1.Relationship of cyclin D1 and P16 with the biological characters of lung cancer

BACKGROUND:Cyclin D1 and P16 promote cellular proliferation and inhibit cellular division by regulating the activity of CDK4. There existes to some extent amplification or overexpression of cyclin D1 gene together with P16 gene the inactivation of P16 gene in the many tumors.OBJECTIVE: To probe into the relationship of the amplification of cyclin D1 and the deletion of P16 and lung cancer.DESIGN: Controlled trial of case samples.SETTING: Department of Biochemistry, the 81 Hospital of Chinese PLA.PARTICIPANTS: Tissue samples of 40 cases of lung cancer were obtained from surgical specimens of tumor tissues and normal lung tissues at the 81 hospital of Chinese PLA between June 1998 and June 1999. Informed consent was obtained from the patients.METHODS: Method of multiplex polymerase chain reaction (PCR) was adopted. Cyclin D1 gene and P16 gene were amplified in the same tube,using extracted genome DNA as template. 10 μL PCR products were separated on a 15 g/L agarose gel.MAIN OUTCOME MEASURES: ① Amplificative result of cyclin D1and P16 in the pathological tissue of the patients with lung cancer. ② Relationship of cyclin D1 and P16 and the biological character of lung cancer.RESULTS: ① The amplified positive rates of cyclin D1 gene and P16gene in lung cancer were 55.0%(22/40) and 65%(26/40) respectively.②Relationship of cyclin D1 and P16 and biological characters of lung cancer :The amplified positive rate of both of them is closely related to the metastasis of lymph node and the staging of clinical pathology. With the incidence and development of metastasis, amplification positive rate of cyclin D1 was increased while the positive rate of P16 decreased.CONCLUSION: The high amplification of Cyclin D1 and the high deletion of P16 gene is closely related to the high activity of proliferation of lung cancer. To detect the changes of these two genes would help us to judge the metastasis, malignant extent and patients' prognosis.
2.The experimental study of poly-DL-lactic acid absorbable membrane in prevention of peripheral nerve adhesion

Objective To study the effect of poly- DL- lactic acid (PDLLA) absorbable membrane on preventing adhesion after repair of peripheral nerve injury. Methods 60 SD rats were divided into 2 groups randomly: the suturing+ PDLLA group (experimental group) and the purely suturing group (contrast group), with 30 rats in each group. Their sciatic nerves were cut off and then sutured microsurgically. In the experimental group, the anastomotic stoma was encapsulated with PDLLA membrane; in the contrast group, there was no such a treatment. We conducted gross observations and histological examinations at 2, 4, 8, 12, and 16 weeks after operation, neural electrophysiological examinations at 12 and 16 weeks after operation, and image analysis by computer at 16 weeks after operation. Results The scar proliferation and adhesion near the anastomotic stoma in the experimental group were less extensive than those in the contrast group; regenerative nerve fibers grew more straight and orderly manner. Electrophysiological examinations and image analyses by computer also showed that the experimental group achieved better results than the contrast group. Conclusion Poly- DL- lactic acid (PDLLA) absorbable membrane can prevent scar proliferation and adhesion after repair of peripheral nerve and promote nerve functional recovery effectively.
3.Treatment of nonunion following intramedullary nailing of femoral and tibial fractures

Objective To explore diagnosis of nonunion following intramedullary nailing of femoral and tibial fractures and the treatment of nonunion of femoral and tibial fractures with Less Invasive Stabilization System (LISS) or locking compression plate (LCP). Methods From February 2003 to December 2004, 7 cases of nonunion after intramedullary nailing of femoral and tibial fractures were diagnosed by X-ray or CT scanning or CT-3D. LISS or LCP fixation and bone grafting were employed to treat the 7 cases of nonunion in the femur and tibia. Their case histories ranged from 10 to 49 (mean 23.3) months. Results All the 7 patients were followed up for a mean of 9.1 (4 to 16) months. The bone union time for all the fractures averaged 4.7 months (ranging from 4 to 6 months). No loosening or breakage of the implants occurred in this series. Conclusions The operation should be done for the patients who feel pain in the lower extremities or the fracture line still exists at the fracture ends long time after intramedullary nailing. In order to improve the healing of bone fractures, micromovement and bone defects at the fracture site should be eliminated. Because of its merits in design, the LISS can be used to effectively treat nonunion at the distal femur or the proximal tibia.
4.Protective Effect of Jiedu Huayu Decoction on Rat Hepatic Immune Injury

Objective To evaluate the protective effects of Jiedu Huayu Decoction(JHD)on hepatic immune injury in mice induced by concanavalin A(Con A) and to explore its mechanism.Methods Sixty NIH mice were randomly divided into 5 groups:normal control group(group A),model group(group B),high-dose JHD group(group C),low-dose JHD group(group D) and bifendate group(group E).Except those in group A,mice in other groups were injected with Con A(at the dose of 20 mg/kg) via the tail vein at the first day of experiment.Group C and group D were orally administrated with JHD at the dose of 72.0 g/kg and 18.0 g/kg,respectively.Group E were orally administrated with bifendate at the dose of 150 mg/kg.All the drugs were given once daily for 5 consecutive days.Four hours after last medication,mice were injected with Con A once again at the same dose of 20 mg/kg.Serum aminotransferase(ALT) activity and interleukin-6(IL-6) concentration were detected at the 8th hour after Con A administration.Hepatic histopathological examination was performed,and the expression of intercellular adhesion molecule-1(ICAM-1) in hepatic tissue was detected by immunohistochemical method.Results ALT activity and IL-6 content in group C and group D were obviously lowered,hepatic histopathological changes were alleviated,and the expression of ICAM-1 in liver tissue was attenuated compared with those in group B(P
5.EXPRESSION OF THE p16 AND Rb GENES mRNA AND PROTEIN LEVELS IN LUNG CANCER

The authors used immunohistochemistry and in situ hybridization to observe the expression of the p16 and Rb mRNA and protein levels in 89 cases of lung cancer. The results showed that the loss of p16 gene expression took place mainly in non small cell lung cancer, and loss of Rb gene expression mainly in small cell lung cancer. The expression of these genes on protein level fundamentally corresponded with mRNA level. It is suggested that the expression of these two genes are related to the histological type of lung cancer, and the p16 and Rb genes can be considered as one of molecular biological targets for gene classification diagnosis. There may exist a mechanism at gene translation level which leads to the inactivation of the p16 and Rb genes.
6.Protective effects of rosiglitazone on non-alcoholic steatohepatitis in rats

ObjectiveTo investigate the preventive effects of rosiglitazone on nonalcoholic steatohepatitis (NASH) rats and to explore the potential mechanisms in modulating peroxisome proliferator-activated receptor gamma (PPARγ),nuclear factor kappa B (NF-κB),and cyclooxygenase-2 (COX-2) expression.Methods Thirty male SD rats were assigned into the normal group ( n =10),the model group ( n =10),rosiglitazone prevention group [ n =10,simultaneously 4mg/( kg · d) gavage daily at beginning].Liver appearance,liver index,and histological changes were assessed.Serum tumor necrosis factor-o (TNF-c) and prostaglandin E2 (PGE2) were determined using enzyme-linked immunosorbent assay.The expressions of PPARγ,NF-κB,and COX-2 in liver were determined using immunohistochemical methods.The mRNA and protein expressions of COX-2 were disclosed by real-time polymerase chain reaction and Western blot analysis.ResultsCompared with the normal group,the liver index significantly increased in model group (3.92 ±0.72 vs.5.71 ± 1.05,P =0.004).HE and Masson staining showed significantly increased steatosis,inflammation,and fibrosis.The serum levels of TNF-α,PGE2 in high-fat-diet-fed rats were significantly increased ( 11.72 ± 2.47 vs.29.39 ± 5.32,P =0.002 ; 236.60 ± 24.90vs.288.24 ± 17.17,P =0.004).Immunohistochemistry showed NF-κB and COX-2 in livers were significantly elevated,but PPARγ wasdecreased in nonalcoholic steatohepatitis rats.Real-time polymerase chain reaction and Western blot found mRNA and protein expressions of COX-2 were increased in the model group (0.57 ± 0.08 vs.2.83 ± 0.24,P =0.0007 ; 0.38 ± 0.03 vs.1.00 ± 0.03,P =0.004).Compared with the model group,the expressions of PPARγsignificantly increased and the expressions of NF-κB and COX-2 significantly decreased ( mRNA:2.83 ± 0.24 vs.0.46 ± 0.11,P =0.002 ; protein: 1.00 ± 0.03 vs.0.62 ± 0.02,P =0.006 ) in the rosiglitazone prevention group.ConclusionBy inhibiting NF-κB and COX-2 expressions,rosiglitazone can reduce insulin resistance and then prevent the occurrence and deve lopment of nonalcoholic steatohepatitis.
7.Role of endoplasmic reticulum stress in omega-3 polyunsaturated fatty acids supplementation for attenu-ating inflammatory response in experimental rat models of colitis

Objective To investigate the effects and mechanisms of omega-3 polyunsaturated fatty acids (ω-3 PUFA) supplementation on colonic macroscopic and histological score , inflammatory response , and endo-plasmic reticulum stress ( ERS ) response in experimental rat models of colitis .Methods Experimental rat models of colitis were induced by trinitro-benzene-sulfonic acid (TNBS).Totally 100 male SD rats were ran-domly divided into 5 groups according to the random data tables:sham operation group ( Sham group ) , inflam-matory bowel disease group (IBD group),ω-3 PUFA supplementation group (IBD+ω-3 group), 5-aminosali-cylic acid group ( IBD +5-ASA group ) , and ERS activation 2-deoxy-D-glucose group ( IBD +ω-3 +2-DG group).Colonic macroscopic and histological scores were evaluated on days 1, 3, 7 and 14 after modeling.The serum levels of tumor necrosis factor-α(TNF-α), interleukin (IL) -1, and IL-6 were measured using en-zyme-linked immunosorbent assay , whereas ERS cytokines including glucose-regulated protein 78 ( GRP78 ) , inositol-requiring enzyme 1 (IRE-1), and C/EBP homologous protein (CHOP) were tested by Western blot. Results Compared with the Sham group , colonic macroscopic and histological score , the serum levels of in-flammation relatived factors (TNF-α, IL-1, IL-6) and ERS relatived factors (GRP78、 IRE-1, CHOP) were significantly increased on the rest of the four groups ( all P<0.001 ) .Compared with the IBD group , ω-3 PUFA supplementation reduced colonic macroscopic [7 d: 3.55 ±0.29 vs.4.37 ±0.39, P=0.03, 14 d:2.46 ±0.17 vs.3.86 ±0.21, P=0.04] and histological score [7 d: (2.56 ±0.27) scores vs.(3.45 ± 0.40) scores, P=0.02, 14 d: (2.23 ±0.20) scores vs.(3.06 ±0.26) scores, P=0.04].Meanwhile,ω-3 PUFA supplementation suppressed the expressions of inflammation [TNF-α:(43.71 ±11.39) pg/ml vs. (84.97 ±13.81) pg/ml, P=0.02, IL-1:(38.51 ±10.60) pg/ml vs.(73.04 ±12.48) pg/ml, P=0.01, IL-6:(28.91 ±7.27) pg/ml vs.(53.45 ±9.40) pg/ml, P=0.02] and ERS relatived factors (GRP78:2.41 ±0.29 vs.1.47 ±0.21, P=0.01, IRE-1:2.83 ±0.31 vs.1.23 ±0.20, P<0.001, CHOP:1.89 ± 0.17 vs.1.32 ±0.11 , P=0.04 ) .However , the salutary effects of ω-3 PUFA would been reversed by ERS activation 2-deoxy-D-glucose [ TNF-α: (72.67 ±10.37 ) pg/ml vs.(43.71 ±11.39 ) pg/ml, P =0.02, IL-1:(57.66 ±13.88) pg/ml vs.(38.51 ±10.60) pg/ml, P=0.02, IL-6: (46.10 ±9.67) pg/ml vs. (28.91 ±7.27) pg/ml, P=0.01, GRP78:1.47 ±0.21 vs.1.82 ±0.24, P=0.03, IRE-1:1.23 ±0.20 vs.2.21 ±0.23, P=0.02, CHOP:1.32 ±0.11 vs.1.61 ±0.16, P=0.04].Conclusion The salutary effects of ω-3 PUFA supplementation on the colitis induced by TNBS appear to be mediated by inhibited inflam -matory responses , which may suppress the activation of ERS response .

9.Protective Effect of Phyllanthus Compound Ⅱon Experimental Hepatic Damage in Mice

【Objective】To observe the protective effect of Phyllanthus Compound Ⅱ(PCⅡ) on experimental hepatic damage in mice.【Methods】Sixty NIH mice were randomized into 5 groups: normal control group,model group,high-,moderate-and low-dose PCⅡ(110,55,and 27.5 g/kg respectively) groups,bifendate(150 mg/kg) group.Mice models of acute hepatic damage were induced with concanavalin A(Con A) and D-galactosamine(D-GalN)respectively.Serum levels of alanine transferase(ALT) and aspartate transferase(AST),and superoxide dismustase(SOD) activity were examined.The pathological changes of hepatic tissue were also observed.【Results】 PCⅡmarkedly decreased serum ALT and AST levels,and decreased hepatic SOD activity(P0.05 compared with bifendate group).The hepatic pathological changes were improved in PCⅡ groups.【Conclusion】PCⅡ exerts a better protective effect on mice hepatic damage induced by Con A and D-GalN,and its protective mechanism may be related with the inhibition of lipoperoxidation damage.
10.Clinical features, levels of protein carbonyl in serum of obstrucitive sleep apnea syndrome with type 2 diabetes mellitus.

OBJECTIVE:
To explore the prevalence, clinical feature and levels of protein carbonyl (PCO) in serum of type 2 diabetes mellitus combining obstrucitive sleep apnea syndrome (OSAHS).
METHOD:
Two hundred and three patients with type 2 diabetes were taken multi lead sleep detection and their AHI, age, height, body mass index (BMI),waistline, duration of diabetes, fast blood glucose, HbA1c level and level of PCO in serum were recorded.
RESULT:
The prevalence of OSAHS was 79.2% in 203 patients, serious apnea 30.4%, moderate apnea 45.4%, mild apnea 24.2%. BMI, waistline, fast blood glucose, HbA1c level and level of PCO in serum were higher in patients complicated with OSAHS (P < 0.01). HbA1c was independently and positively correlated with patients with OSAHS risk (P < 0.05, OR 6.11). The independent correlation factors of AHI included HbA1c level,BMI, waistline, duration of diabetes and level of PCO in serum,with HbA1c as the predominant factor (P < 0.05).
CONCLUSION
The prevalence of OSAHS was higher in patients of type 2 diabetes mellitus combining (OSAHS). Moreover, in these patients poor glucose control and aggravated protein oxidative injury were observed.
