5.Biological effects of adeno-associated virus vectors of human transforming growth factor ?_1 and ?_3 for reversion of rabbit intervertebral disc degeneration

Objective To study the biological effects of pSNAV2-hTGF?1 and pSNAV2-hTGF?3 on the reversion of rabbit disc degeneration. Methods Rabbit nucleus pulpous and annulus fibrosus cells were isolated and cultured. The fluorescence labled pSNAV2 were used to detect the transfect rates of rabbit disc cells at first. Then, the pSNAV2-hTGF?1 and pSNAV2-hTGF?3 were transfected into the degenerated rabbit disc cells respectively. The biological effects of hTGF?1 and hTGF?3 on degenerated rabbit disc cells were detected with Western-bloting and 35S detection to analyze and compare the matrix synthesis of the tranfected cells. Results pSNAV2 could transfect degenerated disc cells effectively in the early stages. Both the pSNAV2-hTGF?1 and pSNAV2-hTGF?3 could stimulate the synthesis of collagen Ⅱ and proteoglycan of the rabbit disc cells. For the early stage of degenerated disc cells, the synthesis of collagen Ⅱ and proteoglycan were greater transfected with pSNAV2-hTGF?1 than transfected with pSNAV2-hTGF?3. The pSNAV2-hTGF?1 could promote the degenerated rabbit annulus fibrosus cells to synthesize collagen Ⅰ and pSNAV2-hTGF?3 could promote the degenerated nucleus pulpous cells of later stage to synthesize the collagen Ⅱ. Conclusion Both pSNAV2-hTGF?1 and pSNAV2-hTGF?3 can promote the degenerated rabbit disc cells of early stage to synthesize the matrix. pSNAV2-hTGF?3 can efficently promote the seriously degenerated nucleus pulpous cells to synthesize the collagen Ⅱ.
6.Pharmacological studies on poly-saccharide sulphate: protective effects on experimental myocardial infarction in rabbits and myocardial necrosis induced by isoprenaline in rats

This study reported the protective effects of poly-saccharide sulphate (PSS) on experimental model of myocardial infaction by high positioned double - ligation of the anterior descending left coronary artery in rabbits, and on ischemic injury of myocardium by intraperi-toneal injection of isoprenaline in rats. The results showed that PSS could decrease the infarct sizes of rabbits model expressed by ECG mappingof ∑ST segment, NST and NQ, and reduce the activities of CPK in serum. It also showed that PSS reduced the severity of myocardial necrosis and the activies of CPK in serum of the experimental rats . It suggested that PSS have the protective effects on ischemic injury of myocardium.

8.Risk factors for infants and young children with wheezing in Zhongshan city

Objectives To analyze the risk factors associated with infant wheezing in Zhongshan city. Methods A multi-center, large sample of case-control study was applied and the data related to risk factors was collected by questionnaire survey. T test and chi-square test were firstly used for univariate analysis, and then the multivariate stepwise logistic regression was used to analyze the independent risk factors associated with infant wheezing. Results A total of nine factors were found rele-vant to infant wheezing by univariate analysis including parental allergic history, way of birth, respiratory syncytial virus infec-tion, Mycoplasma pneumoniae infection, personal allergic history, like crying, parents have constant disagreements, home near the road, and factory around (P<0.05). Parental allergic history (OR=3.441, 95%CI:1.914-6.186, P<0.001), respiratory syncy-tial virus infection (OR=2.910, 95%CI:1.793-4.723, P<0.001), Mycoplasma pneumoniae infection (OR=2.277,95%CI:1.110-4.667, P=0.025), home near the road (OR=2.036, 95%CI:1.280-3.239, P=0.003) and like crying (OR=1.521, 95%CI:1.049-2.206, P=0.027) were approved to be the independent risk factors of infant wheezing in ZhongShan. Conclusions Nine factors have relationship with infant wheezing, including parental allergic history, respiratory syncytial virus infection, Mycoplasma pneumoniae infection, home near the road, like crying, personal allergic history, and that the former five factors are the indepen-dent risk factors.
9.Mild hypothermia on patients with massive hemispheric infarction

Objective To study the influence of mild hypothermia on blood glucose and blood lactate and intracranial pressure (ICP) of the patients with anterior circulation brain infarction (massive hemispheric infarction) and evaluate its clinical effects.Methods All 30 patients with massive hemispheric infarction within the first 24 hours from stroke onset were randomly divided into the control group and the hypothermia group (15 patients respectively). To the latter group, mild hypothermia (32~34℃)was initiated and ICP values were measured in the same time. Rewarming was begun after hypothermic therapy lasted for 20~49(32?9)hours. In both groups, the blood glucose and blood lactate were measured at the time of admission, the second day and the fourth day. According to Neurological Deficit Scale score, the prognosis was evaluated at the time of admission and 4 weeks later.Results In hypothermia group, the initial ICP was (14.2?4.2) mm Hg, which was significantly reduced under hypothermia to (11.3?2.8) mm Hg(P
