,and this method is suitable for the laboratory preparation.
5.Determination of Resveratrol in Serum and Organs of Mice by RP-HPLC

OBJECTIVE: To develop RP-HPLC method for the rapid determination of resveratrol in serum and tissue homogenates (in some organs) of mice.METHODS:HPLC determination was performed using Kromasil C18 column,the mobile phase consisted of methanol - water (48∶ 52) with a flow rate of 1ml/min,the column was under room temperature with its pressure at 12kPa,the detection wavelength was 303nm and the sample size was 20?l. RESULTS: The linear range for resveratrol was 0.25~25?g/ml, with minimal detectable drug concentration at 0.1?g/ml (S/N=3). The average absolute rec_overy of the samples was above 88% and the methodological recovery rates of which ranged from 96% to 100%,RSD were below 10%. CONCLUSIONS: The established determination method is economical, reliable, simple and rapid, and suitable for the blood concentration determination of resveratrol and the study of its distribution in animal body.
6.ITS Sequence analysis of Chinese and Japanese medicinal plants of Angelica L.

Objective To establish the base of molecular system-theory on medicinal plants of Angelica L.and to offer molecular gists for finding the efficacy difference of them.Methods The PCR technique was utilized together with the clone technique to get the ITS gene and measure its genic sequence.Statistical analyzing and branch analyzing were carried out by Phylip software and genetic distance difference in every kinds can be calculated with MEGA3.Results ITS1-5.8 S-ITS2,belongs to one of medicinal plants of Angelica L.,its sequence length extents 600 to 629 bp.Subtracting 5.8 S,its length is 165 bp,the length of ITS is 468 bp.The medicinal plants of Angelica L.are divided into two groups by dendrogram.The genetic distance of A.sinensis from Gansu Province is obviously different from the other medicinal plants of Angelica L.,but the distance difference compared with Levisicum officinale is smaller.Conclusion It suggests that the above result be identical to the efficacy of medicinal plants of Angelica L.It may be an important reason of explaining why the efficacy between A.sinensis and other medicinal plants of Angelica L.is so different.
7.Advances in PI3Kδ selective inhibitors

Phosphoinositide 3-kinase(PI3K), a key regulator in the PI3K/AKT/mTOR signaling pathway, plays a critical role in regulation of cell growth, differentiation, metastasis apoptosis, and angiogenesis processes. Abnormal activation of PI3K is closely related to the development of many types of human cancers. PI3Kδ is an important isoform of PI3K family, mainly expressed in leukocytes, which makes it an attractive target for the treatment of hematological malignancies. Great efforts in both industry and academia have been made to develop PI3Kδ selective inhibitors as anti-cancer agents. This review attempts to provide an overview of recent advances in the research of PI3Kδ selective inhibitors, fueling the progress of the ideal selective PI3Kδ inhibitors in the future.

DNA marked-assisted selection of medicinal plants accelerated the breeding and promotion of new cultivars, and guaranteed the healthy development of Chinese medicinal materials industry. The first disease-resistant cultivar of notoginseng, namely "Miaoxiang Kangqi 1", served as the object of study. We evaluated the Kangqi's resistance of seeds, seedlings and root against the pathological bacteria (Fusarum oxysporum) of root rot. Compared to the traditional cultivars, the disease index of notoginseng seeds declined by 52.0% after inoculation for seven days; the death rate of seedlings and disease index of root respectively decreased by 72.1% and 62.4% after inoculation for 25 days. Additionally, the growth inhibition ratio of notoginseng seeds and seedlings declined after inoculation. The seeds, seedlings and roots of "Miaoxiang Kangqi 1" showed significantly resistant to root rot. The evaluation of disease-resistance of Kangqi provided the basis for the popularization of new cultivar and guaranteed the favoring conduct of notoginseng pollution-free cultivation.
9.Pharmacokinetics and brain distribution differences of baicalin in rat underlying the effect of Panax notoginsenosides after intravenous administration.

Baicalin (BA) is the most well-known flavonoid present in Radix Scutellariae. The aim of this study was to explore whether the pharmacokinetic behavior of BA in rat brain can be affected by Panax notoginsenosides (PNS), and to assess the possible mechanism for the observed effects. Specific HPLC and HPLC/MS/MS methods were developed and validated for the determination of BA in the rat plasma and brain using carbamazepine as an internal standard. BA was found to enter rat brain quickly after a single intravenous dose. When co-administered with PNS, clearance (CL) of BA from rat plasma decreased by 50.00%, while the area under the curve AUC0-t and AUC0-∞ increased 94.69% and 87.68%, respectively. On the other hand, some pharmacokinetic parameters of BA in rat brain had obvious differences after PNS was administered, such as an increase in Tmax from 5 min to 15 min, an increase in AUC0-t and AUC0-∞ by 42.75% and 29.39%, respectively, as well as a decrease in CL by 27.95%. Together, these results indicate that PNS can decrease the elimination rate of BA from rat plasma, promote the penetration of BA into rat brain, increase the concentration and slow down the elimination of BA from rat brain. The data provide important information that compatibility with PNS can promote the consequent effects of BA for the treatment of encephalopathy.

DNA marker-assisted selection of medicinal plants is based on the DNA polymorphism, selects the DNA sequences related to the phenotypes such as high yields, superior quality, stress-resistance and so on according to the technologies of molecular hybridization, polymerase chain reaction and high-throughput sequencing, and assists the breeding of new cultivars. This study bred the first disease-resistant cultivar of notoginseng "Miaoxiang Kangqi 1" using the technology of DNA marker-assisted selection of medicinal plants and systematic breeding. The disease-resistant cultivar of notoginseng contained 12 special SNPs based on the analysis of Restriction-site Associated DNA Sequencing (RAD-Seq). Among the SNP (record_519688) was related to the root rot-resistant characteristics, which indicated this SNP could serve as genetic markers of disease-resistant cultivars and assist the systematic breeding. Compared to the conventional cultivated cultivars, the incidence rate of root-rot and rust-rot in notoginseng seedlings decreased by 83.6% and 71.8%, respectively. The incidence rate of root-rot respectively declined by 43.6% and 62.9% in notoginseng cultivation for 2 and 3 years compared with those of the conventional cultivated cultivars. Additionally, the potential disease-resistant groups were screened based on the relative SNP, and this model enlarged the target groups and advanced the breeding efficiency. DNA marker-assisted selection of medicinal plants accelerated the breeding and promotion of new cultivars, and guaranteed the healthy development of Chinese medicinal materials industry.
