Objective To study the proliferation promoting effect on osteoblast cells induced by sodium orthovanadate and whether nitric oxide (NO) was involved in this process. Objective To study the therapeutic effect on murine allergic asthma with recombinant Bla g 2(rBla g 2) allergen and its possible mechanism.Methods Eighteen BALB/C mice were randomly divided into three groups:normal control group(group A) , asthma model group(group B) , and recombinant protein rBlag2 treatment group(group C).Mice in groups B and C were subcutaneously immunized weekly with rBla g 2(50 mg) formulated in Al(OH)3 adjuvant for three weeks.Group A received only adjuvant emulsified with normal saline.Two weeks after the last inoculation, mice in group C were administered each with rBla g 2(100 mg) /dose, and groups A and B were given PBS.All the mice received eight doses at 2-day intervals.One week after the last immunotherapy, mice in groups B and C were intranasally challenged with 50 mg rBla g 2 daily for seven days, while mice in group A received PBS.Twenty-four hours after the challenge, the following items were examined:airway hyperresponsiveness of mice, total cellular score and cell classification in bronchoalveolar lavage fluid(BALF) , level of rBla g 2-specific IgE and IgG2a in serum, lung inflammation by HE stain, and Bcl-2 expression of eosinophils of lung by immunohistochemistry.Results Compared with group B, group C showed a decreased Penh value of airway hyperresponsiveness(P

AIM: To observe the growthinhibiting cell cycle-modifying and apoptosis-inducing effects of satraplatin on human ovarian carcinoma cell line A2780, and to explore its possible mechanism. METHODS: The effect of satraplatin on A2780 cells proliferation was determined using MTT, and the change in cell cycle was analyzed using PI staining. Morphologic change was visualized by fluorescence and electron microscopy. AnnexinV-FITC/PI staining multiparameter flow cytometry and immuno- histochemical TUNEL assay were used to detect apoptotic cells. The activity of caspase-3 and the effect of pan-caspase inhibitor on cell viability were measured as well. RESULTS: The growthinhibiting and apoptosis-inducing effects of satraplatin were dose-dependent and similar to those of cisplatin. Satraplatin mainly caused A2780 cell accumulation in S phase accompanied by minor accumulation in G2/M phase. Cells treated with satraplatin exhibited typical morphology of apoptosis. Satraplatin-induced increase in caspase-3 activity of A2780 cells was concentration-dependent. The viability of A2780 cells was affected by pan-caspase inhibitor z-VAD-fmk in a dose-dependent manner under certain concentration of z-VAD-fmk. CONCLUSION: Satraplatin-induced apoptosis in A2780 in vitro was observed. Caspase-dependent and independent pathways were involved in apoptosis induced by satraplatin, and the latter included caspase-3 dependent and non-caspase-3 dependent pathways.

ObjectiveTo investigate the effects of low dose testosterone undecanonate (TU) and dehydroepiandrosterone (DHEA) on serum androgen levels in male adult rabbits postinfarction congestive heart failure (CHF) model.Methods48 male 4～5 months old New Zealand rabbits were randomly assigned to ligation of first branch of left coronary artery (n=40) or sham operation (n=8). 3 weeks later, 30 survived model rabbits were randomly treated with TU 3 mg/kg per 2 weeks intramuscular injection (TU group, n=10), DHEA 3 mg/kg/d oral (DHEA group, n=10) or placebo (placebo group, n=10) for 3 months. Serum androgen changes were recorded at 0th, 3rd and 15th week.ResultsAt 3rd week after peration, left ventricular ejection fraction of model animals was 47.6±4.5% lower than that of the sham group (66.7±5.8%) (P<0.05). Serum free testosterone level of model animals returned to normal ranges after low dose TU or DHEA treatment, otherwise DHEAS level was higher than the sham group (P<0.05).ConclusionThe doses used in this research reached physiological dosage to male adult rabbits in postinfarction CHF model.

Objective To explore the nursing after free great toe fibular flap transplant to repair finger pulp defects. Methods 12 cases with finger pulp defect accepted the free great toe fibular flap were reviewed. Results All flaps survived and no vascular crisis occurred. The flap shaped well, and the skin sweated and the two point discrimination was 4-6 mm. Conclusion Close monitoring and appropriate is important after free great toe fibular flap transplant to repair finger pulp defects.

Objective To investigate the whole genomic expression profiles of chronic atrophic gastritis with interleukin(IL)-1β-31CC/-511TT genotype as measured by oligonucleotide microarray technique.Methods Genomic RNA was extracted from gastric biopsies of 12 patients with chronic atrophic gastritis(6 with IL-1β-31CC/-511TT and 6 with IL-1β-31TT/-511CC).The genomic profiles of IL-1β gene polymorphisms 31CC/-511TT and 31TT/-511CC were compared and tested for differential expressed genes associated with 31CC/-511TT using Agilent human whole genomic oligonucleotide microarrays.The results were further analyzed in terms of gene ontology(GO).Results There were 200 differentially expressed genes associated with IL-1β-31CC/-511TT,159 of which were up-regulated and 41 were down-regulated.These genes mainly involved in macromolecule metabolic process,post-translational protein modification,ubiquitin cycle,and protein kinase cascade.Five genes had biological activities,one of which was down-regulated gene(PCSK5)and 4 were upregulated genes(PRKCA,NPLOC4,TRIB3 and MAPKAPK3).Conclusions The chronic atrophic gastritis with IL-1β-31CC/-511TT genotype has molecular phenotypes which is associated with malignance and inflammation.These individuals are needed more intensive preemptive treatment and dynamic surveillance.

OBJECTIVETo discuss the treatment and diagnosis of congenital median perineal cleft in children.

METHODSBetween January 2009 to February 2013, 7 cases were diagnosed as congenital median perineal cleft according to the symptoms. Among them, 4 cases underwent surgery to correct cleft with double triangular perineal flaps. The other 3 cases with minor cleft did not receive surgery management.

RESULTSThere is an median cleft from the perineum to the anus with mucosa on the cleft surface. Primary healing was achieved in all the four patients with satisfactory appearance. The patients were followed up for 1-4 years with almost normal perineal appearance.

CONCLUSIONSCongenital perineal median cleft can be diagnosed according to the symptoms. Double triangular perineal flaps can be effectively correct the cleft to attain normal perineal appearance.

Objective To explore the value of hydrogen sulfide(H2S) in the evaluation of severity of acute pancreatitis(AP).Methods Fifty-one patients with AP from February 2013 to December 2015 in this hospital were divided into severe acute pancreatitis (SAP,n=21) group and mild acute pancreatitis (MAP,n 30) group.Thirty six non-pancreatitis abdominal patients and 9 healthy persons(NC) were chosen as compare groups.The peripheral venous blood samples from the four groups and the Ranson Score of AP group were obtained after admission,the venous blood samples of 12 h,24 h,48 h of AP group were also obtained.Blood samples were used to detect the concentrations of hydrogen sulfide with ELISA method.We compared the concentrations of H2S between the four groups with one way ANOVA,and post hoc;compared the differences of concentrations of H2S in AP group between after admission,12 h,24 h and 48 h with repeated measures data ANOVA;and explored the relationship between the concentrations of H2S after admission and the Ranson scores in AP group with Spearman correlation analysis.Results The concentra tions of H2S was significant higher in SAP group than MAP,abdominal,and NC group(P=0.000);the concentrations of H2S was also significant higher in MAP group than abdominal,and NC group(P=0.000);there is no significant difference between the abdominal and NC group(P =0.131).There is significant difference of H2S concentrations between the four times in AP group(P =0.000),decreasing gradually over time.The H2S concentrations in AP group after admission was significantly related with their Ranson scores(r=0.578,P=0.000).Conclusion There is certain value of the concentrations of H2S in the diagnosis of AP,the higher H2S level suggests the higher severity of the patients with acute pancreatitis.

Objects To investigate the correlation of the single nucleotide polymorphism ( SNP) of clopidogrel related gene CYP2C19, ABCB1, PON1 to the occurrence of clopidogrel resistance ( CR) and TEG among patients after percutaneous coronary intervention.Methods A total of 299 patients after PCI were enrolled from April 2015 to December 2015.It genotyped the CYP2C19(rs4244285,rs4986893)ABCB1 ( rs1045642 ) and PON1 ( rs662 ) gene, measured clopidogrel response by TEG.Accordingly, all the enrolled 299 patients were then divided into CR group (n=17) +non-CR (NCR) group (n=282) or CLR group (n=54) +non-CR (NCR) group (n=245) by TEG(%).All the patients were divided into EM、IM and PM group by CYP2C19 genotype.The age of patient in CR (71.1 ±11.1) years old is higher than NCR (65.02 ±10.51) years old (t=2.559, P<0.05).Results CYP2C19 PM was associated with decreased of TEG(ADP) (Z=-2.065, P=0.039), while it was not related to the age of patient(Z=0.405,P>0.05).There was no significant difference between CR(χ2 =0.175,P=0.916) CLR(χ2 =1.589,P=0.452)and the level of TEG(ADP) (Z=-0.030,P=0.976) in PON1(rs662) polymorphism.There was no significant difference between CR(χ2 =1.722,P=0.423) CLR(χ2 =0.176,P=0.916) and the level of TEG(ADP) (Z=-0.331,P=0.741) in ABCB1(rs1045642) polymorphism.Conclusions CYP2C19 PM is associated with decreased of TEG(ADP).It is considered that no correlation exists between ABCB1(rs1045642) and PON1(rs662) polymorphism and clopidogrel resistance in patients with coronary heart diseases.The loss of function of ABCB1 ( rs1045642 ) and PON1 ( rs662 ) is not associated with decreased of TEG(ADP) in CYP2C19 PM patients.

The control handles( including financing, provider payment, organization, regulation and behavior) proposed by World Bank in health policy research as the basic framework,and the nature and positioning of family doctors′ contract-based service as the precondition, we recommended the system framework of such a mechanism, comprising one contract, three regulations, five control handles and seven supporting policies. Such a framework can serve as the cornerstone for family doctors′ contract-based service experiment in Zhejiang province for the sake of hierarchical medical system.
