1.Determination of Aflatoxins Content in Traditional Chinese Medicine Using Immunoaffinity Column Cleanup and by High Performance Liquid Chromatography

Objective To develop a method for the determination of aflatoxins B1,B2,G1 and G2 in Chinese herbal medicine.Methods The herbal medicine samples were cleaned up on immunoaffinity columns and analyzed by high performance liquid chromatography(HPLC) with fluorescence detection.For chromatographic separation,a Zorbax SB C18(4.6 mm?150 mm,5 ?m) column was employed.The separation was carried out as the mobile phase of methanol-0.01 mol/L KH2PO(44+6)with a flow rate of 0.5 ml/min at column temperature of 35 ℃.The reaction tube temperature of postcolumn derivatization system was 70 ℃.The detection was observed by fluorescence with excitation at 360 nm and emission wavelength at 425 nm.Results The calibration curve showed good linearity in the range of 12.00-300.00 ?g/L for aflatoxin B1,and 24.00-600.00 ?g/L for aflatoxin B2,G1 and G2.The lowest limits of detection for aflatoxins in Chinese herbal medicine were 0.025 ?g/L for AFB1,0.085 ?g/L for AFB2,0.060 ?g/L for AFG1,and 0.055 ?g/L for AFG2,and the correlation coefficients were ≥0.996.Recoveries were 81.7%-101.2% for aflatoxin B1,B2,G1 and G2 spiked to Fructus amomi,Radix angelicae sinensis,Rhizoma polygonati,Rhizoma atractylodis macrocephalae and Rhizoma dioscoreae at the level of 5 ?g/L,and the relative standard deviations were 0.7%-4.9% in all instances.Conclusion The method has good stability,high sensitivity and high selectivity,and it is applicable to the determination of aflatoxins in Chinese herbal medicine.
2.The clinical application of endoscopic technique on maxillofacial benign tumor

OBJECTIVE To explore the method and effect of endoscopic technique on maxillofacial benign tumor.METHODS Ten patients visiting our hospital from January to December 2007 and diagnosed as maxillofacial benign tumor were recruited in this study.The tumor of the patients were composed of 4 sebaceous glands,3 liparomphalus,3 epidermoid cysts.All of them were treated with endoscopic technique.The surgical approach was all in hair line or behind ear lobe and the incision was about 1cm. Subcutaneous tunnel was prepared.The operations were guided by the endoscope and helped with selfmade tritus skin expandor.RESULTS The operations were performed with an average duration of 45 minutes. Patients were all satisfied with the post operative results. CONCLUSION Endoscopic technique could well be applied in surgery of maxillofacial benign tumor.
3.Wear matching performance of dental restoration materials oppositing to dentine

Objective:To compare the wear resistance between dentin and dental restoration material in vitro.Methods:The friction and wear behaviors of natural tooth dentin respectively against highly polished polymer ceramic,glass ceramic and zirconia were investigated in an artificial saliva test environment by UMT-2 friction and wear testing machine.Worn surface morphology was observed by scanning electron microscopy(SEM),roughness was measured by roughness instrument,the rigidity value was weighed by electronic balance.Results:The roughness of the 3 materials was similar to that of dentin(P > 0.05),the rigidity of dentin was less than that of the 3 materials (P < 0.05).After friction and wear test,the abrasion quantity of dentin respectively against the 3 materials was less than that of the materials (P < 0.05).There was positive correlation between the wear loss and the hardness of the 3 materials and dentin(r =0.846).The mass loss of the dentin against Poly Ceramic after grinding abrasion was the closest among the 3 materials.Conclusion:Different materials have varying degrees abrasion against dentine.The Polymer Ceramic has closer abrasion performancez with dentin than the other 2 materials.
4.Latent tuberculosis infection and tuberculosis incidence college freshmen in Chaoyang District during 2013 to 2015

Objective:
To understand the prevalence of latent tuberculosis infection (LTBI) among college freshmen in Chaoyang District of Beijing from 2013 to 2015, and to provide evidence for prevention and control of tuberculosis in schools.
Methods:
The latent tuberculosis infection was identified through a tuberculin purified-protein derivative (TB-PPD) skin test. TB-PPD test results of 65 233 college students in the Chaoyang District during 2013 to 2015 were collected and stored, by using descriptive statistical analysis.
Results:
Among the 65 233 freshmen, 63 135 were effective PPD test results, 11 754 were PPD positive, 2 720 were PPD strong positive, the LTBI status: the PPD positive rate was 18.62% and the PPD strong positive rate was 4.31%. The positive rate and the strong positive rate in male (19.56%, 4.76%) were higher than that in female(17.87%, 3.95%)(χ2=29.37, 24.29), in ethnic minorities were higher than that in Han Nationality (18.25%,4.16%) and in Tibetans (29.45%, 8.69%) were the highest(χ2=102.01, 70.04), among freshmen from the western region (21.88%, 5.11%)were the highest and from Beijing city (16.25%, 3.46%) were lower (χ2=220.63, 99.03, P<0.01), among those with BCG scar (18.94%, 4.34%) are slightly higher than those without BCG scar(16.96%, 4.08%) (χ2=48.46, 10.43, P<0.01). 19 cases of tuberculosis patients were found, the prevalence rate was 29.13/100 000 (19/65 233), of which 68.42% (13/19) were found through the strong positive for PPD. Among the strong positive students (2 707), there were 1 433(52.94%) who received preventive therapy, and during the follow-up two years, 7 were newly diagnosed with tuberculosis, the annual incidence rate was 129.29/100 000 (7/2 707).
Conclusion
Freshmen from high-risk areas of tuberculosis are the focus of tuberculosis screening, for which we should continue to carry out PPD screening. Those with latent tuberculosis infection are high-risk groups of tuberculosis, and should be paid more attention in the prevention and control of tuberculosis in schools.
5.Comparison of genotype distribution of patients with acute hepatitis B infection or chronic hepatitis B infection in Shanghai

Objective To investigate the distribution of genotypes in chronic HBV infection (CHB) and acute HBV infection (AHB) patients in Shanghai. Methods Sixty-two patients with AHB and 73 patients with CHB admitted to ('hanghai Hospital of Shanghai between 2003 and 2007 were studied. Viral genotypes of all the patients were determined by direct gene sequencing.Meanwhile, epidemiological, clinical and biochemical parameters of all patients were collected. Mean values of different groups were compared by t test while frequency was compared by chi square test. Results The major prevalent genotypes in both AHB and CHB patients were genotype B and C (48.4% vs 51.6% in AHB patients and 26.0% vs 74.0% in CHB patients). The proportion of genotype B was higher in AHB patients compared to CHB patients (P= 0.02). Epidemiological factors and clinical outcomes were not statistically different among patients with different viral genotypes. The proportion of genotype C was much higher in CHB patients compared to AHB patients (P=0.006). The main transmission route of AHB was heterosexual interaction which was 18 out of 62 (29.0%), but in CHB patients, it was prenatal transmission which was 38 out of 73 (52.1%). Conclusions In shanghai, the main HBV genotypes in both AHB and CHB patients are genotype B and C. The proportion of genotype B is relatively high in AHB patients while proportion of genotype C is more common in CHB patients. There is no significant relationship between genotypes and the clinical outcomes of AI-IB patients.
6.The surface structure and antibacterial property of Ag-containing coating on casting cobalt chromium alloy

Objective:To study the surface structure and antibacterial property of Ag-containing coating(AgCC)on casting cobalt chromium alloy(Co-Cr).Methods:Plasma spraying technique was adopted to prepare AgCC on Co-Cr.Scanning electron microsco-py,energy dispersive analysis and X-ray diffraction analysis were used to evaluate the surface properties.The antibacterial effects of AgCC against staphylococcus aureus and candida albicans were examined by film attachment method.Fluorescence microscope was a-dopted to evaluate the bacteria adhesion property.Results:AgCC surface was uniform and compact,combined perfectly with sub-strate material.The content of the surface was mainly Ag,Cr and a small amount of Ag2 O and Cr2 O3 .The results of antibacterial as-say showed that on AgCC the number of bacterial colony was fewer(P<0.05)and bacterial adhesion was less than those on Co-Cr(P<0.05).Conclusion:AgCC surface was stable and has antibacterial property.
7.The prognostic value of age for 1030 patients with early stage breast cancer after postoperative radiotherapy

Objective To analyze the prognostic value of age in patients with early stage breast cancer. Methods The clinical characteristics of 1030 patients with early stage breast cancer (the number of positive axillary lymph nodes was less than 3) were retrospectively reviewed. Of all the patients, 468(stage Ⅰ, n = 227; and stage Ⅱ , n = 241) received breast conserving surgery (BCS) and 562 (stage Ⅰ, n =184; and stage Ⅱ, n= 378) received modified mastectomy. Patients were divided into young-age group (≤35,136 patients), middle-age group (> 35-≤60,738 patients) and old-age group (> 60,156 patients).The number of patients without postoperative radiation therapy after BCS is 16, 60 and 39 in the three groups, respectively. Two-dimensional conventional fractionated radiotherapy was administered. The prognostic value of the tumor size, status of axillary lymph nodes or hormonal receptors, postoperative radiation therapy were analyzed. Results The follow-up rate was 97.86%. Of 795 patients followed up more than 5 years, 110,569 and 116 patients were devided into the three groups, respectively. There were 40, 202 and 87 patients without radiation therapy in the three groups. The 5-year recurrence rates of the three groups were 6. 2%, 8. 7% and 10. 4% (χ2 = 1.14, P= 0.567). The 5-year distant metastasis rates were4.3% , 9.5 % and2. 5% (χ2 = 5.31 , P = 0. 070) . The5 - year survival rates were9l. 2% , 92. 6%and 82. 1% (χ2 = 6. 83, P = 0.033). The young-age group had more tumors smaller than 2. 0 cm (65.4%), less positive axillary lymph nodes (13.2%), poorer differential tumor and less positive hormone acceptors (48.0%). Of patients with tumor larger than 2. 0 cm who had no radiotherapy after BCS, the 5-year survival rates were 94%, 87% and 71% (χ2= 20.69, P= 0.000) in the three groups. The corresponding recurrence rates were 23%, 18% ,7%, (χ2 = 9. 97, P = 0. 007), and distant metastasis rates were23%, 25% and 10% (χ2 =8.51, P=0. 014). Conclusions The age is an important prognostic factor in patients with early stage breast cancer undergoing BCS, but not in those undergoing modified mastectomy.
8.Effects of four dental alloys on apoptosis related gene and protein expression of fibroblast L929.

OBJECTIVETo investigate the effects of the leaching liquids of 4 differents kinds of dental alloys (Au alloy, Ag-Pd alloy, Co-Cr alloy, Ni-Cr alloy) on apoptosis related gene and protein of fibroblast L929.
METHODSThe L929 cells of mouse were treated in vitro with leaching liquids of 4 different kinds of dental alloys, Au alloy (group A), Ag-Pd alloy(group B), Co-Cr alloy(group C) and Ni-Cr alloy(group D). The RPMI1640 cell medium containing 10% fetal calf serum was served as a control(group E). The effects of these alloys on the expression of caspase-3, 8, 9 of L929 cells were examined by reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry method. Results After 48 h culture, the mRNA levels of caspase-3 and caspase-9 demonstrated significant differences between the groups expect group A and group E. The mRNA levels of caspase-8 had no change in all groups. The expression of caspase-3 and caspase-9 were significant differences between the groups expect group A and C, group B and D. The expression of caspase-8 had no change in all grotps.
CONCLUSIONThe leaching liquids of 4 different kinds of dental alloys expect Au alloy may induce cell appotosis through mitochondrion pathway.

9.Effects of M-CSF concentration, RANKL concentration and M-CSF preinduction on osteoclastogenesis.

This study was directed to the effects of macrophage-colony stimulating factors (M-CSF) concentration, recerptor activator of nuclear factor kappaB ligand (RANKL) concentration and M-CSF preinduction on osteoclastogenesis and the related resorption function. Bone marrow mononuclear cells were isolated and were divided into 4 groups. Group A underwent osteoclastogenic induction with the use of 30 ng/ml M-CSF and 50 ng/ml RANKL, while Group B received 50 ng/ml M-CSF and 100 ng/ml RANKL treatment. Both C and D Group underwent preinduction with the use of 30 ng/ml M-CSF for 3days, and then they were treated with 30 ng/ml M-CSF and 50 ng/ml RANKL, 50 ng/ml M-CSF and 100 ng/ml RANKL, respectively. Osteoclastogenesis was examined by TRAP staining 6 days after induction, and dentin resorption lacunae were detected by Scanning Electron Microscope 9 days after induction. TRAP positive multinuclear cells were observed in all groups of cells, and resorption lacunae were formed in all of them. However, more TRAP positive multinuclear cells were observed and more large resorption lacunae were detected in groups B and D than in groups A and C, respectively. The number of TRAP positive cells, number of resorption lacunae and lacuna areas in groups C and D were also greater than those in groups A and B, respectively. Higher concentration of M-CSF and RANKL and preinduction with M-CSF may benefit osteoclastogenesis and increase resorption function of osteoclast.

10.The biological safety and biocompatibility evaluation of the mixture of Paris polyphylla-chitosan for denture adhesive

Objective:To study the biological safety and biocompatibility of the mixture of Paris polyphylla-chitosan.Methods:According to the GB/T 16886.12-2005 standard,YY/T 0279-1995 standard and GBT16886.5-2003 standard,samples were prepared and tested by oral mucous membrane irritation test,cytotoxicity test and flow cytometry.Results:No local response to the mixture of Paris polyphylla-chitosan was found,and the visual observation and pathological findings of oral mucosa were normal and similar to that of the control group.Therefore,the mixture of Paris polyphylla-chitosan had no irritation response to oral mucosa.The mixture of Paris polyphylla-chitosan showed no cytotoxicity to L929 cells,and did not affect the cycle distribution and apoptosis of L929 cells.Conclusion:The mixture of Paris polyphylla-chitosan has good bio-safety and biocompatibility.
