1.Protective effects of piperine on alveolar bone and collagen in a periodontitis model

BACKGROUND:Piperine in models of pancreatitis, gout, middle cerebral artery infarction has anti-inflammatory, antioxidant and immune regulatory effects, but its effects on periodontitis model are not clear.
OBJECTIVE:To observe the protective effect of piperine on bone absorption and degradation of col agen in experimental rat models of periodontitis.
METHODS:Rat models of periodontitis were established by ligaturing the dental cervix of rat mandibular first molar with 3-0 silk. On day 1 before model establishment, rats were intragastrical y administered piperine 50 and 100 mg/kg. There were healthy control group and model group. Related detection was performed 8 weeks after model establishment.
RESULTS AND CONCLUSION:(1) Results of quantitative CT analysis:compared with the healthy control group, the distance from the first molar enamelo-cemental junction to the alveolar ridge crest was significantly lower in the model group (P<0.05). The degree of alveolar damage was significantly improved in the 50 and 100 mg/kg piperine groups compared with the model group (P<0.05). (2) Related factor protein expression:compared with the model group, matrix metal oproteinase-8,-13 and interleukin-1βprotein expression was significantly decreased in the 100 mg/kg piperine group (P<0.05);matrix metal oproteinase-8 protein expression was significantly decreased in the 50 mg/kg piperine group (P<0.05). (3) Col agen fiber morphology:compared with the model group, col agen fibers arranged orderly and col agen fiber area significantly increased in the 50 and 100 mg/kg piperine groups (P<0.05). (4) Results confirmed that piperine could reduce the alveolar bone resorption, reduce the degradation of col agen fibers and protect the periodontal tissues in models of periodontitis. Its mechanism is associated with the inhibition of matrix metal oproteinase-8,-13 and interleukin-1βprotein expression.
2.Clinical Attack Type and Electroencephalogram Features of 36 Cerebral Palsy Children with Epilepsy

ObjectiveTo investigate the relationship between the clinical type of attack and electroencephalogram (EEG) features of cerebral palsy (CP) children with epilepsy (EP).MethodsThe EEGs of 36 CP cases with EP were recorded under natural circumstances.Results20 cases (55.6%) had focal high amplitude spikes and wave complexes; 4 cases (11.1%) had typical hypsarrhythmia; 3 cases (8.3%) had varied hypsarrhythmia. Among them, the spikes and wave complexes of 9 cases were bilateral.ConclusionThe most of the clinical attack and EEG characters of CP children with EP are partial type.

4.Determination of Seven Antibiotics in Cosmetics by Ultra Performance Liquid Chromatography

Objective To establish an ultra performance liquid chromatography method for the determination of 7 kinds of antibiotics(minocycline hydrochloride,ox tetracycline,tetracycline hydrochloride,chlortetracycline hydrochloride,doxycycline hydrochloride,chloramphenicol,metronidazole) in anti-acne cosmetics.Methods Seven kinds of antibiotics were separated on a Waters UPLC BEH C18 column by gradient elution using A(methanol/acetonitrile = 1/2) and B(1% formic acid aqueous solution) as the mobile phase at a flow rate of 0.2 ml/min,with the detection wavelength of 268 nm.Results The detection limits were 0.3-0.5 ng,the calibration curves were linear in the range of 2-100 ng(1-50 mg/L),the precision was not more than 4.8% and the rate of recovery were 82.5%-105.6%.Conclusion The method established in the present paper is rapid,sensitive and suitable for the determination of 7 kinds of antibiotics in anti-acne cosmetics.

6. Carfilzomib inhibits the growth of lung adenocarcinoma via upregulation of Gadd45a expression

Proteasome inhibitors have shown remarkable success in the treatment of hematologic neoplasm. There has been a lot of attention to applying these drugs for solid tumor treatment. Recent preclinical study has signified the effectiveness on cell proliferation inhibition in lung adenocarcinoma treated by carfilzomib (CFZ), a second generation proteasome inhibitor. However, no insight has been gained regarding the mechanism. In this study, we have systematically investigated the CFZ functions in cell proliferation and growth, cell cycle arrest, and apoptosis in lung adenocarcinoma cells. Flow cytometry experiments showed that CFZ significantly induced G2/M cell cycle arrest and apoptosis in lung adenocarcinoma. MTS and colony formation assays revealed that CFZ substantially inhibited survival of lung adenocarcinoma cells. All results were consistently correlated to the upregulation expression of Gadd45a, which is an important gene in modulating cell cycle arrest and apoptosis in response to physiologic and environmental stresses. Here, upregulation of Gadd45a expression was observed after CFZ treatment. Knocking down Gadd45a expression suppressed G2/M arrest and apoptosis in CFZ-treated cells, and reduced cytotoxicity of this drug. The protein expression analysis has further identified that the AKT/FOXO3a pathway is involved in Gadd45a upregulation after CFZ treatment. These findings unveil a novel mechanism of proteasome inhibitor in anti-solid tumor activity, and shed light on novel preferable therapeutic strategy for lung adenocarcinoma. We believe that Gadd45a expression can be a highly promising candidate predictor in evaluating the efficacy of proteasome inhibitors in solid tumor therapy.
7. Prediction of occult carcinoma in contralateral nodules based on the ultrasonic features of unilateral papillary thyroid carcinoma

Objective:
To investigate the occurrence of occult carcinoma in contralateral lobes based on the ultrasonic features of unilateral papillary thyroid carcinoma.
Methods:
The study included 202 consecutives cases of unilateral papillary thyroid carcinoma with benign nodules in the contralateral lobe identified by preoperative ultrasound or fine-needle aspiration from June 2014 to December 2015. All patients received total thyroidectomies, and with postoperative pathological examination they were divided into two groups, one including 60 cases with positive occult cancer and another one consisting of 142 cases with negative occult cancer. Univariate and multivariate analyses were performed to analyze the sonographic features of unilateral papillary thyroid carcinoma relevant to the occurrence of occult carcinoma in the contralateral nodules.

9.Characteristics and risk factors of abdominal lymph node metastasis in esophageal cancer

Objective To investigate the characteristics and risk factors of abdominal lymph node metastasis in thoracic esophageal squamous cell cancer.Methods The clinical data of 586 patients with thoracic esophageal cancer who underwent surgery via transabdominal and transthoracic approaches between June 2009 and June 2014 at the Sichuan Cancer Hospital were retrospectively analyzed.All the patients received resection of esophageal cancer and lymph node dissection, and the transabdominal right thoracic approach or cervico-thoracicabdominal triple incision was selected according to the condition of patients.No.18, 19, 20 lymph nodes were dissected seperately and No.16, 17 and lesser curvature lymph nodes were separated.All the specimens of lymph nodes were detected by regular pathological examination.Measurement data with normal distribution were presented as x ± s and count data were described as rate.Comparisons of rate between 2 specimens and among the multiple specimens were respectively analyzed using the chi-square test and partition of chi-squared.The multivariate analysis was done using the logistic regression.Results The number of lymph node dissected in 586 patients was 12 524 with an average number of 20 ± 11 per case, and the rate of lymph node metastasis was 55.63% (326/586).The number of mediastinal lymph node dissected was 7 012 with an average number of 12 ± 5 per case, and a rate of mediastinal lymph node metastasis was 40.96% (240/586).The number of abdominal lymph node dissected was 5 512 with an average number of 9 ± 8 per case, and a metastasis rate was 31.74% (186/586).The abdominal lymph node metastasis rate of the upper, middle and lower thoracic esophageal cancer were 13.73% (14/102), 31.51% (92/292) and 41.67% (80/192), respectively, showing a significant difference among the above 3 indexes (x2 =25.91, P < 0.05).The lymph node metastasis rate in No.16, 17, 18,19, 20 and lesser curvature lymph nodes were 12.80% (75/586), 16.89% (99/586), 1.71% (10/586),0.68% (4/586), 1.71% (10/586) and 2.05% (12/586), respectively, with a significant difference among the above 6 indexes (x2 =287.95, P < 0.05).The results of univariate analysis showed that the tumor location,surgical procedure, T stage, N stage, G stage, pathological stage and mediastinal lymph node metastasis were risk factors affecting abdominal lymph node metastasis of thoracic esophageal cancer (x2 =24.02, 23.97, 37.87,136.85, 38.79, 7.70, 154.27, P < 0.05).The tumor in the lower thoracic portion, N3 stage and stage Ⅳ were independent risk factors affecting abdominal lymph node metastasis of thoracic esophageal cancer in the multivariate analysis (RR =5.80, 2.36, 2.76, 95% confidence interval: 1.022-1.813, 1.317-3.950, 1.652-12.351, P < 0.05).Conclusions Abdominal lymph node metastasis is common in thoracic esophageal cancer in which No.16 and 17 lymph nodes predominate, and it is easy to occur in patients with lower thoracic esophageal cancer, and advanced N stage and pathological type.
10.Study on application of formative assessment and summative assessment in sanitary microbiol-ogy teaching

Objective To explore the new model of teaching assessment and evaluation of sanitary microbiology studied by students majoring in preventive medicine. Methods Combination of formative assessment and summative assessment was applied in the teaching for students in under-graduate preventive medicine class in Grade 2007-2010. To get better effect, method was improved constantly. Pearson correlation analysis was used to students' formative assessment and summative as-sessment, and comprehensive evaluation achievement(the first two added). Moreover, the effects were assessed by teachers' self-evaluation and their exchanging ideas with students. Results Ranging from 10.06 to 12.22, the standard deviations of four grades' summative assessment results were the biggest number, so the summative assessment was more effective. Students' formative assessment, summative assessment, and comprehensive assessment for this course were positively correlated(P<0.01) for suc-cessive four years. Teachers and students approved of the new mode. Conclusion The effective com-bination of formative assessment and summative assessment was helpful for student to develop and assess learning ability and overall qualities, and for teachers to improve teaching level.
