1.Effects of ulinastatin on cytoldnes and respiratory index during open heart surgery with CPB

Objective To investigate the effects of ulinastatin on pro- and anti-inflammatory cytokines and respiratory index during open heart surgery under CPB. Methods Twenty ASA Ⅱ- Ⅲ patients of either sex (9 males, 11 females) scheduled for elective valve replacement under CPB were randomly divided into two groups of 10 each : control group (C) and ulinastatin group (W) . Patients with hepato-renal dysfunction or taking glucocorticoid were excluded. The patients were premedicated with intramuscular phenobarbital 0.1 g and scopolamine 0.3 mg. Anesthesia was induced with midazolam 0.1 mg?kg-1 , fentanyl 10?g?kg-1 and vecuronium 0.1mg?kg-1 . After tracheal intubation anesthesia was maintained with midazolam and fentanyl. The patients were mechanically ventilated. VT was set at 8-12 ml?kg-1 , RR 10-12 bpm and I: E 1:2. PETCO2 was maintained at 35-45 mg. In ulinastatin group, the patients received ulinastatin 12 000 U?kg-1 of which half was given i.v. before CPB and half was added to the priming fluid, while group C received normal saline instead of ulinastatin. Blood samples were taken from radial artery for determination of plasma TNF-?, IL-6, and IL-10 concentrations and blood gas analysis before operation (T1 ) , 30 min after initiation of CPB (T2), 1 h (T3 ) , 4 h (T4 ) and 24 h (T5 ) after CPB. Respiratouy index (PA-aDO2/ PaO2) was calculated at T1-5 .Results There were no significant differences in sex, age, weight, height, duration of CPB, and aortic cross-clamping time between the two groups. Objective To investigate the localization of human herpesvirus (HHV-8) in lesions of Kaposi′s Sarcoma (KS) and explore the role of HHV-8 in the pathogenesis of KS in Xinjiang. Methods HHV-8 DNA was detected by fluorescence in situ polymerase chain reaction. 3.Epidemiological analysis of imported malaria prevalence in Luoyang City from 2010 to 2013

Objective To understand the epidemiological characteristics of imported malaria in Luoyang City,so as to pro-vide the evidence for malaria prevention and control in this city. Methods The Epidemic situation data from network reports, as well as the case survey and the epidemiological investigation data of imported malaria were collected and analyzed in Luoyang City from 2010 to 2013. Results A total of 98 imported malaria cases were reported in Luoyang City from 2010 to 2013,includ-ing 35 cases of vivax malaria,57 cases of falciparum malaria,4 cases of ovale malaria and 2 cases of quartan malaria. All the cases were confirmed by laboratory detection. Seventy-one cases(72.44%)returned from African countries,and 27(27.55%) cases returned from Southeast Asian countries. The majority cases were young man,and 78.57%of the cases were diagnosed by different levels of centers for disease control and prevention. There was no significant seasonal variation in onset time. The medi-an time from onset to seeing doctor was 6 days. Conclusions The epidemic situation of imported malaria is quite serious in Luoyang City. It is necessary to further strengthen the professional training and multi-sectoral cooperation ,and take effective pre-vention and control measures to reduce the hazard of imported malaria.

5.Effects of tacrolimus on the proliferation of HepG2.2.15 cells and the replication of hepatitis B virus in vitro

BACKGROUND: The relationship between liver cancer recurrence and hepatitis B virus recurrence remains poorly understood and it is considered to be related to application of immunosuppressive agent after liver transplantation. OBJECTIVE: To investigate the effects of tacrolimus (FK506) on the proliferation of HepG2.2.15 cells and the replication of hepatitis B virus in vitro. METHODS: HepG2.2.15 cells were in vitro cultured. After passage 3 HepG2.2.15 cells were cultured for 24 hours, they were interfered with different concentrations of FK506. 0 g/L FK506-interfered group served as control group, 50 g/L FK506-interfered group as low-concentration FK506 group, 100 g/L or 500 g/L FK506-interfered group as medium-concentration FK506 group, and 1 000 g/L or 3 000 g/L FK506-interfered group as high-concentration FK506 group. RESULTS AND CONCLUSION: Moderate- and high-concentration FK506 exhibited inhibitory effects on the proliferation of HepG2.2.15 cells, while low-concentration FK506 exhibited no inhibitory effects with correlation. High-concentration FK506 made HepG2.2.15 cells arresting at G0/G1 stage. FK506 decreased CyclinA expression in HepG2.2.15 cells in a dose-dependent manner. Higher concentration of FK506 leaded to lower expression of CyclinA. FK506 did not produce effects on the replication of hepatitis B virus in HepG2.2.15 cells. These results indicate that FK506 inhibits the proliferation of HepG2.2.15 cells in vitro, which occurs possibly due to Cyclin A, but it would not affect the replication of hepatitis B virus in vitro.
6.Quick Hand Disinfection versus Six Steps Hand Washing in ICU

OBJECTIVE To explore an effective method of hand hygiene in ICU.METHODS Doctors and nurses in ICU washed hand with quick hand disinfectant or six steps hand washing at random,the time and compliance of two methods were recorded and their effect was checked.RESULTS The compliance and effect of quick hand disinfection were better than six steps hand washing(P=0.000),the time of quick hand disinfection was shorter than six steps hand washing(P=0.000).CONCLUSIONS The quick hand disinfection has good effect and high compliance.It is a good way for hand hygiene in ICU to improve clinical effects and values.

Purpose The aim is to breed a highly productive γ-linolenic acid mutant strain.Methods The original strain Mortierella ramanniance AS 3.3413 was treated by 5-FU,UV,LiCl as mutanting agent.Results A highly productive γ-linolenic acid mutant strain F5 was successfully obtained.The γ-linolenic acid yield of mutant strain F5 was increased by 300% as compared with that of the original strain.Pass-generation test showed that the heredity character of mutant strain F5 was stable.After the optimal medium and culture condition was established, the yield of γ-linolenic acid was up to 1 153.63 mg/L,and was increased by 335.87% as compared with that of the original strain.Conclusion The mutant strain can be used for the industrial production of GLA.

Objective To investigate the infectious status,etiological spectrum and epidemiological characteristics of rotavirus (group A/B/C),calicivirus (novovirus Ⅰ/Ⅱ,sapovirus),astrovirus and enteric adenovirus in diarrhea cases below 5 years old from 2008 to 2015 in Henan provinces.Methods Totally 2541 stool samples were collected from cases below 5 years old in four sentinel hospitals.All stool specimens were tested for group A rotavirus by double antibody sandwich ELISA method.G/P genotyping of group A rotavirus was determined by nested multiplex PCR.Viral RNA was extracted from all samples and rotavirus (group B/C),calicivirus,astrovirus and enteric adenovirus were detected by two-step reverse transcription-polymerase chain reation (RT-PCR)/PCR.Results One thousand four hundred and twenty-one out of 2 541 samples were positive with a total positive rate of 55 .9%,among which,102 were mixed infection.The isolation rate of rotavirus was 36.0% (914 samples)(group A:785 cases,group B:36 cases,group C:93 cases),calicivirus was 12.1 % (308 samples)(novovirus Ⅰ:64 cases,novovirusⅡ:193 cases,sapovirus:51 cases),astrovirus was 5 .9% (151 samples),enteric adenovirus was 1 .9%(48 samples).The group A rotavirus gene type combinations were composed mainly of G9P[8],G2P[4], G3P[8 ],G1P [8 ]and most cases were identified from September to November and March to May. Novovirus Ⅱ was predominant in calicivirus and most cases were identifed between March and May. Rotavirus or calicivirus infection was mainly among children aged 4—12 months or 3—5 years, respectively.Clinical manifestations included fever,diarrhea,vomiting,dehydration.Gender and region distributions differed according to the types of pathogen.Conclusions Group A rotavirus and novovirus Ⅱare the major viral pathogen in diarrhea cases younger than 5 years old in Henan province.Different viral infections exhibit extinct epidemiologic and clinical characteristics.
10.Effect of angiotension-Ⅱ on rat cardia fibroblasts

AIM: To investigate the effects of angiotensin-Ⅱ (Ang-Ⅱ) on the proliferation and collagen synthesis in rat cardiac fibroblasts and the expression of Ang-Ⅱ receptor on fibroblasts. METHODS: [3H]-TdR and [3H]-prolin at different concentrations were incubated with the confluent fibroblasts of newborn rats stimulated by Ang-Ⅱ. Receptor of Ang-Ⅱ on fibroblasts and intracellular free calcium ions were examined by autoradiograms and fluorescence technique. RESULTS: In the presence of Ang-Ⅱstimulation, the increase in incorporation of [3H]-TdR and [3H]-prolin was much higher than that of control in a dose-dependent manner. Autoradiograms showed that Ang-Ⅱ was uniformly distributed over the membrane of the fibroblasts. The silver grains on fibroblasts were obviously decreased after adding unlabeled Ang-Ⅱ by the autoradiograms. The concentration of free calcium ions was increased in fibroblast. CONCLUSION: These findings suggest that Ang-Ⅱ promotes fibroblast proliferation and the syntheses of collagen protein, in which Ang-Ⅱ binds to the membrane receptor of fibroblasts to activate the signal system in cells, such as intracellular free calcium ions.
