1.Expression and Significance of Anti EZH2 Autoantibodies IgG in Hepatocellular Carcinoma Patient's Serum Samples

Objective To detect the anti EZH2 autoantibodies IgG expression in hepatocellular carcinoma patient's serum samples and investigate the relationship of anti EZH2 autoantibodies IgG expression with clinicopathological parameters.Methods ELISA method was used to detect the anti EZH2 autoantibodies in serum of 120 cases of HCC patients and compared with 115 healthy controls.Results Serum anti EZH2 antibodies in patients with hepatocellular carcinoma group the content of IgG was 2.015±0.568 μg/ml group,anti EZH2 antibodies IgG content control health was 1.185±0.289 μg/ml.There was significant difference between two groups (t=12.48,P<0.001).Anti EZH2 antibodies of IgG in serum of pa ticnts with hepatocellular carcinoma was up-regulated and the degree of tumor size,tumor the degree of differentiation,TNM grading and intrahepatic metastasis (t=8.485 ~ 57.93,P<0.001).And age,sex,drinking,whether HBV infection had no significant difference (t=1.22~5.20,P>0.05).Conclusion The expression of anti EZH2 autoantibodies in serum was related to the occurrence and development of hepatocellular carcinoma,and the detection of serum anti EZH2 autoantibodies in serum of liver cancer may be helpful to judge the malignant behavior and prognosis of hepatocellular carcinoma (HCC).which may be used as a refcrcnce for the evaluation of the prognosis of HCC patients with IgG antibody against IgG.
3.The correlation between granulomatosa cheilitis and odontogenic infectious foci

Clinical data of granulomatosa cheilitis of 33 cases were analysed.Odontogenic infectious foci were found in each patient.Elimination of odontogenic infectious foci was followed by regression or disappearance of the swellings in all the 33 cases,suggesting a close relationship between granulomatous cheilitis and odontogenic infectious foci.Elimination of odontogenic infectious foci can be an important measure for the treatment of granulomatous cheilitis.
4.Expression of serum anti-IMP1 autoantibody IgG in patients with hepatocellular carcinoma and itsrelationship with clinicopathological parameters

Objective To investigate serum anti-insulin-like growth factor-Ⅱ mRNA binding protein1(IMP1) autoantibody IgG expression in hepatocellular carcinoma(HCC) patient and its relationship with clinicopathological parameters.Methods ELISA method was used to detect serum anti-IMP1 autoantibody IgG in 120 cases of HCC,then the detection results were compared those in 115 healthy individuals(control group).The relationship between serum anti-IMP1 autoantibody IgG level with clinicopathological parameters in HCC patients was analyzed.Results The serum anti-IMP1 autoantibody IgG levels in the HCC group and control group were(1.512±0.685),(1.080±0.301)μg/mL respectively,the HCC group was significantly higher than the control group with statistical difference(P<0.001);serum anti-IMP1 autoantibody IgG in HCC patients was related with the tumor size,tumor differentiation degree,TNM grade and intrahepatic metastasis(P<0.001),and had no obvious relation with the age,sex,drinking and HBV infection(P>0.05).Conclusion Serum anti-IMP1 autoantibody IgG expression increased is increased in HCC patient.The detection of serum anti-IgG autoantibody IMP1 level in HCC is conducive to judge the malignancy degree of HCC.
5.Prepare and clinical application of HCV genotyping oligochip

Objective To study the preparation of hepatitis C viruses (HCV) genotyping oligochip and its application in the detection of 76 hepatitis C patients.Methods Oligonucleotide probes and primers were designed in the 5’noncoding region and core region of HCV. The HCV typing chip was prepared by spotting the modified probes onto nylon membrane. Products of the second PCR were labeled with Dig-dUTP. Furthermore, 6 PCR products were sequenced.Results Using the chip,15 subtypes in 11 types of HCV were analyzed.Results of hybridization indicates that 76 hepatitis C patients were all positive and 20 health people were negative.Among 76 patients, 64 cases were 1b type, 11 cases were 2a type and 1 case was 3a type. Mix infection was not found. The results obtained by sequencing 6 samples and chip arraying were the same.Conclusion The HCV genotyping chip could be used in detecting serum HCV RNA and analyzing its genotypes.
6.Identification of Codonopsis Radix and Its Adulterants Using the ITS/ITS2 Barcodes

The ITS/ITS2 barcodes were used to simply and effectively identify Codonopsis Radix and its adulter-ants. In this study, ITS (internal transcribed spacer of unclear ribosomal DNA) regions were amplified using PCR (polymerase chain reaction) from thirty-three samples of Codonopsis Radix and ITS2 regions were obtained from the ITS sequences using the hidden Markov model (HMMer)-based annotation methods. The sequences of ITS/ITS2 regions were aligned and the genetic distances were computed by MEGA5.0. Species identification efficiency of ITS/ITS2 sequences were evaluated using BLAST1 and nearest distance methods. The results indicated that The sequences lengths of ITS regions of Codonopsis Radix were 654-655 bp, and the lengths of ITS2 regions were 239 bp. The intraspecific genetic distances among Codonopsis Radix were obviously lower than the interspecific genetic distance between Codonopsis Radix and its adulterants. Therefore, ITS/ITS2 regions can stably and accu-rately distinguish Codonopsis Radix and its adulterants.
7.One-stage urethroplasty using circular fasciocutaneous preputial skin flap for the treatment of complex anterior urethral strictures

Objective To evaluate the efficacy of 1-stage urethroplasty using pedicle circular fascioctaneous preputial flap for the treatment of complex anterior urthral strictures.Methods Between January 2006 and January 2013, 37 patients with complex anterior urethral stricture were treated by 1-stage urethroplasty using pedicle circular fascioctaneous preputial flap.The mean age was 41 years ( 22 -71 years) .The etiology of stricture included trauma of 13 cases, iatrogenieity of 13 cases, gonorrhea infection of 2 cases, unknown reason of 9 cases.The penile urethral stricture was found in 22 cases, the bulbourethral stricture in 9 cases, and stricture extending from penile to posterior urethra in 6 cases.The mean length of anterior urethral stricture was 8.1 cm (range 5.0-14.0 cm).A circumferential island of the preputial/distal penile skin was mobilized by the technique of preserving penile fasciocutaneous wide vascular pedicle. The pedicle is composed of two layers of the dartos and the superficial lamella of Buck′s fascia, and the flap was divided in the midventral/middorsal plane back to the penoscrotal junction to convert the circular configuration to a longitudinal trip for urethral reconstruction.The dorsal and ventral inlaid flap urethroplasty was performed in 27 cases and tubularized flap urethroplasty was performed in 10 cases.Results The mean operative duration was 3.1 h (2.5-3.5 h).The mean length of the circular fascioctaneous preputial flap was 10.4 cm (range 9.0 -14.0 cm).All the patients were followed up for mean 22 months (3 -51 months).Thirty-two cases voided well and the mean peak urinary flow rate was 22.3 ml/s (15.0-29.0 ml/s).One-stage healing achieved in 32 cases (86.5%).Recurrent stenosis was noted in 4 cases, and meatal stenosis occurred in 1 patient, who required re-operation.Re-repair succeeded in 4 cases and total success rate was 97.3% (36/37).Conclusions The pedicle circular fascioctaneous preputial flap has advantage of good blood supply and autograft for new meatus.It could be a reliable and durable method for the treatment of complex anterior urthral strictures(≥5 cm) in 1-stage urethroplasty.
8.Practice and prospect of radiotherapy and immunotherapy for lung cancer

In recent years, immunotherapy has become the hottest topic in the field of oncology. Both the Keynote189 study and the Keynote407 study have confirmed that progression-free survival is significantly prolonged in patients who have been benefited from immune checkpoint blockades in lung cancer. In an article published in The New England Journal of Medicine in 2012, a case report of radiation abscopal effects caused by immunization combined with conventional radiotherapy has attracted great attention in the field of oncology. The Pacific study, published in 2017, expanded the indications for immunotherapy from advanced to locally-advanced non-small cell lung cancer. The second analysis of Keynote001, published in the Lancet Oncology in the same year, suggested that radiation therapy may mediate the immune memory effects, whereas the mechanism and time window are still unclear. With the publication of PEMBRO-RT study and several pieces of work by our team in recent years, various details of radiotherapy combined with immunotherapy (iRT) have become more mature. In clinical practice, iRT is involved in the full treatment of lung cancer. However, iRT is not a hodgepodge or stew that needs further refinement and sorting. In this article, the principles, efficacy in clinical practice, and exploration of the details of iRT were discussed.
9.Status of screening myopia among primary and middle school students in Linfen Community of Shanghai from 2019 to 2023

Objective:
To analyze the prevalence and trend of screening myopia among primary and middle school students in Linfen Community of Shanghai from 2019 to 2023, so as to provide a reference for the prevention and control of myopia from the perspective of the community.
Methods:
From 2019 to 2023, all primary(5) and middle(2) school students aged 6-15 years in Linfen Community of Shanghai were screened. Statistical analysis was performed using the Chi square test and trend Chi square test. The curve fitting model was used to fit the model of the increase rate of screening myopia among primary and middle school students in 2019, 2021 and 2023.
Results:
The overall rate of screening myopia among primary and middle school students in Linfen community from 2019 to 2023 was 55.17%. The prevalence rate of screening myopia was 79.43% in boys and 81.92% in girls in middle school, and the difference was statistically significant ( χ 2=5.71, P =0.02). In 2019, 2021, and 2023, the peak age of screening myopia among primary and middle school students in Linfen Community gradually occurred earlier, at the age of 7(12.13%), 6( 12.28 %), and 6(14.99%) years old, respectively. The growth rate of screening myopia in students aged 8-12 years in 2023 was lower than that in 2019 and 2021.
Conclusions
The screening myopia rate of primary and middle school students aged 6-15 years in Linfen Community is relatively high, with primary school girls higher than boys, and growth spurt accelerates. It is suggested that prevention and control of myopia in the community should focus on preschool children and adolescent girls.
10.Analysis of transcriptional factors expression profile during dedifferentiation of liver cell cultured in vitro.

Primary hepatocytes are widely used in drug metabolism and toxicity assessment. As the culture of primary hepatocytes in vitro is a process of dedifferentiation, hepatocytes lose normal metabolic detoxification function gradually. The mechanism of hepatocyte dedifferentiation has been not clear so far. TFs play an important role in the dedifferentiation and non-parenchymal cells can maintain the function of hepatocytes in vitro. However, the current methods cannot be used in effective identification and quantitative analysis of a large number of TFs. In this paper, the mo-culture system (only primary hepatocytes) and co-culture system (primary hepatocytes and non-parenchymal cells) were established. The cells were cultured for 24 h, 48 h, 72 h as monolayer. The changes of TFs during the culture were obtained by TOT (Transcription factor response elements on tip) transcription factor enrichment method and mass spectrometry. A total of 219 TFs were identified in three individual replicates. The result revealed that up-regulated TFs were enriched in cell proliferation, death and immune response pathways, and down-regulated TFs were involved in metabolism pathway. The establishment of such culture-TFs identification system is of great significance to reveal the mechanism of primary hepatocyte dedifferentiation and crosstalk between hepatocytes and non-parenchymal cells.
