1.Establishment of radioligand binding assay of gastrin receptors in rat gastric parietal cells

AIM To establish radioligand binding assay of gastrin receptor in rat gastric parietal cell. METHODS Using 125 I labeled [Leu 15 ] gastrin 17 I as the radioligand, rat gastric parietal cells as the receptor preparations. RESULTS In the range of the study, a Scatchard plot of the binding was made by Ligand program with an equilibrium K d of approximately 1 249?10 -10 mol?L -1 and a maximum binding capacity of 4 4604?10 -12 mol?L -1 . The amount of gastrin binding was strongly associated with the cell concentration of rat gastric parietal cell preparations ranged from 0 25?10 9 to 2?10 9 cells?L -1 . The variant coefficent of the binding assay was 7 04% within the same sample. The competitive inhibiting analyses showed that 125 I gastrin was bound to gastric parietal cell preparations with a high specificity. CONCLUSION The radioligand binding assay of gastrin receptor in rat gastric parietal cell preparations meets the criteria for establishing receptor binding assay.

3.Clinical analysis of coronary artery bypass grafting:a report of 286 consecutive cases

Objective To evaluate the technique of coronary bypass grafting and its early result in 286 consecutive patients.Methods Two hundred and eighty-six patients from June 2002 to December 2005 underwent coronary artery bypass grafting.The average unmber of grafts was 3.3.Two hundred and eighty-three patients had left internal mammary artery bypassed to the left anterior descending branch.Other grafts were from saphenous veins.Results All patiens were clinical recovery.Respirator assisted within 0~28 hours(average 10).Conclusion Coronary artery bypass can be safely done with low mortality excellent surgical results even in patients with multi-vessel coronary artery disease.
4.Pathological Observation of Therapeutic Action of Kuijieling Decoction on Ulcerative Colitis Rats

Objective To observe the therapeutic effect of Kuijieling Decoction on experimental ulcerative colitis rats.Methods Ulcerative colitis rat model was induced by enteroclysis with trinitro-benzene-sulfonic acid(TNBS).Three dosages(18.3 g/kg,9.2 g/kg,4.6 g/kg)of Kuijieling Decoction were used to treat the model rats.After treatment,the serum of rats were obtained to detect the level of IL-1? by enzyme-linked immunosorbent assay.Then the rats were killed for the observation of the colonic pathologic changes by naked eye and for the estimation of the damage scores of colon mucosa.Finally,the colon tissue was fixed to make pathological section and the damage index of colon mucosa was observed under microscope.Results The tissue damage scores of high-and moderate-dosage groups of Kuijieling Decoction were(2.41? 0.79)and(2.83? 0.71)respectively,and were significantly lower than that of model control group(3.75? 0.75;P

[Objective] To observe the regulatory effect of astragalosides (AST) on the binding capacity of gastrin receptor in gastric parietal cells (GPC) of rats with spleen deficiency. [Methods] SD rats were given gastric gavage of Radix et Rhizoma Rhei (Dahuang) decoction 10 g/kg to induce spleen deficiency. Gastric parietal cells were isolated and purified by Lewin gastric sac method and Percoll density gradient separation method. 2 mL suspension of GPC from rat models was incubated in vitro with AST 20 ?L at the concentrations of 10, 20 and 30 g/L respectively. Suspension of GPC from rat models incubated with phosphate buffered saline (PBS) served as the model group and suspension of GPC from the normal rats incubated with PBS as the normal group. The binding capacity of gastrin receptor in GPC was detected by radioligand receptor assay. Achievements of the 30-year research work on Pi-Wei Doctrine were summarized. They are: (1) ratio of salivin activity and excretion rate of D-xylose in patients with spleen deficiency are different from those in the normal subjects;(2) the principle of syndrome differentiation and treatment is embodied in the methodology of herbal medicine pharmacology for complex prescription; (3) the above findings are further confirmed by the research in molecular and cellular level which indicates that TCM syndromes and pharmacological mechanisms of herbal medicines have their material basis and the regulatory effect of complex prescription on TCM syndromes is actually the pharmacological mechanism of herbal medicines. It is suggested that the research of herbal medicine should be based on the guidance of traditional Chinese medicinal theory and carried out in clinical practice.

BACKGROUND: Constipation, especially that caused by spleen insufficiency in transportation, often not responses to common purgatives, however,the therapy to strengthen the spleen and regulate qi so as to promote transportation and dredge stasis may be an ideal way to resolve the problem.OBJECTIVE: To investigate the effect of zhizhu decoction, a classic famous prescription, on gastric motility of mice with spleen insufficiency constipationDESIGN: A randomized controlled trial.SETTING: Department of Traditional Chinese Medicine (TCM) of Guangdong Medical College and Spleen-stomach Institute of Guangzhou University of Traditional Chinese Medicine.MATERIALS: The experiment was completed from June to December2002 at Spleen-stomach Institute of Guangzhou University of Traditional Chinese Medicine. Totally 328 NIH mice of general grade were chosen.METHODS: Mouse models with spleen insufficiency constipation were set up by irregular diet and overstrain combined with water deprivation. The influence of zhizhu decoction in large, middle and small doses on defecation in both spleen insufficiency mice and normal mice was investigated, taking Chinese patent drug bianmi tong (bowel-relaxing) as a positive control. In observation of normal mice defecation, there were five groups of large dose,middle dose and small dose of zhizhu decoction, bianmi tong and normal control. In observation of defecation of spleen insufficiency constipation mice, there were six groups: the above five and a natural recovering. In observation of loading mice trial, besides the above five groups, there was an atropine group. After administration, the time when the mice first defecated dark stool, the number of stool grains and the stool quantity were observe;and the percentage of small intestine advance was calculated.MAIN OUTCOME MEASURES: [1] Observation of the effect of zhizhu decoction on the defecation in both normal and spleen insufficiency mice.[2] The influence of zhizhu decoction on bowel advance in both normal and atropine loading miceRESULTS: Totally 328 mice were involved in the trial and 319 entered into the final result analysis, the other 9 died because of mistake in administration by gavage. The effect of zhizhu decoction on the defecation: [1] For normal mice, compared with the distilled group, in the zhizhu decoction groups of various doses the time when the mice first defecated dark stool was obviously reduced, and the number of stool grains and stool quantity were obviously increased, but the effect in the large and middle dose groups was best. [2] Compared with the normal group, in the model group the time when the mice first defecated dark stool was obviously increased, and the number of stool grains and stool quantity were obviously decreased. Zhizhu decoction in large and middle doses could obviously reduce the time when the mice first defecated dark stool, and increase the number of stool grains and stool quantity, the effect was better than that of the control drug bianmi tong. The influence ofzhizhu decoction on intestinal advance in mice: [1]Zhizhu decoction in various doses could obviously increase the rate of intestinal advance compared with distilled water group for normal mice. [2]Adding atropine load could reduce the rate of intestinal advance compared with the normal control group, and the percentage in all zhizhu decoction groups was obviously increased compared with the atropine group.CONCLUSION: Zhizhu decoction can reduce the time when the mice first defecated dark stool, increase the number of stool grains and stool quantity were obviously increased to both normal mice and mice with spleen insufficiency mice; it can also increase the intestinal advance rate in normal mice, and had a good antagonism against the inhibition of atropine to intestinal advance. In the trials the effect of zhizhu decoction in large and middle doses was more obvious, with a certain dose-effect relationship, the effect was better than that of the control drug bianmi tong.
