1.ACUTE TOXIC RESPONSE OF METRONIDAZOLE DERIVATIVE IN MICE
Academic Journal of Second Military Medical University 1985;0(05):-
Metronidazole derivative (CM) administered intraperitoneally or intravenously could cause a marked dose dependent neurotoxic response. The mean LD50 was 5034 mg/kg in the three batches of CM intraperitoneally administered and their effect was statistically identical, and the mean LD50 was 3186 mg/kg in the three baches of CM intravenously administered and their effect was same although there was a significant difference between 8638 and 8646 batches which was probably due to the individual difference in Sensitivity to intravenous injection.

In this experiment, we studied the effects of rhIL-3, rhlFN-r and DMSO on the growth and differentiation of the human leukemia cell line HL-60. The results showed that three agents were capable of suppressing the proliferation of the HL-60 cells and inducing terminal differentiation of the cells, with the exception of early augmentation of the cell growth by the rhIL-3. The differentiation effect was most obvious in DMSO treated cells, then in rhIL-3 and least in rhIFN-r The enhanced effects occurred when the HL-60 cells were treated in combination with DMSO and rhIL-3 or rhIFN-r

In this study, several physiological factors were studied for their effects on the growth and differentiation of U937 cells in vitro .The results showed that (1)rhTNF-?,PHA-LCM and c-RA exerted a dose and time-dependent suppression of proliferation of U937 cells; (2) the synergistic effect oc-cured when rhTNF-? was combined with c-RA in the cell culture; and (3)rhIL-1-? augmented the differentiation of U937 cells induced by c-RA though it had no effect on the cell line by itself. According to the [3H]-TdR incorporation, it can be concluded that the reduction of DNA synthesis of U917 cells by rhTNF-? ,PHA-LCM,and c-RA is attributed to the inhibition of the growth. The results might have clinical significance in the induction of differentiation of leukemia cells.

Objective To compare the clinical effects between rivaroxaban and warfarin on preventing deep vein thrombosis (DVT) in elderly hip fracture patients with diabetes mellitus (DM) after operation. Methods One hundred and sixty-eight elderly hip fracture patients with DM were randomly divided into the observation group and the control group. The observation group (88 cases) were treated with rivaroxaban (10mg, po, qd) after surgery, and the control group were treated with warfarin and low molecular weight heparin calcium for 14 days. Respectively after 7, 14, 21 and 28 days, the color doppler ultrasonography was used to examination vein blood flow and the DVT of affected lamb. In addition, the levels of D-dimer and bleeding were observed during treatment. Results The incidence of DVT in observation group (2 cases, 2.23%) was significantly lower than that in control group (7 cases, 8.75%), and DVT of control group was happened on 7th day after surgery. In addition, the level of D-dimer on 7th day post-surgery in observation group significantly decreased, while it significantly decreased on 14th day post-surgery in control group (P<0.05). However, the incidence of bleeding events in observation was less than that in control group (P<0.05). Conclusion Rivaroxaban can be more effective compared with warfarin in preventing diabetes mellitus elderly hip fracture patients with postoperative DVT formation and significantly reduce the bleeding risk, with more safe and convenient.

Objective To analyze the changes of the urine sodium and serum sodium in different types and different duration and different temperature in children with febrile seizures,to explore its clinical significance. Methods The children with febrile seizures(n =50)were selected as the group Ⅰ,and the children with fever but without seizure(n =50)were selected as the group Ⅱ,both of them were from pediatric ward. The control group(n =20)without fever or seizure was from Children's Health Section. All the children were checked serum sodium level and urine sodium and urine creatinine level. Results The sodium/creatinine ratio in the groupⅠ[(25.07 ±6.517)] was significantly higher than that in the group Ⅱ[(20.43 ±5.48)]and group Ⅲ[(14.88 ±5.11)](F =33.519, P <0.05),but there was no significant difference of the sodium/creatinine ratio in different ages and different types of the group Ⅰ,and there was negative relevance between the serum sodium and the sodium/creatinine ratio in the groupⅠ(r =-0.517,P <0.05).The mean serum sodium level in the groupⅠ[(133.50 ±3.14)mmol/L]was signifi-cantly lower than that in the group Ⅱ[(137.60 ±2.59)mmol/L]and group Ⅲ[(138.90 ±2.24)mmol/L](F =39.220,P <0.05),but there were no significant differences of the serum sodium in different types and different dura-tion and different temperature of seizure in the group Ⅰ.Conclusion The study shows that serum sodium is lower and the urine sodium is higher in children with febrile seizures, the changes of the urine sodium and serum sodium are not related to the types. The children with febrile seizures should be timely monitoring of serum sodium, urine sodium levels, and according to changes in the level of serum sodium, urine sodium, early appropriate supplement containing sodium liquid and maintaining the blood sodium to normal levels can reduce the recurrence of seizure.

China Oncology 2000;0(06):-
Purpose:To investigate the relation of cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) to tumor progression of cervical carcinoma. Methods:The expression and localization of COX-2 and iNOS protein in the 25 patients with cervical carcinomas were determined by immunohistochemical and the gene expression of COX-2 and iNOS were examined by reverse-transcription polymerase chain reaction ( RT-PCR). Results:Immunohistochemical staining for COX-2 and iNOS expression was strongly positive in 15 of 25 (60 %) and 20 of 25 (80 %) cases, respectively. Increased COX-2 and iNOS mRNA levels were confirmed by RT-PCR. There was negative correlation between COX-2 expression and tumor cell differentiation(r=-0.420, P

Chinese Pharmacological Bulletin 1986;0(06):-
AIM To establish radioligand binding assay of gastrin receptor in rat gastric parietal cell. METHODS Using 125 I labeled [Leu 15 ] gastrin 17 I as the radioligand, rat gastric parietal cells as the receptor preparations. RESULTS In the range of the study, a Scatchard plot of the binding was made by Ligand program with an equilibrium K d of approximately 1 249?10 -10 mol?L -1 and a maximum binding capacity of 4 4604?10 -12 mol?L -1 . The amount of gastrin binding was strongly associated with the cell concentration of rat gastric parietal cell preparations ranged from 0 25?10 9 to 2?10 9 cells?L -1 . The variant coefficent of the binding assay was 7 04% within the same sample. The competitive inhibiting analyses showed that 125 I gastrin was bound to gastric parietal cell preparations with a high specificity. CONCLUSION The radioligand binding assay of gastrin receptor in rat gastric parietal cell preparations meets the criteria for establishing receptor binding assay.
