1.Studies on In-Vitro Biological Activity of Albiflorin R1 in Counteracting Cell Proliferation

[Objective] To study the in-vitro biological activity of albiflorin R1 (AR1), a new monoterpene glycoside from Paeonia lactiflora,in counteracting cell proliferation. [Methods] Two kinds of human cancer cell strains (HeLa cells and K562 cells) and J774 macrophage cell strain were cultured in vitro to observe the effects of albiflorin Rl on cell proliferation. [Results] Albiflorin Rl could markedly inhibit the proliferation of J774 macrophages and also inhibit the proliferation of HeLa and K562 cell stains in vitro. [Conclusion] Albiflorin Rl may have an anti-neoplastic, anti-inflammatory and immunosuppressive activity.

3.Studies on Anti-inflammatory Mechanism of Sinomenine

【Objective】The effect of sinomenine on human cyclo-oxygenase(COX) and interleukin-1?(IL-1?) was observed to explore the anti-inflammatory mechanism of sinomenine.【Methods】Peripheral blood monocytes(PBMC) were isolated and incubated with sinomenine.After incubation,total RNA from PBMC was extracted and the effect of sinomenine on COX and IL-1? expression with or without the stimulation of lipopolysaccharide(LPS) was observed by the method of semi-quantitative reverse transcription polymerase chain reaction(RT-PCR).【Results】Sinomenine had no effect on COX-1 gene expression,had a slight inhibitory effect on COX-2 gene expression and had a strong inhibitory effect on IL-1? gene expression.【Conclusion】 The anti-inflammatory mechanism may be related to the inhibition of inflammatory cytokines.
4.Correlation between plasma homocysteine levels and behavioral and psychological symptoms in patients with mild to moderate vascular dementia

Objective To explore the correlation between serum homocysteine(Hcy) levels and behavioral and psychological symptoms dementia (BPSD) in patients with mild to moderate vascular dementia (VaD).Methods Two hundred and ninety-three patients with mild to moderate vascular dementia (aged 40 or over) admitted to the department of neurology of the First Affiliated Hospital of Liaoning Medical College from January 2012 to January 2015.Patients were diagnosed with dementia by NINDS-AIREN criteria,MMSE scores ≤26,Hachinski ischemic scale(≥4) and clinical dementia rating(1≤ C DR ≤ 2).VaD patients were divided into high Hcy(HHcy) group (Hcy≥ 15 μ,mol/L,n=188) and control group(Hcy<15 μmol/L,n=105).The total scores and the scores of 12 functional domains of behavioral and psychological symptoms in NPI were analyzed by using comparative statistical methods.Results Prevalence of high homocysteine was 64.16% (n=188) among mild to moderate VaD (n=293).The incidence of BPSD in HHcy group(80.9%) was significantly higher than that in the control group (57.1%),the difference was statistically significant (x2=18.932,P<0.01).HHcy patients (27.95±5.04) had a significantly higher total score of NPI compared with control patients (16.87± 1.87),the difference was statistically significant (t=3.753,P<0.01).In terms of scores in 12 functional domains in NPI,the scores in sleep disorders (2.99± 1.40),high (2.10±0.53),irritability/mood swings (2.64± 1.43),abnormal behavior (1.74±0.52),disinhibition (1.40±0.43),agitation (2.02±0.74) were higher than those in control groups (1.85±0.37,0.21±0.05,1.80 ±0.56,0.36±0.09,0.45±0.07,0.68±0.23),all the difference were significant(t=2.327,t=2.012,t=2.136,t=2.066,t=2.050,t=2.007,all P<0.05).Binary logistic regression analysis showed that there were positive correlation between scores of NPI and plasma Hcy levels (OR=1.164,95% CI:1.052-1.288,P=0.003).Compared with HHcy group and control group,there were no statistical significance in regard to infarcted brain focus including the frontal lobe,parietal lobe,temporal lobe,occipital lobe,basal ganglia,brain stem and cerebellum (x2=0.528,x2=0.043,x2=0.630,x2=0.166,x2=0.657,x2=1.010,x2=0.019,allP>0.05).Conclusion High homocysteine levels are correlated with behavioral and psychological symptoms in patients with mild to moderate VaD,including performance of sleep disorders,high,irritability/mood swings,abnormal behavior,disinhibition and agitation.Moreover,the severity of BPSD is positively associated with homocysteine levels.The higher the homocysteine level,the more severe the BPSD.The serum homocysteine levels are no correlated with infarcted brain focus.
5.Effect of multiple IPL treatment on cell senescence markers in skin fibroblasts

Objective To study the effect of multiple IPL treatment on cell senescence markers of skin fibroblasts using UVA as a control and to make clear whether the multiple IPL treatment may result in cell senescence.Methods Cells were divided into three groups: one group without irradiation as a control,one group receiving IPL treatment with 15 J per cm2,and the last group receiving UVA irradiation with 9 J per cm2.IPL and UVA irradiation were performed once a day during five days.On the sixth day,the cells were collected.Senescence-associated β-galactosidase (SA-β-Gal) staining,cell cycle,reactive oxygen species (ROS) and telomere length were determined.Results Our results showed that five consecutive days of IPL irradiation had no effect on the activity of SA-β-Gal and telomere length and decreased the G1 % of cell cycle and the level of ROS in comparison with the control group (P<0.05).On the contrary,five consecutive days of UVA irradiation increased the activity of SA β Gal and the level of ROS,shortened the length of telomere and no obvious change in the G1 % of cell cycle in comparison with the control group.Conclusions Multiple UVA irradiations induce cell senescence.On the contrary,multiple IPL treatments could not induce cell senescence.
6.RAPD Analysis of Pogostemon cablin from Different Habitats

Objective To compare the genetic difference of Pogostemon cablin from different habitats.Methods Forty-six primers were used to analyze the fingerprint of Pogostemon cablin by Random Amplified Polymorphic DNA,and 6 of them were identifiable.Primer S358 could reflect the tiny polymorphic differences of Pogostemon cablin from different habitats which had 3 same amplified strips.Primers S358,S359,S443 could be used for the identification of Shipai sample from its other species.Results The results of cluster analysis showed that the two samples from Gaoyao were classified as one type,the four samples from Zhanjiang,Hainan,Shipai as another type,one sample from Caizhilin and one from the pharmacy of the First Affiliated Hospital of Guangzhou University of TCM as the the third type,and the sample from the Qingping Traditional Chinese Herbal Market as the fourth type.Conclusion The RAPD analysis can be used for the identification of Pogostemon cablin from different habitats.
7.Effect of Naringin on Proliferation and Differentiation of 3T3-L1 Preadipocyte

Objective To observe the effect of naringin on proliferation and differentiation of 3T3-L1 preadipocyte,and to investigate its possible mechanism.Methods The cultured 3T3-L1 cells were pretreated with naringin,and then MTT method was used to detect the proliferation of the cells,oil red O staining method and spectrophotography were applied to analyze the degree of differentiation.The expression of PPAR? 2 and C/EBP? mRNA associated with adipocytes differentiation were detected by reverse transcription PCR(RT-PCR).Results Naringin suppressed the proliferation and differentiation of 3T3-L1 preadipocytes and down-regulated the expression of PPAR? 2 and C/EBP? mRNA.Conclusion Naringin can inhibit the proliferation of 3T3-L1,and can suppress their differentiation through down-regulating the expression of PPAR? 2 and C/EBP? mRNA.
8.Expression of and significance of RORγt and IL-17 in newborn mice with cyto-megalovirus infection

Objective:By comparing the expression levels of retinoic acid-related orphan nuclear receptorγt( RORγt) and in-terleukin-17 ( IL-17 ) in newborn rats′spleen tissue with cytomegalovirus infection with normal newborn to provide experimental evidence for the pathogenesis of CMV infection.Methods:Forty-eight newborn BALB/c mice were randomly divided into control group and MCMV group.Mice in virus group was given intraperitoneal injection of MCMV virus suspension,while the control group were given the same dose of normal saline as controls.Eight mice in the two groups were killed at day 3,7 and 14.The animal were sacrificed at day 3,7 and 14( n=8 for each interval) and spleens were obtained from the two groups.MCMV DNA,RORγt mRNA,IL-17 mRNA and protein of RORγwere detected by using RT-PCR and Western blot between the two groups.Results:Expression of MCMV DNA was in-creased in the MCMV group but absent in the control group.RORγt mRNA, IL-17 mRNA and protein expression of RORγt were significantly higher than the normal control group,with the extension of the infection time gradually increased( P<0.05 ) .Conclusion:The IL-17 and RORγt in spleen tissue may take part in the inflammatory response induced by MCMV, and may be involved in the pathogeneses of MCMV injury.
9.Expression of SOX10 in cerebral white matter in immature rats with postnatal infections

Objective To explore the effect of postnatal infection on SOX10 expression in cerebral white matter in immature rats. Methods A total of 96 newborn SD rats were randomly divided into hypoxia group, lipopolysaccharide (LPS) group, and control group. At day 3 and 6 after birth, the rats in LPS group and hypoxia group were intraperitoneally injected with 0.25 mg/kg of LPS while the rats in control group were injected with normal saline. Meanwhile the rats in hypoxia group were maintained in a hypoxic tank under atmospheric pressure and thermostatic water bath at 37℃for 2 hours of ventilation with mixed gas con-taining 8%O2 and 92%N2 at a rate of 2 L/min starting 3 days after birth. At day 7, 10, 14, 21 after birth, eight rats in each groups were sacriifced and the cerebral white matter was extracted. HE staining was performed to observe the pathological changes of cerebral white matter by light microscopy. The expression of SOX10 in cerebral white matter was determined by immunohisto-chemical and Western blotting analysis. The expression of TLR-4 was determined by Western blotting. Results In LPS group and hypoxia group, the SOX10 positive cells and expressions of SOX 10 and TLR-4 were increased at day 7, reached the peak at day 10, and then gradually declined. There were signiifcant differences between any two time points (P<0.05). In control group, there were a few positive cells and limited expressions of SOX 10 and TLR-4 and there were no differences between any two time points (P>0.05). At each time point, the difference in the SOX10 positive cells and the expressions of SOX 10 and TLR-4 were statistically signiifcant among three groups (P<0.05) in the order of hypoxia group>LPS group>control group and there were signiifcantly differences between each groups (P<0.05). Conclusions Postnatal infections can lead to cerebral white mat-ter lesions in immature rats. The existence of both hypoxia and infection can aggravate the brain injury. The high expression of SOX 10 may have the protective effect.
10.On-pump coronary artery bypass grafting:a report of 350 cases

Objective To summarize the clinical experiences of coronary artery bypass grafting ( CABG) in the treatment of coronary artery disease. Methods CABG were performed on 395 consecutive cases from January 2002 to December 2012,including 299 male and 96 female with a mean age of 62. 3 years old. All the operation were performed under cardiopulmonary bypass (CPB ) with moderate hypothermi-a. Left ventricular aneurysm plasty operation were performed in 18 patients. Results The mean number of grafts was 3. 2,the mean CPB time was 88 min( 62~170 min) ,aortic cross-clamping time was 68 min( 25~102 min) ,mean ventilation time was 18 h( 12~72 h) . There were 8 deaths with a mortality of 2. 0%. Six patients died of multiple organ failure,1 patients died of ventricular fibrillation after operation,1 patients died of acute myocardial infarction. Postoperative follow up was carried out on 280 cases,follow-up time was from 8 months to 11 years. Five of them died of unknow causes. The heart function of the rest was significantly improved. 195 patients were free of angina. 85 pa-tients’ s symptom got better. Conclusion CABG performed under cardiopulmonary bypass ( CPB ) with moderate hypothermia is safe and effective for the treatment of coronary artery disease.
