1.Effect of laparoscopic cholecystectomy on quality of life index and liver function in patients with cholecystolithiasis

Objective:To analyze the effect of laparoscopic cholecystectomy on quality of life index and liver function in patients with cholecystolithiasis.Methods:The clinical data of 160 patients with cholecystolithiasis in Zhejiang Wenzhou Tianning Hospital from January 2015 to January 2018 were retrospectively analyzed. Among them, 80 patients were treated with small incision cholecystectomy (control group), and 80 patients were treated with laparoscopic cholecystectomy (observation group). The operation time, bowel movement recovery time, visual analogue score (VAS) 24 h after operation, quality of life index, total bilirubin (TBIL), direct bilirubin (DBIL), alanine aminotransferase (ALT), aspartate aminotransferase (AST), C reactive protein (CRP) and tumor necrosis factor α (TNF-α) were compared between 2 groups.Results:The operation time, bowel movement recovery time and VAS 24 h after operation in observation group were significantly lower than those in control group: (57.8 ± 8.4) min vs. (89.8 ± 10.5) min, (25.3 ± 4.8) h vs. (38.8 ± 6.2) h and (7.33 ± 1.05) scores vs. (9.18 ± 1.85) scores, and there were statistical differences ( P<0.05). The quality of life index 1, 6 and 12 months after operation in observation group was significantly higher than that in control group: (113.2 ± 8.9) scores vs. (108.6 ± 7.4) scores, (126.9 ± 10.2) scores vs. (115.9 ± 9.3) scores and (134.3 ± 10.1) scores vs. (123.0 ± 8.1) scores, and there was statistical difference ( P<0.05). The CRP, TNF-α, DBIL, TBIL, AST and ALT 3 and 7 d after operation in observation group were significantly lower than those in control group, and there were statistical differences ( P<0.05). Conclusions:Laparoscopic cholecystectomy is effective in treating cholecystolithiasis and can effectively reduce the quality of life index and liver function.
2.Study on the relationship between Gln223Arg variation in leptin receptor gene and hypertension complicated with obesity in Wenzhou population

Objective To study the genotype distribution of Gln223Arg in Han population in Wenzhou area,and discuss its association with hypertension patients with obesity.Methods The genotypes of Gln223Arg variant in leptin receptor gene were determined by polymerase chain reaction-restriction fragment length polymorphisms(PCR-RFLP) assay in 321 unrelated subjects of Wenzhou Han population,including 111 normal subjects(the control group),106 cases of hypertension patients with obesity and 104 cases of hypertension patients without obesity(the case group).Results The frequencies of GG,GA and AA genotypes in patients of hypertension with obesity were 0.604,0.377 and 0.019,respectively,0.712,0.279 and 0.010 in patients without obesity;0.802,0.189 and 0.009 in control group.The frequencies of G and A allele were 0.769,0.231 respectively in patients of hypertension with obesity,0.865,0.135 in patients of hypertension without obesity,0.869,0.104 in control group.The genotype frequency of Gln223Arg variant in leptin receptor gene in the control group and the case group had a significant difference(?2=6.310,P
3.Effects of phycocyanin on apoptosis of human laryngeal cancer HEP-2 cells

[ ABSTRACT] AIM:To investigate the effect of phycocyanin on the apoptosis of human laryngeal cancer HEP-2 cells and to explore the inhibitory mechanism of phycocyanin to tumor.METHODS:Highly purified phycocyanin was ex-tracted from spirulina.The effects of phycocyanin at different concentrations on the growth of human laryngeal cancer HEP-2 cells were detected by MTT assay.In addition, the cell structures were observed under electron microscope.The cell ap-optosis was analyzed by flow cytometry.The production of reactive oxygen species ( ROS) was measured by flow cytometry. Enzymatic activities of caspase-3,-8 and-9 were measured by chemical colorimatry.The expression of Bax, Bcl-2, Fas, P53, caspase-3 and caspase-9 at mRNA and protein levels was determined by RT-PCR and Western blot.RESULTS:MTT test confirmed that phycocyanin inhibited the cell activity of HEP-2 cells with time and dose dependent manners.The result of electron microscope observation and flow cytometry indicated that phycocyanin induced the apoptosis of HEP-2 cells.The intracellular content of ROS was increased.The activities of caspase-3, -8 and -9 were increased.RT-PCR showed that the mRNA expression of Bax, Fas, P53, caspase-3, caspase-9 was increased and Bcl-2 was decreased.The results of Western blot were consistent with the results of RT-PCR.CONCLUSION:Phycocyanin might induce apoptosis of HEP-2 cells by down-regulating Bcl-2, up-regulating Bax, Fas and P53, and the transduction of apoptotic signals in the human laryngeal cancer cells.
